        해양생물 크릴(Euphausia superba)이 흰쥐의 혈청 유리지방산 및 전해질 농도에 미치는 영향

        본 연구는 식이조성에 동결 건조한 krill (Euphausia superba) meal을 첨가 하였을 때 Sprague Dawley계 수컷 흰쥐의 유리지방산(non-esterified fatty acid, NEFA), blood urea nitrogen (BUN), creatinine, uric acid 및 전해질에 미치는 영향에 대하여 조사를 하였다. 이조성은 기본식이를 급여한 대조군인 GC군(krill meal 미함유)을 비롯하여 기본식이에 10.0%, 20.0%, 30.0%의 krill meal 급여군을 각각 G10군, G20군, G30군으로 구분하였다. 5주간 실험 사육한 결과, 흰쥐의 NEFA와 BUN 농도는 krill meal 급여군(G10군, G20군, G30군)에서 대조군(GC 군)보다 유의적으로 감소된 결과를 보였고, creatinine과 uric acid 농도는 G10군에서는 GC군과 유의적 차이가 없었으나, G20군과 G30군에서 유의적으로 감소하였다(p<0.05). 전해질 농도에서 total calcium (T-Ca)은 G20군과 G30군에서 유의 적으로 증가하였으며, 인(Pi) 농도는 각 군간에 유의적 차이가 없는 것으로 관찰되었다(p<0.05). The purpose of this study was to investigate the change of hematology and serum chemistry values on Sprague-Dawley rats, while used krill (Euphausia superba) meal diet for 5 weeks. Seven-week-old male rats were divided into four groups (n=6) and fed experimental diets containing three different krill meal contents and control group; 10.0% krill meal (G10), 20.0% krill meal (G20), 30.0% krill meal (G30), and control group (GC). Concentrations of non-esterified fatty acid (NEFA), blood urea nitrogen (BUN) in serum were significantly lower in the G10, G20, G30 than GC group (p<0.05). Statistically significant differences, at the confidence level of 95%, for the creatinine, uric acid, electrolyte (T-Ca) parameters in the sera were observed in G20 group, G30 group. The concentration of electrolyte (Pi) in serum was no significant difference among the groups (p<0.05). The results indicate that a krill meal diet was effectively reduce the NEFA.

        두유 커드를 생산하는 김치 유래 젖산균의 동정

        본 연구의 목적은 두유 curd를 형성하는 미생물을 분리하는 것이다. 두유 curd를 형성하는 미생물은 채소를 젖산균으로 발효시킨 전통적인 한국의 음식, 김치로부터 분리하였다. 분리 균주 196개 중 10개의 균주(strain No. 2-2-2, 2-15-2, 2-18-1, 2-19-2, 3-4-1, 3-4-2, 3-8-1, 3-8-3, 3-17-1, 4-39-5)가 단단한 두유 curd를 형성하였고 분자생물학적?생화학적 분석법에 의해 동정되었다. 분리균주로부터 추출한 genomic DNA는 16S rDNA 지역의 PCR 증폭을 위한 주형으로 사용하였다. GenBank 데이터로 16S rDNA 염기서열을 비교한 결과, 분리 균주들은 Leuconostoc mesenteroides group과 Lactobacillus sakei group으로 동정되었다. 두유 curd를 형성하는 균주들의 계통 발생학적 위치와 분류군은 neighbor-joining 방법을 이용하여 확인하였다. 또한, L. mesenteroides group은 생화학적 특성에 의해 L. mesenteroides subsp. dextranicum으로 동정되었다. 하지만 L. sakei group은 생화학적 특성 비교시 다양성을 보여 Lactobacillus sp.로 명명하였다. The purpose of this study was to isolate soy curd forming bacterial strains. Soy curd forming bacteria were isolated from Kimchi, a traditional Korean vegetable food that is fermented using lactic acid bacteria. Among 196 bacterial strains, ten isolates (strain No. 2-2-2, 2-15-2, 2-18-1, 2-19-2, 3-4-1, 3-4-2, 3-8-1, 3-8-3, 3-17-1, 4-39-5) formed firm soy curd. The isolated bacterial strains were identified by molecular biological and biochemical analyses. The genomic DNAs extracted from the isolated bacterial strains were used as a template for PCR amplification of 16S rDNA region. By comparing the results of the 16s rDNA sequences with GenBank data, the isolated strains were identified as Leuconostoc mesenteroides group and Lactobacillus sakei group. The phylogenetic position of soy curd forming strains and their related taxa were investigated using neighbor-joining method. L. mesenteroides group was further identified as L. mesenteroides subsp. dextranicum based on biochemical properties. L. sakei group was named Lactobacillus sp., because it showed a variety of biochemical properties.

In this study, the energy rating of Apartment building is evaluated by applying various thermal insulators and window glasses.

        Korea is classified as one of water deficient countries. A great deal of water use is used in the buildings. Hence the developed and improved sanitary plumbing system application is essential in building facility design for water use saving. The totally integrated pluming system for cooling, heating and hot water was adopted on design step to reduce the water use quantity. These systems are very effective in not only energy saving but environmental pollution reduction due to excessive waste water. According to appropriate sanitary plumbing system, the amounts of water use reduction and energy saving was studied in a government office building case.

