        Diazinon이 계배(鷄胚)의 기형 유발에 미치는 효과 I. 계배(鷄胚) 형태와 콜린성 봉쇄약물에 미치는 효과

        Teratogenic effects of diazinon were assessed on morphology of chick embryos cholinergic blocking agents. Diazinon at doses ranging from 25 to $2000{\mu}g/egg$, was injected on Day 3 of incubation. $TD_{50S}$, were different for the various teratogenic sings such as wry neck, micromelia, abnormal feathering, abnormal beak and curled claws. The threshold dose for wry neck was higher than the threshold dose for other signs; $40{\mu}g/egg$ produced substantial micromelia, abnormal feathering, abnormal beak and curled claws, but gave no sings of wry neck. In contrast to the teratogenic doses, the $LD_{50}$ of diazinon was very high(above $2000{\mu}g/egg$). One of the characteristics of diazinon-induced teratogenesis was reduction of body weight(78.8%) and body length(73.8%). Maximal teratogenic effects, scored as sings of retarded growth, wry neck, micromelia, abnormal feathering, abnormal beak, and curled claws, were produced when the insectcide was administered on the third or fourth day. The threshold dose for type II teratogenic sings including wry neck and short neck was higher than for type I including micromelia and abnormal feathering. Morphorlogical studies, using atropine and gallamine, suggested that nicotinc but not muscarinic receptors may be involved in the mechanism of diazinon-induced type II malformations.

        쥐(rats)의 full-thickness skin grafts에 대한 hyperbaric oxygen과 α-tocopherol의 효과

        To document that effects of hyperbaric oxygen(HBO) and ${\alpha}-tocopherol$ on full-thickness skin grafts in rat, we performed full-thickness skin grafts bilaterally on each rats. The HBO-treated rats were received HBO twice daily for 90 minutes at 2 ATA. Surgical control rats were not treated with HBO. ${\alpha}-tocopherol$ treated rats were received the agent via oral gastric tube daily for 3 days preoperative and a fourth dose 1 to 2 hours postoperative. HBO plus ${\alpha}-tocopherol$ treated rats were received HBO and ${\alpha}-tocopherol$ as mentioned above. Biopsy specimens were taken from each rat at the time of grafting and on days 2, 4, 7, 10, 14, 21, and 28, then were processed for tissue-concentration of total glutathione(GSHt), oxidized/reduced glutathione level, and thiobarbituric acid-reactive substance(TBARS) levels. The percentage of viable graft on day 10 ranged from 67 to 93%, and was not significantly different among the each other groups. The percentage of viable graft were, however, higher in HBO plus ${\alpha}-tocopherol$ treated rats(78.6%) than in HBO alone treated rats(59.1%), ${\alpha}-tocopherol$ alone treated rats(66.7%) and surgical control rats(58.2%). TBARS concentration had a significant increase from preoperative concentration at day 2, and peak concentration at day 4(p<0.01). Concentration then decreased to preoperative concentration at day 28. GSHt concentration of free skin graft had a similar patteren of change in four groups and decreased significantly from preoperative concentration at day 2, returning to preoperative concentration by day 7(surgical control, HBO-treated, and ${\alpha}-tocopherol-treated$, alone) and 28(HBO plus ${\alpha}-tocopherol-treated$). Percentage of the concentration of reduced glutathione decreased in surgical control, HBO-treated and, ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treared$(p<0.01) on day 7 after surgery, whereas the concentration of oxidized increased significantly in HBO-treated(p<0.05), ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treated$(p<0.01).

        내측광근 강화운동을 이용한 슬개-대퇴 동통 증후군 치험 2례

        슬개-대퇴 통증 증후군 환자 2례에 내측광근 강화 운동을 이용하여 치료한 결과, Knee X-ray(Skyline view) 상의 일치각 및 NRS, WOMAC Index의 호전을 확인하였다. 이에 저자는 본 증례를 보고하는 바이다. Objectives: The purpose of this case report is to investigate the effect of exercise for vastus medialis to knee pain patients with patello-femoral pain syndrome. Methods: Two patients with patello-femoral pain syndrome were treated by exercise for vastus medialis. To measure the outcome of the patients' improvement, we observed the X-ray of both knee sky-line view, Numerical Rating Scale(NRS), and Western Ontario and McMaster Universities(WOMAC) index. Results: After being treated by exercise for vastus medialis, the patients' X-ray of both knee sky-line view, Numerical Rating Scale(NRS), and Western Ontario and McMaster Universities(WOMAC) index were significantly improved. Conclusions: These results suggest that exercise for vastus medialis is effective treatment to patients with patello-femoral pain syndrome.

        Phytomitogen에 의한 기니픽 임파구의 Blast Transformation : I. 유사분열에 미치는 배지, 유사분열촉진물질 및 세포농도의 효과

        The present study has been carried out to investigate the optimal condition on the blastogenesis of guinea pig lymphocytes. A microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the in vito optimal condition of guinea pig lymphocytes. Data were presented to show many variables that are Involved in studying the responses of guinea pig lymphocyte in a microculture system to the stimulation of Concanavalin A(Con A) and lipopolysaccharide (LPS). Analysis indicated that the conditions for optimal Con A as measured by incorporation of $^3H$-TdR include : (1) use of RPMI-1640 as culture medium, (2) use of $6{\mu}g$ of Con A, per culture, (3) use of $1{\times}10^6$ cells per culture. Conditions for optimal stimulation with LPS mitogen were similar to those used for Con A.

