      • Anti-inflammatory effects of luteolin: A review of in vitro, in vivo, and in silico studies

        <P><B>Abstract</B></P> <P><B>Ethnopharmacological relevance</B></P> <P>Luteolin (3′, 4′, 5,7-tetrahydroxyflavone) has been identified as commonly present in plants. Plants with a high luteolin content have been used ethnopharmacologically to treat inflammation-related symptoms. Both isolated luteolin and extracts from luteolin-rich plants have been studied using various models and exhibited anti-inflammatory activity.</P> <P><B>Aim of the review</B></P> <P>This paper uses recent research findings with a broad range of study models to describe the anti-inflammatory activity of luteolin, particularly its mechanisms at the molecular level; provide guidance for future research; and evaluate the feasibility of developing luteolin into an anti-inflammatory drug.</P> <P><B>Materials and methods</B></P> <P>We summarize reports about the anti-inflammatory activity of luteolin published since 2009, which we found in MEDLINE/PubMed, Scopus, Web of Knowledge, and Google Scholar. To acquire broad information, we extended our search to online FDA documents.</P> <P><B>Results</B></P> <P>Luteolin is a flavonoid commonly found in medicinal plants and has strong anti-inflammatory activity <I>in vitro</I> and <I>in vivo</I>. Some of its derivatives, such as luteolin-7-O-glucoside, have also shown anti-inflammatory activity. The action mechanism of luteolin varies, but Src in the nuclear factor (NF)-κB pathway, MAPK in the activator protein (AP)− 1 pathway, and SOCS3 in the signal transducer and activator of transcription 3 <B>(</B>STAT3) pathway are its major target transcription factors. A clinical trial with a formulation containing luteolin showed excellent therapeutic effect against inflammation-associated diseases.</P> <P><B>Conclusion</B></P> <P> <I>In silico</I>, <I>in vitro</I>, <I>in vivo</I>, and clinical studies strongly suggest that the major pharmacological mechanism of luteolin is its anti-inflammatory activity, which derives from its regulation of transcription factors such as STAT3, NF-κB, and AP-1. Much work remains to ensure the safety, quality, and efficacy of luteolin before it can be used to treat inflammation-related diseases in humans.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        p38-dependent c-Jun degradation contributes to reduced PGE2 production in sodium orthovanadate-treated macrophages

        In particular, the phenomenon of c-Jun degradation within theinflammatory response has not yet been fully analyzed. In orderto verify this, we investigated LPS-stimulated murine macrophagespre-treated with sodium orthovanadate (SO) in order to uncoverthe regulatory mechanisms of the MAPKs which regulate c-Jundegradation within the inflammatory response. Through our study,we found that SO suppressed the production of prostaglandinE2 (PGE2) and the expression of COX-2 in LPS-stimulated RAW264.7cells. Additionally, SO decreased total c-Jun levels, withoutaltering the amount of mRNA, although the phospho-levels ofp38, ERK, and JNK were strongly enhanced. Through the usageof selective MAPK inhibitors, and knockdown and overexpressionstrategies, p38 was revealed to be a major MAPK which regulatesc-Jun degradation. Further analysis indicates that the phosphorylationof p38 is a determinant for c-Jun degradation, andis sufficient to induce ubiquitination-dependent c-Jun degradation,recovered through MG132 treatment. Therefore, our results suggestthat the hyperphosphorylation of p38 by SO contributesto c-Jun degradation, which is linked to the suppression of PGE2secretion in inflammatory responses; and thus, finding drugs toincrease p38 activity could be a novel strategy for the developmentof anti-inflammatory drugs.

      • Thymoquinone Suppresses IRF-3-Mediated Expression of Type I Interferons via Suppression of TBK1

        <P>Interferon regulatory factor (IRF)-3 is known to have a critical role in viral and bacterial innate immune responses by regulating the production of type I interferon (IFN). Thymoquinone (TQ) is a compound derived from black cumin (<I>Nigella sativa</I> L.) and is known to regulate immune responses by affecting transcription factors associated with inflammation, including nuclear factor-κB (NF-κB) and activator protein-1 (AP-1). However, the role of TQ in the IRF-3 signaling pathway has not been elucidated. In this study, we explored the molecular mechanism of TQ-dependent regulation of enzymes in IRF-3 signaling pathways using the lipopolysaccharide (LPS)-stimulated murine macrophage-like RAW264.7 cell line. TQ decreased mRNA expression of the interferon genes <I>IFN-α</I> and <I>IFN-β</I> in these cells. This inhibition was due to its suppression of the transcriptional activation of IRF-3, as shown by inhibition of IRF-3 PRD (III-I) luciferase activity as well as the phosphorylation pattern of IRF-3 in the immunoblotting experiment. Moreover, TQ targeted the autophosphorylation of TANK-binding kinase 1 (TBK1), an upstream key enzyme responsible for IRF-3 activation. Taken together, these findings suggest that TQ can downregulate IRF-3 activation via inhibition of TBK1, which would subsequently decrease the production of type I IFN. TQ also regulated IRF-3, one of the inflammatory transcription factors, providing a novel insight into its anti-inflammatory activities.</P>

        Tumor-suppressive functions of protein lysine methyltransferases

        Protein lysine methyltransferases (PKMTs) play crucial roles in histone and nonhistone modifications, and their dysregulation has been linked to the development and progression of cancer. While the majority of studies have focused on the oncogenic functions of PKMTs, extensive evidence has indicated that these enzymes also play roles in tumor suppression by regulating the stability of p53 and β-catenin, promoting α-tubulin-mediated genomic stability, and regulating the transcription of oncogenes and tumor suppressors. Despite their contradictory roles in tumorigenesis, many PKMTs have been identified as potential therapeutic targets for cancer treatment. However, PKMT inhibitors may have unintended negative effects depending on the specific cancer type and target enzyme. Therefore, this review aims to comprehensively summarize the tumor-suppressive effects of PKMTs and to provide new insights into the development of anticancer drugs targeting PKMTs.

