        새송이버섯 수확 후 배지로부터 surfactin 생성 Bacillus amyloliquefaciens YJ07의 분리 및 특성

        Spent mushroom substrates (SMS) is a by-product remaining after a crop of mushrooms. About four surfactin-producing strains were isolated from SMS (Pleurotus eryngii). Among of them, one isolate, which designated to YJ07, potentially showed the antifungal activity against Aspergillus flavus and Aspergillus ochraceous producing mycotoxin on PDA medium. The biochemical characteristics of the strain YJ07 was similar with Bacillus subtilis and Bacillus amyloliquefaciens by Bacillus ID kit and VITEK 2 system. Comparative 16S rDNA gene sequence analysis of the strain YJ07 also showed that the strain YJ07 was most closely related to Bacillus amyloliquefaciens with sequence similarity of 99.5%. On the basis of their biochemical characteristics and phylogenetic distinctiveness, the strain YJ07 was classified within the genus Bacillus as Bacillus amyloliquefaciens YJ07. The antifungal compound from B. amyloliquefaciens YJ07 was similar to lipopeptide surfactin from Bacillus subtilis by TLC and HPLC analysis.

      • 미토콘드리아 및 Mating type 유전자를 이용한 느타리버섯류의 분자표지 개발

        버섯은 세계적으로 양송이, 표고 느타리버섯류 재배가 주류를 이루고 있으며, 한국의 시중에서 재배하고 있는 버섯품종이 1969년 이후 꾸준히 증가하여 약 24품목 224품종이 신고되거나 품종보호등록되어 농가에 보급되었다. 그 중에서 느타리와 큰느타리가 전체 생산 및 소비가 약 50%를 차지하고 있다. 이러한 상황에 비춰 하나의 품종이 다른 이름으로 신고되거나 등록되는 경우가 존재하여 먹는 버섯만 해도 구별하기가 쉽지 않은 상황이다. 과거에는 일반인이 외국에서 품종을 수집하여 그대로 판매신고를 하여 상품으로 판매하는 사례도 있는 실정이다. 지금은 품종등록을 한 종균을 허가없이 사용할 경우 로열티를 지불하거나 소송이 진행될 수 있음을 간과해서는 안된다. 일례로 새송이버섯 사건이 대표적인 예라 할 수 있다. 이 사건의 품종판별 방법으로 유전자 식별법을 사용하였다고 한다. 이것은 형태적 형질만으로는 품종구분 방식에 한계가 있음을 보여주고 있다. 우리는 미토콘드리아 유전자를 이용하여 품종구분을 하였는데 느타리에서 원형느타리, 수한1호, 춘추2호 등이 뚜렸한 차이를 보여주어 품종을 구분할 수 있는 마커로서의 가능성을 보여주었다. 그리고 교배육종된 교잡주의 모본유래를 확인함으로서 육종 마커로 사용할 수 있으리라 판단된다. 또한 육종효율 증진을 위해 mating type을 결정하는 homeodomain 및 pheromone receptor 유전자를 이용하여 교배형을 결정함으로서 교배육종 시 시간과 경비를 절감할 수 있으리라 사료된다.

        Di-Mon 교잡법에 의한 큰느타리버섯 교잡주의 특성

        Pleurotus eryngii is not edible and medicinal mushrooms indigenous to Korea. To improvement of strain suitable to the geographic setting of Korea, we are mated with 22 dikaryons and 47 monokaryons isolated from Pleurotus eryngii ASI 2547 by Di-Mon mating. 19 strains forming fruit body obtained from clamped 253 bred strains. 7 excellent strains are selected from 19 bred strains by various morphological features of fruit body. Among the selected 7 strains, H6 strain were identified into ASI 2547-like recombinant hybrids with URP uniprimer by RAPD analysis. This suggested that Di-Mon crossing is one of rapid and easy breeding method for strain improvement with molecular techniques.

