        소의 동결수정란이 수태에 미치는 영향 2 . 자당부유액에 의한 2 단계 평형의 영향

        Twenty frozen embryos obtained from high genetically potential donors were introduced by cooperation with the American Holstein Association. The 47 recipients at the National Livestock Breeding Station were intramuscularly injected once or twice with 25㎎ PGF₂α for synchronization of estrus cycle with donors. The frozen embryos were rehydrated in 2-steps sucrose media after thawing at 37℃ and embryo transfer was performed by both of surgical and non-surgical methods. In addition, fresh embryos collected from 3 donors of the Station were transferred. The results were as follows: 1. Twenty-two of 23 fresh embryos obtained from 3 superovulated donors showed good morphology and average 7.3 embryos per donor were collected. 2. Among 47 heads treated, rate of estrus induced by twice of PGF₂α was higher (87.5%) than by once (78.7%), and the rate of heifers was higher than cows. 3. The quality of corpus luteum of 45 recipients synchronized in estrus was composed of 15 cows (33.3%) in excellent or good, 23cows (51.1%) in fair or little poor and 7 cows (15.6%) in no corpus luteum or cystic follicle, respectively. Most of recipients showed fair or little poor corpus luteum. 4. All frozen embryos except seven showed geneally good qualities in results of examination before transfer. 5. Twelve (40%) among 30 embryos were impregnated and all of them were Holstein breeds. 6. Two of 12 frozen and 10 of 18 fresh embryos were concepted, respectively. The conception rate of fresh embryo was higher than that of frozen embryo. 7. Forty-five percentage (rune of 20 recipients) of conception rate by non-surgical were higher than 30% (three of 10 recipients) by surgical method. 8. The surgical conception rates (66.7%) of fresh embryos were higher than non-surgical method (53.3%). However, surgical method (14.3%) of the frowns were lower than non-surgical (20.0%).

        Nineteen frozen embryos with high genetic-potency were imported from Holstein Registration Association of America and transferred to recipients of the National Livestock Breeding Station. The synchronization was induced by twice intramuscular injections of 25㎎ PGF₂α with 14 days interval. The frozen embryos thawed at 37℃ were rehydrated within 6 steps in order to remove glycerol of the medium. The embryos were transferred to 19 recipients with non-surgical method and the results were as follows. 1. On 1st and 2nd intramuscular injection of PGF₂α, twentyeight (68.3%) and thirty-six (87.8%) among 41 treated showed estrus within 4 days after treatment. 2. The rate of the standing heat on day 3 after injection of PGF₂α was 53.7 and 73.6% at 1st arid 2nd injection, respectively. 3. The rate of the heifers showing standing heat after 1st and 2nd injection was 79.2 and 91.7%, but that of the cows was 52.9 and 82.4%. The rate was higher in heifer than cow. The rate of heifer selected for embryo transfer among synchronized recipients was also higher (62.5%) than that of cow (23.5%). 4 The evaluation of 19 frozen embryos (12 Holsteins and 7 Limousins) exception of 4 eggs with broken zona pellucida was generally good quality blastocysts as pre-freezing embryos. The pregnancy was obtained only from blastocyst. 5. At the examination of pregnancy at 60 days after nonsurgical transfers, three (15.8%) of 19 recipients were concepted. As the results, 2 female Holsteins (16.7%) and 1 male Limousin (14.3%) were delivered in terms of normal pregnant period.

        수정란의 급속동결융해법에 관한 연구 1 . 마우스 동결수정란에 대한 1단계 Straw 법이 난자생존성에 미치는 영향

        This is a preliminary study to simplify freezing, thawing and transfer procedure of cattle embryos using mouse embryos. The experiments were designed to investigate the effective concentration of glycerol and sucrose by one-step straw method, and to determine the optimum thawing temperature. Effects of viability among permeable cryoprotec-tants and impermeable sugar dilutions was also studied. Results obtained were summarized as followed; 1. A total of 1,340 embryos were cryopreserved among which 1,390 embryos collected from 136 mice. 2. Post-thaw development to expanded blastocysts, was highest when embryos were frozen in 1.0 M glycerol+0.5 M sucrose. However, proportions of morphologically normal embryos was highest when they were frozen in 0.25 M glycerol+0.5M sucrose. 3. Post-thaw development to expanded blastocysts was highest when embryos were frozen in a lower concentrated sucrose compared to higher concentration. 4. The temperature thawed at 20℃ and 30℃ resulted in a significantly higher embryos survival rate after 72 hr. in culture than did at 35℃. However no significant differences was denoted between 20℃ and 30℃ (p$lt;0.05). 5. In effects of non-permeable sugar dilutions on in-vitro development of frozen-thawed mouse embryos, raffinose and trehalose were compared favorably with sucrose dilution in morphological recovery and survival rates. 6. When the embryos were frozen-thawed rapidly in 1.0 M glycerol and trehalose concentration, the proportions of embryos developed to expended blastocyst were significantly better in 1.25 M and 0.5 M than other higher concentration (p$lt;0.01). 7. In effects of permeable cryoprotectants and sucrose concentration, 1.5 M of permeable freezing media were significantly higher survival rate than that of 2.0 M and propanediol was resulted in slightly higher survival rate than glycerol. However, no significant differences showed among sucrose concentrations exception of null dilution(p$lt; 0.01). The results from this experiment suggested that the use of propanediol and raffinose in the rapid freezing technique offers slightly better results than does the use of glycerol and sucrose.