        복분자 열매(Rubus coreanus Miquel)의 항산화 활성 및 생리활성물질

        The black raspberry (Rubus coreanus Miquel) contains anthocyanin, tannins, gallotannin, gallic acid, ferulic acid and phenolics. It brightens the eyes and protects the liver and kidneys. It was effective for anti-aging. Thus, the purpose of this study was to inform the excellence of black raspberry and to screen antioxidant activity to ensure the possibility as a functional material. In this study, bioactive compounds in black raspberry were determined. Additionally, black raspberry was extracted by CM (chloroform:methanol, 2:1, v/v), 70% methanol and 70% ethanol, and were investigated and compared in vitro methods. Total phenol and flavonoid contents were measured to compare each different solvents. In addition, DPPH radical scavenging activity and ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power were determined to measure the antioxidant activity. The results were the highest in 70% ethanol extracts, and the higher the concentration showed a significantly higher antioxidant capacity. When observed the relationship between the study, antioxidant activity of black raspberry was supposed to affect by the anthocyanin, phenol and flavonoid contents.

        블랙 초크베리 추출물의 항산화 활성 및 아질산염 소거 활성

        The purpose of this study was to measure the antioxidant activity and bioactivity of black chokeberry (Aronia melanocarpa). The black chokeberry was known to contain many physiologically active substance, such as ascorbic acid, anthocyanin, quercetin. We extracted black chokeberry powder from four solvents such as chloroform:methanol(CM, 2:1, v/v), distilled water (DW), 70% ethanol, 70% methanol. After that, we determined anthocyanin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power and Nitrite scavenging activity of black chokeberry extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of black chokeberry extracts was the highest in 70% methanol (p<0.05). The results suggest that black chokeberry can be used as nutraceutical foods and natural antioxidant.

        Culture Condition for Listeria monocytogenes 1421 Biofilm Formation and the Effect of Kimchi on Biofilm

        식품으로부터 유래되어 치명적인 질병을 유발하는 Listeria monocytogenes는 다양한 지지체 위에 biofilm을 형성 할 수 있다. 이러한 biofilm은 여러 가지 소독제나 살균과정으로부터 L. monocytogenes의 생존력이나 저항성을 증가시켜 준다. 본 연구에서도 L. monocytogenes는 다양한 배양용기에서 biofilm을 형성하였으며, BHI를 배지로 하여 30℃에서 4일 동안 배양하였을 때 최대의 biofilm을 형성하였다. L. monocytogenes의 biofilm의 형성에 미치는 김치의 효과를 살펴본 결과 김치를 첨가함에 따라 biofilm의 양이 줄어들기도 하고 늘어나기도 하여, 김치에 따라 다른 영향을 미치는 것으로 여겨졌으며, 이러한 결과는 L. monocytogenes의 생육에 미치는 김치의 효과와는 차이가 있었다. Listeria monocytogenes, a fatal food-borne pathogenic bacteria, can form a biofilm on many different supports. The biofilm gives L. monocytogenes more viability and resistance to disinfectants and sterilization procedures. L. monocytogenes formed biofilms on various culture vessels tested in this experiment and showed the maximum amount of biofilm when it was cultured for 4 days at 30℃ in BHI broth. In this study, biofilm formation was stimulated or inhibited by addition of different Kimchi samples. That was not in accordance with the effect of Kimchi on the growth of L. monocytogenes.

