        국내 육성 장미 품종 꽃잎 유래 체세포배 발생 캘러스 유도

        This study was conducted to induce somatic embryogenic callus (SEC) derived from petals in rose. The petal explants of 3 cultivars ('Ice Wing', 'Orange Eye' and 'Pink Beauty') with different flower colors were placed on three types media (MS, SH and WPM) supplemented with 11 mg/L 2,4-D, respectively, and then cultured in the dark for 47 days. Calluses were formed at explants of all three cultivars. Also, 'Ice Wing', which were cultured in the SH as the basal medium, showed the highest callus formation rate. However, somatic embryos were generated from only petal-derived callus of 'Ice Wing', which were induced on the WPM as the basal medium, transferred it to SH basal medium supplemented with 3 mg/L 2,4-D, and 300 mg/L L-proline, and cultured for 5 weeks. The SEC has been proliferated every four weeks at the subculture interval. In addition, as a results of making a comparison of expression of RhSERK3 and RhSERK4, which is used as signal for generation of somatic embryo from callus in rose, between the SEC and petal-derived callus from 'Ice Wing' by RT-qPCR, the former showed 10 times higher RhSERK3 expression and 700 times higher RhSERK4 expression than the latter.

        Hebb의 학습 법칙과 화소당 가중치 최소화 기법에 의한 적응학습 및 그의 전기광학적 구현

        Hopfield 모델에 화소당 가주치를 도입하고 이를 최적화하여, 서로간에 상관관계가 높은 "0"에서 "9"까지의 10가지 숫자를 성공적으로 기억, 재생시킬 수 있는 $6{}8$ nodes 연상기억 시스템을 소개한다. 다른 많은 신경회로와는 달리, 이 모델은 "6","8","3","9"와 같이 상관관계가 매우 큰 영상에 대해서도 높은 오차 교정 능력을 가짐을 볼 수 있다. 화소당 가중치의 최적화 무제는 최소자승평균 오차 알고리듬에 기초한 적응학습 과정으로 볼 수 있으며, 이는 또한 Widrow-Hoff 신경회로로 구현 할 수 있다. 가중치 최적화 회로의 전기 . 광학적 구현을 위한 설계도 소개한다. Introducing and optimizing bit-significance to the Hopfield model, ten highly correlated binary images, i.e., numbers "0" to "9", are successfully stored and retrieved in a $6{}8$ node system. Unlike many other neural network models, this model has stronger error correction capability for correlated images such as "6","8","3", and "9". The bit significance optimization is regarded as an adaptive learning process based on least-mean-square error algorithm, and may be implemented with Widrow-Hoff neural nets optimizer. A design for electro-optic implementation including the adaptive optimization networks is also introduced.

      • 간(肝) 지방(脂肪) 침윤(浸潤)에 대한 초음파(超音波) 소견(所見)과 혈액(血液) 생화학적(生化學的) 검사(檢査)의 상관관계(相關關係)에 관한 연구(硏究)

        In medical selection of Daehan Education Insurance, We doubted that 140 cases(male 117 cases, female 23 cases) might have the fatty infiltration of the liver by ultrasonography. We examined the weight and the hypertension of 140 patients by biochemically analyzing the blood extracted under fasting according to the age-distribution. The results are as follows: 1. 90% out of them had body weight than normal subject. 34.29% out of them were hypertension patients and most of them were $30{\sim}39's$ and $40{\sim}49's$. 2. As compared with the normal subjects in serum lipid value, HDL cholesterol value was low, but total cholesterol, Triglyceride, phospholipid and total lipid value were significantly high. Especially the statistical value of Triglyceride was very significantly high. 3. Total abnormal rate of sGOT value in Liver function test is 34.43% and the mean value of it is 66 unit and the abnormal rate of sGPT value is all 3.14% and the value of it is 70 unit. 4. The abnormal rate of Alkaline phosphatase value is 9.29% and the abnormal rate of total Bilirubine value is 14.29%. As HBsAg positive rate is 2.14% and HBs Ab positive rate is 31.43%, the positive rate of HBsAg represents Lower positive rate than Korean mean value. 5. The abnormal rate of the fasting blood glucose is 15.00% and the abnormal rate of Blood sugar pc 2hrs is 30.71%. 6. The abnormal case of protein, Albumin, Globulin, BUN, craetinine and Hemoglobin value was not found to any of them and in the above liver function test except serum lipid value, the rate not showing the abnormal value represents 45.3%(male) and 52.17%(female).

