This study evaluated the possibility of the 3-dimensional attachment of human periodontal ligament fibroblasts to a periodntally involved root surface after an EDTA treatment in vitro. The human PDL fibroblasts were isolated from the middle third of the root of periodontally healthy teeth extracted for orthodontic reasons. The cells were cultured in a medium containing Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum at $37^{\circ}C$ in humidified air containing 5% $CO_2$. Eight single-rooted teeth were obtained from patients diagnosed with periodotitis. After scaling and root planing, four teeth were etched with 24% ethylenediaminetetracetic acid (EDTA) for two minutes (Experimental group). The other four teeth were not treated with EDTA and were used as the control group. The human PDL fibroblasts were placed in the total root surface and cultured for 4 weeks. The teeth were fixed in 2.5% glutaraldehyde in PBS before preparation for the scanning electron microscopy (SEM) examination. The human PDL fibroblasts showed a healthy morphology on the root surfaces treated with EDTA (Experimental group) and a relatively unhealthy appearance on the treated root surfaces (Control group). This suggests that EDTA favorably affects the 3-dimensional attachment of human PDL fibroblasts cultured on the root surfaces. which may play an important role in periodontal healing and regeneration.
Fast Protein Liquid Chromatography를 이용한 황색포도상구균 장독소 B 분리

황색포도상구균 식중독은 해당 균이 세포 밖으로 분비하는 장독소에 의해 발병되며, 지금까지 밝혀진 16가지독소 중 내열성이 가장 강하고 흡입중독으로 쉽게 질환을일으킬 수 있는 Staphylococcal enterotoxin B형을 분리해내는 방법에 대해 연구하였다. 분자량 26-29 kDa의 단순단백질인 SEB를 분리 및 정제하기 위한 방법으로 fast protein liquid chromatography (FPLC)를 이용하였으며, 정제율을 높이기 위해 ion exchange chromatography와 gel filtration chromatography의 두 과정을 거쳤다. 실험결과 SEB의 분리 조건은 CIEX에서 WorkBeads 40S컬럼의 분리능이 가장 좋았고, 염 농도 15%에서 SEB가 분리됨을 확인하였다. CIEX에서 획득한 분획으로 단백질을 크기별로 나누는 GF를 실시하였을 때, Sephacryl S-100컬럼을 사용하여 정제된 SEB를 획득할 수 있었다. 시료가 컬럼을 통과할 때 염 농도가 효율적으로 조절될 수 있도록 하고 SEB를 농축하려 시도한 투석 및 완충액 치환방법은 전체적인 단백질량 손실을 가져오는 것으로 나타났다. 본 연구에서 개발된 독소 분리 및 정제 방법은 향후 다른 장독소 분리에도 활용할 수 있으며, 식중독 신속진단법 개발 및 항체 생산 발전에 효율을 높일 수 있을 것으로 기대된다. The aim of this study is to develop a method to separate and purify the Staphylococcal enterotoxin B (SEB). The method was based on fast protein liquid chromatography (FPLC) along with gel filtration (GF) after cation exchange chromatography (CIEX) treatment. Various columns such as CM sepharose HP, Cellufine C-500, WorkBeads 40S for CIEX, and GF on Sephacryl S-100 for GF were compared in terms of separation efficiency. Consequently, Work- Beads 40S along with Sephacryl S-100 produced best separation performance. Separated SEB with developed method was verified with ELISA assay. Purified SEB with the method can be used to manufacture rapid detection kit or antibodies.

Lipoma is the most common benign mesenchymal neoplasm composed of mature fat. Although lipoma can occur in any part of the body, it is infrequently found in the oral cavity. Especially, lipoma of the tongue is rare and there have been no previous reports of this in the Korean dermatology literature. Lingual lipoma, which accounts for 0.3% of all tongue neoplasm, is typically described as well-circumscribed, submucosal mass and it is located on the lateral border of the anterior two-thirds of the tongue surface. We herein report on a rare case of lipoma of the tongue that occurred in a 77-year old male patient. (Korean J Dermatol 2010;48(5):440~441)
