        부상분리 기술을 이용한 현장형 조류수거시스템의 발전 과정 및 개발 방향

        현장형 조류수거시스템(이하 OACS)은 녹조 현상이 심각할 때 긴급 대책으로 투입되는 대안 중의 하나로, 대규모 수역에 투입되는 OACS가 갖추어야 할 조건은 신속대응성, 효율성 및 대용량이다. 부상분리 기술을 이용한 OACS의 성능은 부상․포집․수거 공정의 효율에 의해 결정되며, 처리용량 및 효율성에 가장 중요한 인자는 포집과 수거이다. 이들이 자동화되면 전체 작업속도와 일일 처리수량이 증가한다. OACS에 적용된 기술은 진보될수록 장비의 단순화․소형화와 포집과 수거공정 자동화가 발전하여 신속대응성과 효율성이 향상되었으며, 잔류슬러지를 크게 줄여 친환경성과 수질 지속효과를 향상시켰고, 슬러지 수거량도 증가하여 대용량이 실현되는 방향으로 개선되었다. 그 결과 시스템은 조류증식비율을 극복할 수 있는 조류제거량을 달성할 수 있었고, 처리용량 대비 운영비가 감소되어 기술의 경제성을 크게 향상시켰다. On-site Algae Collecting System (OACS) is one of urgent countermeasures to clean the raw water when the algae blooms severely. Rapid reaction capability, high efficiency and large capacity are required when applying OACS to large water areas. The total performance of OACS are always determined by unit process named Flotation, Trapping and Collection. The working efficiency and daily treatment quantity of OACS can be increased when it runs automatically. As the rapid development of OACS technology, in the first place, equipment are miniaturized and simple. And in the second place, automation process from Trapping to Collection are advanced. So, They produce results higher working efficiency, smaller residual sludge on treated water, system’s advanced environmental friendly features and the increased amount of sludge by Collection process to achieve large capacity. Now OACS has overcome the algae multiplication rate to ensured the amount of removal algae. In another aspect, it is high economically feasible because of reducing operation cost against the large capacity.

        An analysis of the general composition of wheat flour (hard and semihard wheat flour which is sold in the market was made. Water soluble proteins were investigated by using discelectrophoresis in 7.5% polyacrylamide gel. The results obtained were as follows. 1) Protein contents from first grade of semi hard wheat flour was 8.3-8.6 percent. 2) 7.5 Percent acrylamide gel concentration gives the best separation of water soluble protein from wheat flour. 3) Eight bands of protein were found in hard wheat flour using 2% NaCl solution extraction. Six bands were found in semihard wheat flour.

        In this study, the general composition, amino acid composition and separated protein patterns, using discelectrophoresis, of Dioscorea Japonica Thunberg (yam) were investigated. The results obtained were as follows. 1) Protein content of yam was as high as 3.3 percent compared to that of 1.9 percent of irish potato and 0.7 percent of sweat potatoes. 2) In the amino acid analysis of Yam protein, 17 kinds of amino acid were determined, especially among them tryptopham which is lacking in cereals was as abundant as 0.27 percent. 3) 7.5 percent acrylamide gel concentration gives the best separation of yam protein. One band was found in raw yam using 2% NaCl solution for extraction. Seven bands were found in yam powder using either 2% NaCl solution or 60% ethanol. 4) One band of glycoprotein was found when the acrylamide gel was stained with shiff's reagent.

        참깨박(粕) 단백질의 분리와 조성

        White and black sesame produced in Korea were defatted with ethyl ether or n-hexane. Defatted sesame meal was extracted with water and salt solution, and protein extraction was precipitated at various pH 1 through 12, with trichloro acetic acid (TCA), tannic acid and ammonium sulfate, respectively. Protein was purified by Sephadex A-25, G-75, G-100 and G-200, and identified its protein fraction by polyacrylamide gel electrophoresis. Amino acids composition of protein in white sesame was analyzed by automatic amino acid analyzer. Protein contents of white sesame, black sesame and sesame meal are 20.5%, 19.2%, and 44.7%, respectively. n-Hexane was the most suitable solvent for extraction of oil from sesame. Crude protein precipitation was better in higher pH. The protein extraction was more effective with the solution containing sodium chloride under the pH 8. Globulin in total protein was high and prolamin was less than in other cereal proteins. Glutamic acid contents of white sesame and sesame globulin were 17.1%, and 20%, respectively. Both proteins contained relatively high levels of essential amino acids. 12-13 bands w ere found in water soluble protein and 2 bands in salt soluble protein were detected by the disc gel electrophoresis, and were identified in both of white and black sesame. The salt soluble protein of white sesame could be purified by Sephadee G-100 and G-200.

      • Phase Titration에 의한 酒類中의 알콜 含量의 分析

        The alcohol content of alcoholic beverages was determined using benzyl alcohol in a phase titration. 1) Benzyl alcohol is added to the unknown beverages until distinct turbidity appears, caused by the separation of a second phase. The amount of benyl alcohol necessary to cause turbidity is a function of the ethanol content of the Sample. Volume Percent of alcohol in alcoholic beverages is determined by comparing the titer with the standard colibration curve. 2) Nine samples of alcoholic beverages were taken in downtown for determination of alcohol content. 3) For determination of alcohol content of "Tack Joo", "Tack Joo" was centrifuged or filtered before phase titration. 4) The procedure is simpler and quicker than the standard pycnometric method, and gives comparable results.