        The Stimulus Factors Influencing Intention to Participate in Shopping during the Distribution of the 12.12 Online Shopping Festivals in Malaysia

        Purpose: Online shopping festivals have quickly become the newest trend in online shopping worldwide due to the COVID-19 pandemic. This has led to marketing distribution channels that traditionally emphasized traditional techniques having turned to electronic commerce platforms. Although the pandemic scenario encourages online purchasing, other factors, such as the influence of participation intention to shop during the Online Shopping Festival, must also be considered. Research design, data and methodology: Multiple linear regression analysis was used to test the hypothesis based on data from 121 respondents who are actively involved with online shopping activities in Klang Valley, Selangor. Results: The results of this study show that promotion categories and the perceived influence of mass participation have a significant influence on participation intention. Meanwhile, the perceived temptation of price promotion and perceived fun promotional activities did not significantly influence participation intention. Conclusions: Theoretically, this study contributes to the literature by using the Theory of Planned Behavior and Stimulus-Response models to explain the factors that drive participation intention for online shopping. In practice, this study attracts and encourages customers to shop during the festival day because various attractive promotions are offered by sellers in Malaysia

        Optimization of extractive desulfurization of Malaysian diesel fuel using response surface methodology/Box–Behnken design

        Systematic experiments were conducted to investigate the most appropriate extraction conditions forsulfur reduction. Initial extraction studies showed that DMF was the most potentially effective solvent. The influence of DMF to diesel ratios, extraction times and temperatures, addition of co-solvent, solventmixtures, double extractions were explored. The use of co-solvent and solvent mixtures showed poorefficiencies of sulfur removal. A two-stage extraction with ratio of 1:1 (DMF/Diesel) was successfullyremoved over 88.0% of sulfur in diesel. Under Box–Behnken design, the model showed that maximumextraction ability was observed at 30 8C, 30 min and 1.0 of DMF/diesel ratio to achieved 67.5%.

        Synchronous Occurrence of Bilateral Malignant Otitis Externa: Report of a Rare Case

        Malignant otitis externa (MOE) is a rare and potentially life-threatening disease of the ear and temporal bone. Bilat-eral simultaneous MOE is extremely rare. Due to bilaterally symmetrical facial nerve palsy, it can easily be missed at the initial presentation, causing delay in management. Here, we report a case of bilateral MOE managed aggres-sively with regular ear toileting, ear packing with a ribbon gauze soaked with topical antimicrobials, and long-term intravenous and oral antibiotics. The patient showed good improvement in pain control, facial nerve status, and ear findings.

        Enhancement in photocatalytic degradation of methylene blue by LaFeO3-GO integrated photocatalyst-adsorbents under visible light irradiation

        Perovskite LaFeO3 photocatalyst prepared by using sol-gel glucose method was assembled on graphene oxide sheets to produce integrated photocatalyst-adsorbents (IPCA) and investigated as photocatalyst for the degradation of methylene blue under visible light irradiation. The prepared photocatalyst was characterized by FTIR, XRD, FESEM, BET specific surface area measurement, TEM/HRTEM and UV-Vis spectroscopy analysis. The FTIR, FESEM and TEM analysis has suggested that the photocatalyst LaFeO3 has been successfully embedded at the surface of the graphene oxide (GO) sheets due to a strong interaction between the photocatalyst and the adsorbents matrix. Methylene blue degradation shows that IPCA possesses higher photodegradation kinetics compared to bare LaFeO3 photocatalyst. The resultant photocatalyst also possesses magnetic properties which can overcome the difficulty in recollecting and removal of photocatalyst suspension in water after photocatalytic treatment.

      • Improving the Performance of Industrial Boiler Using Artificial Neural Network Modeling and Advanced Combustion Control

        In heat generation process, performance improvement is a critical factor and essential. An alternative solution is by designing an advanced combustion controller based on neural-predictive control strategy. However, for accomplishing such goal it requires adequate boiler model as well as combustion model. Although heat transfer and combustion processes in boiler are too complex to be analytically described with mathematical model, it can be approximated by artificial neural network model. This paper presents an alternative strategy to model the boiler and combustion process as well as proposes an advanced control strategy that takes the advantage of artificial neural network’ ability as a universal function approximation. A feedforward neural network algorithm is applied to construct the models and the gradient descent technique seeks the optimal network weights, from which the nonlinear predictive control law under the reduced excess air level is derived. Direct application of this control strategy to real-time data taken from a running boiler system at an oil refinery plant demonstrated the benefit of the algorithm to improve the boiler combustion performance.

      • Improving Performance of PID Controller Using Artificial Neural Network for Disturbance Rejection of High Pressure Steam Temperature Control in Industrial Boiler

        The temperature of high-pressure steam is very important to be controlled in order to perform other processes safely, especially for boiler-turbine system. Typically, PID regulator with fixed parameter is used for that purpose. However this method may usually deteriorate the control performance, particularly if the systems exhibit highly coupled behaviour. This paper will present the integration of intelligent control technique, especially artificial neural network, to challenge some deficiencies of PID regulator in dealing with such problem. The proposed control algorithm consists of a neural network controller, which is implemented parallel to the PID controller. The presented neural network controller involves HP steam temperature and its set-point as input and error control signal as a learning signal to be minimized. The ability of proposed algorithm is tested through step-like load disturbance into boiler plant model. Remarkable results have been obtained during this disturbance test. These results showed better performance to reject the disturbances compare with the controller which involves PID regulator alone.
