To develop a HPV-18 oral vaccine, HPV-18 E6 gene was cloned into an expression vector, under the control of CaMV 35S dual promoter and 5`UT region of TEV. The constructed vector (pUS-E6) was used to transform tobacco plants by the Agrobacterium-mediated leaf disc method. The transformants were then regenerated to whole transgenic plants. The HPV-18 E6 gene was found to be expressed in the transgenic tobacco plants as determined by PCR, RT-PCR, Northern analysis and Western analysis. These results show a possibility that HPV oral vaccine may be developed by expressing the HPV proteins in tomato or other vegetables.

담배에서 human papil1omavirus-18형 E6(HPV-18 E6) oncogene을 발현시키기 위하여 E6 유전자를 Cauliflower mosaic virus(CaMV)의 35S dual promoter와 tobacco etch virus(TEV) 5'-untranslated region의 조절을 받도록 벡터를 제조한 후, Agrobacterium-mediated leaf disc 방법으로 담배에 도입하였다. Kanamycin 저항성을 갖는 형질전환체를 선별하고, 완전한 식물체로 재생한 형질전환체의 잎조직에 대하여 PCR 방법과 Northem blot, Westem blot 분석을 한 결과, HPV-18 E6 유전자가 형질전환체 담배에서 성공적으로 발현됨을 알 수 있었다. DNA uptake in dry seeds of gramineous and leguminous plants by imbibition in DNA solution was detected by monitoring the stable expression of β-glucuronidase(GUS) gene. Gene expression vector used in this study contained β-glucuronidase reporter gene and neomycin phosphotransferase II (NPT II) selectable marker gene. Putative transformed plants were analyzed by Southern blot hybridization. Southern blot results confirmed that the two reporter genes are stably integrated into the plant genome.

Purpose: This study aims to explore income-based disparities in breast cancer (BC) incidence and stage at presentation in a national population in South Korea, where a National Cancer Screening Program (NCSP) has been implemented. Methods: In 2007, new patients with BC were identified using the Korea Central Cancer Registry database. We calculated adjusted odds ratios (aORs) to evaluate the association between individual income level and the risk of distant stage BC at presentation, adjusting for women’s age, body mass index, disability registration, employment, region of residence, and year of diagnosis. Results: The cumulative age-standardized incidence of BC in the 11 years was highest among women in the richest quintile (2,040 per 100,000 women for 11 years), whereas the proportion of distant stage at presentation was the highest (10.2%) among the medical aid beneficiaries. The aOR of distant stage diagnosis at presentation was higher for lower-income quintiles, and the risk was the highest in the medical aid beneficiaries (aOR, 2.25; 95% confidence interval, 1.97–2.58) than in the richest quintile. The income-based gradient in aORs for distant stage did not differ between younger (< 40 years) and older patients. Conclusion: A higher risk of distant stage BC at presentation among the lower-income and medical aid groups in the context of a NCSP was observed. A more focused approach toward women in lower-income groups is necessary to alleviate the disparity in the risk of advanced BC.

디지털도서관의 활용도와 중요성이 높아짐에 따라, 사용성(Usability)이 중요한 화두가 되었다. 한편, 사용자 인터페이스는 사용성에 가장 직접적인 영향을 미치는 요소로, 누구나 쉽고 편리하게 서비스를 사용할 수 있도록 설계되어야 한다. 본 연구는 디지털도서관의 사용자 인터페이스 평가지표를 개발하고, 이를 국립중앙도서관 디지털도서관에 적용해 문제점을 분석하고 개선방안을 제시하였다. 평가지표는 문헌연구 및 사례분석을 통해 개발하였으며, 디지털도서관의 특수성을 반영하고자 하였다. 디지털도서관 운영 목적 및 사용자 지원, 시스템메뉴와 네비게이션, 동작과 상호작용, 페이지가 제공하는 정보, 시각적 디자인, 피드백 및 도움, 검색 총 7 가지 분류항목에 따라 세부기준을 마련하였다. 이를 바탕으로 실시한 국립중앙도서관 디지털도서관 분석을 통해서는 총 267 개의 문제점을 발견하였으며, 각각에 대한 구체적인 개선방안을 제시해 디지털도서관 개선 작업 시 실무에서 즉시 활용할 수 있도록 하였다. As popularity and importance of digital libraries are getting higher, usability of the digital libraries are becoming increasingly more crucial. The user interface is one of the most influential factors affecting the usability, thus the user interface should be designed to support easy and convenient usage of the corresponding system. In this study, we developed user interface evaluation heuristics, and applied them to The National Library of Korea’s digital library. The heuristics were developed through literature review and case analysis, while considering the characteristics of digital libraries. The evaluation heuristics include specific criteria according to the seven classification schemes such as purpose of the digital library and user support; menu and navigation; action and interaction; provided information; visual design; feedback and help; and information retrieval. Based on the heuristics, 267 problems were found in The National Library of Korea’s digital library. Practical and specific improvement suggestions were made for each problem to enable immediate implementation of the suggestions when the renovation of the digital library commences.