        아위느타리 신품종 ‘비산2호’의 육성 및 자실체 특성

        To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-68 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2798-24 derived from ASI 2798 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 x ASI 2798-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as ‘Beesan No.2’. The ‘Beesan No.2’ was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803, ASI 2798 and Beesan No.1 . Analysis of the genetic characteristics of the new cultivar ‘Beesan No.2’ showed a different DNA profile as that of the control strains, ASI 2803, ASI 2798 and ‘Beesan No.1’, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were 25oC and pH5~8, respectively. Fruiting body production per bottle was about 123.3 g. And also the stipe was long. This new cultivar ‘Beesan No.2’ of Pleurotus ferulae was characterized white-like variety of King oyster mushroom in the color of stipe, that was long and low yield compared to that of other cultuvar ‘Beesan No.1’. We therefore expect that this new strain will increase of export and substitute cultivar of King oyster mushroom.

Analysis of the genetic characteristics of the new cultivar ‘Beesan No.1’ showed a different DNA profile as that of the control strains, ASI 2803 and ASI 2850, when RAPD(Random Amplified Polymorphic DNA) primer URP6 was used. The optimum temperature and pH arrange for mycelial growth were 25oC and pH5~8, respectively. Fruiting body production per bottle was about 245 g using demonstration farms. And also the stipe was thick and long. This new cultivar ‘Beesan No.1’ of Pleurotus ferulae was characterized by fast fruitbody formation, the stipe was thick and long and high quality yield compared to that of other cultuvars. We therefore expect that this new strain will increase of farmer’s income by construction of stabilized production system.

        신령버섯의 기형으로부터 배양불능세균의 PCR 검정

        Agaricus blazei Murill is a important medicinal mushroom for a powerful immune system builder and tonic. Currently, it is known about a new disease phenomenon that appears to be occurring on a number of mushroom farms. We described a straightforward approach in which molecular methods was used to survey the presence of potentially endo- and epiphytic bacteria infected with the Agaricus blazei. The 16S rDNA was amplified with universal eubacterial primers directly from pure cultures of Agaricus blazei mycelium and fruit body. The 16S rDNA sequences were almost identical (96 to 97% similarity), and phylogenetic analysis placed them in a single unique rRNA branch belong to the uncultural bacterium phylogroup. PCR detection of uncultural bacterium in the malformed tissues of Agaricus blazei were carried out by using 16S rRNA sequenced specific probe. It was strongly amplified at the malformed pileus region of fruit body and also spore print was impossible.

        무포자느타리 선발을 위한 단핵화 균주의 분리 및 Spo11 마커의 이용

        For the development of a sporeless strain of P. ostreatus we used sporeless strain ASI 2069. We have recovered both nuclear types of strain ASI 2069 as monokaryons of nh9, nh15, nh26 and nh36 (here after referred to as neohaplonts) by protoplasting the mycelium. Crosses between neohaplonts and SSI’s(single spore isolates) obtained from a sporulating commercial strain ASI 2180. Five excellent strains are selected from 30 bred strains by quality of fruitbodies and spore number. To development of molecular markers linked to sporeless strain of P. ostreatus, we are screened helicase, recombinase(DMC1) and topoisomerase(Spo11) genes related meiosis by PCR and sequencing. Among three genes, Spo11 gene was identified into molecular marker of sporeless from neohaplonts and bred strains of P. ostreatus.

        세포질전환 기법에 의한 신품종 느타리 ‘천화심’의 육성 및 자실체 특성

        Hybrid strains was selected by crossing between dikaryotic strain ASI 0628 (Dagul) bred from ASI 2596 (Suhan No.3) and ASI 2782(Black pileus mutant) and monokaryotic strain ASI 2344-84. The strain 84 that shown the best cultural characteristics was selected to be a new cytoplasmic hybrid variety and named as ‘Chunhwashim’. The ‘Chunhwashim’ shown incompatibility among them forming the confrontation growth line against parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho). Fruiting body produced about 161.4±4.8 g per bottle. And also the individual generation of fruit body is multiple than chunchu2ho as 40.9. The ‘Chunhwashim’ was cytoplasmic hybrid included hybrid DNA bands of parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho) by Random Amplified Polymorphic DNA (RAPD) primer, and mitochondria DNA band of monokaryotic ASI 2344 (chunchu 2ho) using mitochondria microsatellite DNA marker. This new cytoplasmic hybrid variety 'Chunhwashim' of oyster mushroom is characterized by multiple of individual generation and deeply grey color of pileus.