        어성초(Houttuynia cordata Thunb) 유산균 발효물의 항산화 활성

        본 연구에서는 항산화 활성을 가지고 있는 것으로 이미 알려진 어성초의 효능을 더욱 증가시켜 고부가가치를 지닌 기능성 원료로써 활용하고자, Leuconostoc mesenteroides 4395와 Lactobacillus sakei 383 두 유산균을 이용하여 발효시킨 후 그 추출물의 항산화 활성을 측정하였다. 항산화 활성 척도로써 DPPH 라디칼 소거능을 측정하였는데 10일간의 발효에서 두 균주 모두 발효 5일차에, 발효온도는 Leuconostoc mesenteroides 4395가 30℃에서 71.67±0.52%, Lactobacillus sakei 383는 35℃에서 70.11±0.67%로 각각 최대의 활성이 나타났으며, 두 가지 균 모두 대조군인 미발효물에 비해 DPPH 라디칼 소거능이 약 20% 상승하였다. 총 페놀함량에서는 대조군이 35.90±0.61 mg/g 함유한 반면에 발효한 후 두 균주 모두 약 10 mg/g씩 증가하였고, 총 플라보노이드 함량에서도 대조군의 21.69±1.52 mg/g에 비해 약 6 mg/g 이상 증가함을 나타내었다. SOD 유사활성에서는 발효의 효과가 크게 나타나지 않았으나, 대조군에 비해서는 미약하게 증가한 활성을 나타내었다. 이러한 결과들은 어성초의 유산균 발효물이 발효하지 않은 어성초에 비해 항산화 활성이 더욱 높아, 고부가가치를 지닌 기능성식품의 원료로써 활용할 가능 성이 있는 것으로 판단되었다. This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-oxidative activity of extracts from Houttuynia cordata Thunb. Houttuynia cordata Thunb. was fermented by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-oxidative activities of Houttuynia cordata Thunb. extracts were analyzed both before and after fermentation. Anti-oxidative activity was determined by in vitro assays to measure 1,1-diphenyl- 2-picrylhydrazyl (DPPH) scavenging and super oxide dismutase (SOD)-like activities, and by determining total flavonoid and total phenolic compound contents. The extracts of fermented Houttuynia cordata Thunb. had higher anti-oxidative activity than the unfermented control. The DPPH scavenging activity of the extracts after fermentation by Leuconostoc mesenteroides 4395 at 30℃ for 5 days was 71.67±0.52%, and after Lactobacillus sakei 383 fermentation at 35℃ for 5 days was 70.11±0.67%; these activities were both about 20% higher than the control. Increases of about 10 mg GAE/g of total phenolic compounds were found in both fermented extracts and both contained about 6 mg quercetin equivalents/g of total flavonoids, compared with 35.90±0.61 mg/g and 21.69±1.52 mg/g in the control, respectively. These results also suggested that fermentation time and temperature were important factors in determining the anti-oxidative effect of extracts from fermented Houttuynia cordata Thunb. These findings should be valuable for the development of medicines or functional foods with antioxidative activity.

        Bacillus subtilis MC31 와 Lactobacillus sakei 383의 혼합배양으로 향상된 풍미와 GABA 함량을 지닌 청국장의 품질 특성

        청국장은 혈전용해능효과, 항산화효과, 항암효과 등의 여러 가지 기능성을 가지고 있으나 특유의 불쾌취로 인해 어린이들은 좋아하지 않는다. 청국장의 GABA함량과 풍미를 증진시키기 위하여 B. subtilis MC31과 L. sakei 383을 이용하여 혼합배양조건을 연구하였다. 두 균주의 비율이 1:1일 때 불쾌취가 크게 감소되었으며 점질물의 양과 점도는 일반 청국장과 비슷하였다. B. subtilis MC31과 L. sakei 383으로 37℃에서 72시간동안 발효하여 얻어진 청국장의 GABA함량이 가장 높게 나타났다. 한편, 청국장 발효 기간 동안 혼합 발효 청국장의 생균수는 B. subtilis MC31가 6일째에 최대치(log 9.13 CFU/g)를 나타냈으며 L. sakei 383은 2일째에 최대치(log 6.78 CFU/g)를 나타내었다. 혼합 발효 청국장의 일반성분으로 수분은 61.71%, 조회분은 2.05%, 조단백은 17.54%, 조지방은 8.36%, 조섬유는 1.95% 의 함량을 나타내었다. 아미노태질소는 혼합 발효한 청국장이 B. subtilis MC31을 단일 발효한 청국장보다 함량이 낮았으며 암모니아태 질소와 환원당은 혼합 발효한 청국장이 일반 삶은 콩보다 함량이 높았다. 아미노산 분석기로 측정된 Glutamic acid와 GABA의 함량은 각각 1.40 mg/g 과 0.47 mg/g 이었다.이러한 결과를 보아 단일발효보다 B. subtilis MC31과 L. sakei383를 1:1 혼합발효시 불쾌취가 줄고 발효가 진행됨에 따라 GABA함량이 가장 높게 증가하는 것을 알 수 있었다. Chungkookjang has several functional properties, such as fibrinolytic activity, anticancer effects, and antioxidant effects. However, children do not like Chungkookjang because of its foul odor. A mixed culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 was used to improve the production of GABA in Chungkookjang and its flavor. Most of the foul odor of Chungkookjang was removed. The slime content and viscosity of Chungkookjang fermented in the mixed culture were similar to those of commercial Chungkookjang when B. subtilis MC31 and Lactobacillus sakei 383 were inoculated in a 1:1 ratio. The maximum GABA content was obtained when Chungkookjang was fermented with B. subtilis MC31 and L. sakei 383, which was fermented at 37℃ for 72 hr. During the period of fermentation, the viable cell number of B. subtilis MC31 reached a peak (log 9.13 CFU/g) at six days, and L. sakei 383 reached a peak (log 6.78 CFU/g) at two days. The moisture, crude ash, crude protein, crude fat, and crude fiber contents were 61.71%, 2.05%, 17.54%, 8.36%, and 1.95%, respectively. The amino-type nitrogen content of Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 was less than Chungkookjang fermented by B. subtilis MC31 alone. The ammonia-type nitrogen and reducing sugar content of the Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 were higher than that of steamed soybean. The glutamic acid and GABA content detected with an amino acid analyzer were 1.40 ㎎/g and 0.47 ㎎/g, respectively. These results suggest that fermentation with B. subtilis MC31 and L. sakei 383 in a 1:1 ratio removes more of the foul odor and increases the GABA content compared with single fermentation.