      • 혈장 Plasminogen 과 Prothrombin 의 생산에 관한 연구

        토끼의 정맥에 주사된 [$^{75}Se$]-L-methionine에 편입된 혈장 plasminogen과 prothrombin의 편입율은 주사후 30분 부터 출현하기 시작하여 3시간 후에 최고치에 도달한 뒤 plateau를 이루었다. Plasminogen과 prothrombin은 다 같이 혈장에 존재한다는 것은 잘 알려진 사실이다. (Biggs & Macfarlane, 1962). plasminogen은 섬유소(fibrin)를 용해시키기 위해서 plasminogen activator에 의해서 활성 형인 plasmin으로 전환된 다음에 섬유소에 작용한다는 것도 이미 다 알려져 있지만 plasmin의 체내 생산장소에 대해서는 아직까지 확실히 알려져 있지 않고 다만 면역형광법에 의한 관찰 결과, 골수의 호산성과립구(eosinophilic leukocyte)에서 생산되어 순환내로 유리되는 것이 아닌가 추측되고 있다(Barnhart & Riddle, 1963). 한편 prothrombin은 혈액이 응고될 때에 prothrombinase에 의하여 활성화 되어 thrombin으로 되며, 이 thrombin이 칼슘이온과 함께 섬유소원(fibrinogen)에 작용하여 섬유소(fibrin)로 전환시키며, 또 이 prothrombin의 생산장소는 간이라는 것은 잘 알려진 사실이다(Finkbiner et al., 1959; Rapaport et al., 1960). 저자는 혈액응고 과정에서 섬유소원을 섬유소로 전환시키는데 작용하는 thrombin의 zymogen형인 prothrombin과 섬유소를 용해시키는데 작용하는 plasmin의 zymogen형인 plasminogen이 체내의 생산장소로부터 각각 생산된 뒤에 순환혈액으로 유리되는 방식(mode)을 관찰하기 위하여, 추적자로서 [$^{75}Se$]-L-methionine을 토끼의 정맥에 주사한 뒤에, 일정한 시간 간격으로 도살하고, 혈장 plasminogen과 thrombin에 각각 편입된 [$^{75}Se$]-L-methionine의 방사능을 계측하였다. Both plasminogen and prothrombin incorporated by [$^{75}Se$]>-L-methionine in the plasma of rabbits were rapidly increased up to 3 hours after [$^{75}Se$]-L-methionine infusion, and then showed plateaus, respectively.

      • 한국인(韓國人)의 혈청지질(血淸脂質) 연구(硏究)

        In Med Dept. Daehan Education Insurance, Total 690 cases(male 376 cases, female 314 cases) were inspected to study the change of serum lipid value. The objects of investigation were total cholesterol, HDL cholesterol, triglyceride, phospholipid and total lipid value. The results are as follows: 1) In the normal subjects, with the relation of serum lipid value and the age. triglyceride, phospholipid and total lipid value showed the increasing tendency by 70 age, but total cholesterol value and HDL cholesterol value showed the increasing tendency by 59 age and the decreasing tendency after 59 age. 2) In the normal subject the difference of serum lipid value between sexes did not make the significant difference of level statistically in all items which can be inspected. 3) The weight change and serum lipid value are very significantly related. Total cholesterol, HDL cholesterol and total lipid value are high in female if the weight is light. But they are high in male the weight is heavy phospholipid value is opposite to the above result. Triglyceride value is high in male regardless the weight, but the significant difference happened in the high weight group. 4) As serum lipid value of hypertension patients is compared with that of the normal subjects, HDL cholesterol, triglyceride, phospholipid and total lipid value are significantly high. 5) The doubtful group of the fatty infittration of liver among the hypertension pafients have very significant difference in serum lipid value compared with not only the value of the normal subject but also that of the obesity group. 6) The doubtless group of the fatty infitration of liver among the hypertension patients have the significant difference in all serum lipid value compared with that of the normal subject, but compared with the obesity group males had the significant difference of HDL cholesterol value and both sexes had the significant difference in total lipid value, but had no significant difference in total cholesterol, triglyceride and phosprolipid value. 7) The ratio of total cholesterol per HDL cholesterol had the significant difference in the doubtful hypertension patients of the fatty infittration of liver and the relative weight, but it had no significant difference compared with the normal subject in the doubtless hypertension patients.