        수정란의 급속 동결융해법에 관한 연구 2 . 비침투성 동결보호제가 급속동결한 마우스 수정란의 생존성에 미치는 영향

        This experiment was carried out to investigated effects of trehalose and sucrose concentration as a nonpermeable cryoprotectants on in vitro development of mouse embryo frozen-thawed by direct plunging from room temperature to liquid nitrogen. The results obtained from the experiment are as follows: 1. When the embryos frozen-thawed ultrarapidly in 3.5M glycerol and 0.25 or 0.5M sucrose, the proportions of morphologically normal embryos and those developed to expanded blastocysts were slightly higher in 0.5M(l00% and 80%) than in 0.25M(62.5% and 72%), but there was significant difference between those with and without sucrose(p$lt;0.01). 2. In effects of glycerol concentration added to 0.5M sucrose or 0.5M trehalose on in vitro development of frozen-thawed mouse embryos, 3.5M glycerol resulted in a higher embryos survival rate and proportion of embryos developed to expanded blastocysts. Also the group of sucrose (83%) added medium showed significantly higher embryos survival rate than trehalose(65.6%) group(p$lt;0.01). 3. In effects of permeable cryoprotectants added to 0.5M trehalose on in vitro development of the embryos, 3.5M glycerol and 3.5M propanediol resulted in better embryos recovery and survival rates than other dilution of freezing medium. In difference between the groups of glycerol and propanediol in the freezing medium, glycerol resulted in a significantly higher embryos survival rate than propanediol(p$lt;0.01). 4. When the embryos were frozen-thawed ultrarapidly in mixtures of 25% glycerol, 25% propanediol and surcrose solution, highly survival and recovery rates more than 90% were obtained.

        Expanding 사료 가공 기술이 산란계의 계란 품질 및 영양소 소화율에 미치는 효과

        This study was conducted to investigate the effects of expanding feed processing technologies on the egg characteristics and nutrient digestibility in laying hens. A total of two hundred seventy laying hens were randomly allocated into three treatments with fifteen replications for six weeks. Dietary treatments include 1) MS(mash), 2) EC(expanded crumble) and 3) EP(expanded pellet). In the egg production, the EC and EP treatments were significantly higher than MS treatment(p<0.05). However, no significant difference was founded in egg weight(p>0.05). Egg shell breaking was increased in EC and EP treatments compared with MS treatment(p<0.05), however, egg shall thickness showed no significant difference(p>0.05). Egg production and egg shell breaking were significantly higher in EC and EP treatments than MS treatment(p<0.05), but egg weight and egg shell thickness were no significantly difference among treatment(p<0.05). Yolk color was increased in EC treatment compared with other treatments(p<0.05). Also, yolk index was significantly improved in EC and EP treatments compared with MS treatment(p<0.05). Haugh unit was increased in EC treatment compared with MS treatment(p<0.05). EC treatment had improved DM, Ca and P digestibility. In conclusion, Expanded crumble and pellet processing could improve the egg characteristic and nutrient digestibility in laying hens. 본 시험은 expanding 가공 기술이 산란계의 생산성, 계란 품질과 영양소 소화율에 대하여 알아보고자 실시하였다. 49주령 Hy-line Brown 270수를 공시하였고, 6주간 시험을 실시하였다. 1) MS(mash), 2) EC(expanded crumble) 및 3) EP( expanded pellet) 으로 3처리구로 구성하였으며, 처리 당 15반복, 반복당 6수씩 완전임의 배치하였다. 산란율은 EC와 EP 처리구가 MS 처리구와 비교하여 유의적으로 높게 나타났다(p<0.05). 그러나 난중은 처리구간에 유의적인 차이는 없었다(p>0.05). 난각 강도는 EC와 EP 처리구가 MS 처리구와 비교하여 유의적으로 높게 나타내었지만(p<0.05), 난각 두께는 처리 구간에 유의적인 차이를 나타내지 않았다(p>0.05). 난황색은 EC 처리구가 MS 처리구와 비교하여 유의적으로 높게 나타났다(p<0.05). 난황 계수는 EC와 EP 처리구가 MS 처리구와 비교하여 유의적으로 높게 나타났다(p<0.05). Haugh uint에 있어서 처리구간의 유의적인 차이는 없었으나(p>0.05), 변화량에 있어서 EP 처리구가 가장 높게 나타났다(p<0.05). 영양소 소화율에 있어서 건물, 칼슘, 인 소화율에 있어서 EP 처리구가 소화율을 개선시켰다(p<0.05). 본 연구 결과는 2차적인 사료 가공 기술이 산란계에 서 계란 품질과 영양소 소화율을 개선하였다.

        돼지 수정란이식에 관한 연구 2 . 수정란의 회수 및 이식

        This study of embryo transfer was carried out to investigate the possibility of introducing new genetic material into closed herds for disease control and of transporting embryos between countries. The results obtained were summarized as follows: 1. On the basis of the number of ovulation points counted, 69.1% of embryos were recovered. 2. A very high proportion (86.6%) of 1-cell embryos were recovered from the oviducts of donor sows at 37 to 41 hours after first mating. 3. Sixty-seven embryos were transferred to 5 recipients. Four recipients farrowed a total of 18 pigs, giving a survival rate of 26.9%. 4. Average pig weight at birth was 1.46㎏.