        한국 4년제 치위생학과의 교육과정

        Education should consider the social consensus that changes according to the times and the social environment, and it is also necessary to consider the technically useful subjects. We collected and reported the 4-year course curriculum of dental hygiene when Korean dental hygiene education has continued over 50 years. Each 4-year course curriculum was collected by searching each university website, or requested by email. The curriculum of 23 among 27 schools was collected. According to the classification of dental hygiene in the dental hygienist national examination classification or the Korean society of dental hygiene science, the subjects of the course were divided into clinical dental hygiene, clinical dental treatment support, basic dental hygiene, social and educational dental hygiene, and collected 23 curriculum courses. The average major curriculum was 104.9 credits and the average number of subjects in major courses was 34.3. The average subjects' number of clinical dental hygienic courses was 33.9 (37.5% of the total major credits), clinical dental treatment support was 30.6 (30.2% of the total major credits), basic dental hygiene was 21.8 (20.8% of the total major credits), and social and educational dental hygiene courses was 13.6 (13.0% of total major credits). Integrated subjects' name in clinical dental hygiene was used in all schools of the survey, such as clinical dental hygiene education (and practice), comprehensive dental hygiene education, and integrated dental hygiene. There were 13 schools (56.5%) that use the integrated name in clinical dental treatment support, such as clinical dentistry. There were 14 schools (60.9%) to open dental clinic management, and 22 schools (95.7%) to open national health insurance claims. The basic dental hygiene curriculum maintained the title of each subject and social and educational dental hygiene education was established in most schools, such as community dental health, oral health statistics, and oral health education. Other subjects were English conversation in dental clinic (8 schools) education, clinical dental treatment support, basic science in dental hygiene, social and educational dental hygiene. We knew the clinical dental hygiene and clinical dental treatment support were changing into the integrated subjects, and most schools run dental hygiene research.

        Expression of an artificial gene encoding a repeated tripeptide lysyl-g1utamyl-tryptophan in Tobacco Plant

        식물 단백질의 영양가 향상을 위한 일환으로 필수아미노산의 조성이 풍부한 인공단백질을 암호화하는 인공유전자를 담배 식물체에서 발현을 시도하기 위하여, 식물에서 외래유전자의 발현에 널리 사용되는 Cauliflower mosaic virus (CaMV)의 35S promoter를 이중으로 중첩되도록 하고, (Lys-Glu-Trp)이 64번 반복되는 인공유전자 및 nopaline synthase (nos) terminator를 갖고있는 binary vector pART4-4를 구성하였다. 이 재조합 플라스미드는 Agrobacterium tumefaciens를 이용한 형질전환에 의해 Nicotiann tabacum (Var. Xanthi)으로 도입되었다. Kanamycin이 포함된 신초 유도 배지 및 뿌리 유도배지를 이용하여 정상적으로 재생된 담배 식물체로부터 도입된 인공유전자의 발현을 분석하였다. 추출한 genomic DNA를 EcoRI으로 자른 다음 Southern blot 분석에 의하면, 효소 절단 시 예상되는 1.1 kb에서 band를 형성하였으며 각각의 형질전환 식물체에 인공유전자가 1 또는 3 개씩 도입되어 있음을 확인하였다. Northern blot 분석에 의하면 약 1.2 kb 전사체가 비교적 안정하게 발현되었으며, 잎, 줄기, 뿌리로부터 RNA를 분리하여 promoter의 조직 특이성 발현을 분석한 결과, 잎에서 생성되는 RNA가 줄기나 뿌리 조직보다 안정하게 발현되었다. 형질전환 식물체에서 Western blot에 의한 단백질 분석 결과, 잎에서 추출한 단백질로부터 원하는 크기인 33 kDa의 인공단백질이 생성됨을 확인하였으며 발현 수준은 전체 세포 단백질의 0.1%로서 낮은 수준이었다. To investigate expression of the artificial gene encoding a repeated tripeptide lysyl-glutamyl-tryptophan in tobacco plant, the plant binary vector, pART404 has been constructed, which contains the duplicated CaMV 35S promoter, an artificial gene coding for repetitive polymer (Lys-Glu-Trp)$_{64}$, and nopaline synthase (nos) terminator. The recombinant expression vector was introduced in Nicotiana tabacum (var. Xanthi) via Agrobacterium tumefaciens-mediated trans-formation. The transgenic calli selected by kanamycin containing medium were then regenerated to whole plants. Southern blot analysis indicated that five transgenic plants (No. 1, 7, 9, 43, 45) showed the hybridizing signals at 1.1 kb of the expected size on EcoRI digestion and each of the transgenic plants contained 1 or 3 copies of the artificial gene inserted into its genome. By northern blot analysis, the size of the hybridized total RNA was estimated to be approximately 1.2 kb and the RNA appeared generally to have the integrity. Western blot indicated that the protein was detected at the position of 33 kDa and the expression level of the polypeptide in the transgenic plant (No. 45) was measured to approximately 0.1% of the total protein.