        기니픽그에서의 실험적 trichothecene (T-2) 독소중독증

        Guinea pigs were administrated with T-2 toxin at a rate of 1 and 0.6mg/kg body weight per day for 21 days to study the immunological and pathological effects of T-2 toxin in guinea pigs. Blood was collected before T-2 treatment and on days 7, 14 and 21 of the trial for hematological and biological examinations and for the mitogen assay using lymphocytes. Myeloid: erythroid ratios were examined from the fernur bone marrow samples taken a day before T-2 toxin treatment began, on day 12 and at death. Guinea pigs received with 1mg/kg body weight of T-2 toxin daily showed leukopenic, lymphopenic and anemic signs on day 7 and 14. The mitogenic responses to the T-cell mitogen, Concanavalin A and B-cell mitogens, lipopolysaccharide were significantly depressed on day 7. Histologically, marked cellular damages including karyorrhexis and depletion of lymphocytes were observed in the actively dividing cells of the gastrointestinal tract, lymph node, spleen and bone marrow of guinea pigs.

        Vomitoxin의 면역 독성효과

        The present study were carried out to investigate the effects of Vomitoxin(Deoxynivalenol, DON) induced dysregulation of IgA synthesis. The data presented here demonstrate that exposure to dietary DON increases serum IgA and concurrently decreases IgG and IgM. Peyer's path(PP) and splenocytes from mice fed dietary DON produced increased IgA in mitogen stimulated and macrophage(MØ) perform essential role for Ig synthesis and was affected by the presence of DON. The synthesis and secretion of IgM and IgG under the presence of DON do not require T cell helper activity, whereas those of IgA do require such activity. These results suggest that dietary DON alters the regulation of IgA production and may act as an immunostimulator.

        과잉치 치수유래 줄기세포의 분화제 처리 기간에 따른 상아모세포 발현 특성

        본 연구의 목적은 치과분야에서 줄기세포 공급원으로써 과잉치의 활용 가능성을 알아보고자 Real Time Quantitative Reverse Transcription Polymerase Chain Reaction (Real Time qRT-PCR)법을 이용하여 발치된 과잉치 치수 유래 줄기세포 (supernumerary dental pulp stem cells, sDPSCs)로부터 상아모세포로의 분화여부를 관찰해 보는 것이었다. 이를 위해 상아모세포의 대표적인 발현인자로 알려진 alkaline phosphatase (ALP), osteocalcin (OC), osteonectin (ON), dentin matrix acidic phosphoprotein 1 (DMP-1) 그리고 dentin sialophosphoprotein(DSPP)의 발현을 분화제 처리 후 0일, 8일 그리고 14일째에 각각 Real Time qRT-PCR 법을 통해 상대적인 mRNA의 발현 양을 비교하여 변화 양상을 알아보았다. 또한 Alizarin-red solution 의 염색을 통해 sDPSCs가 분화제 처리 7일, 14일, 21일 그리고 28일째에 석회화 결절을 형성하는 정도를 시각적으로 확인해 보았다. Real Time qRT-PCR 결과 분화제 처리 8일째에 가장 높은 발현 양을 보이다가 14일째에 감소하는 추세를 나타내었으며, Alizarin-red solution 염색 결과는 7일째부터 흐리게 나타나다가 14일째에는 배지 전반에 걸쳐 진하게 염색되는 소견을 보였다. 따라서, sDPSCs를 이용한 연구에서 Real Time qRT-PCR법을 위한 분화제의 처리 기간은 8일 정도가 적절하며, Alizarin-red solution 염색은 14일이 적당한 것으로 사료된다. The aim of this study was to investigate the possibility of the supernumerary teeth for the stem cell source in dentistry. The Real Time Quantitative Reverse Transcription Polymerase Chain Reaction (Real Time qRT-PCR) method was used to evaluate the differentiation toward the odontoblast of the supernumerary dental pulp stem cells (sDPSCs). Supernumerary dental pulp stem cells were obtained from 3 children (2 males and 1 female, age 7 to 9) diagnosed that the eruption of permanent teeth was disturbed by supernumerary teeth. The common genes for odontoblasts are alkaline phosphatase (ALP), osteocalcin (OC), osteonectin (ON), dentin matrix acidic phosphoprotein 1 (DMP-1), dentin sialophosphoprotein (DSPP). The sDPSCs were treated for 0 days, 8 days and 14 days with additives and then Real Time qRT-PCR was performed in intervals of 0 days, 8 days and 14 days. The alizarin-red solution staining was performed to visualize the stained color for the degree of calcification at 7 days, 14 days, 21 days and 28 days after treating additives to the sDPSCs. From the result of the Real Time qRT-PCR, the manifestation exhibit maximum value at 8 days after additive treatment and shifted to a decrease trend at 14 days. Alizarin-red solution staining exhibit light results at 7 days after staining and generalized dark result at 14 days. Consequently, in studies with sDPSCs, appropriate treatment time of additives for Real Time qRT-PCR is 8 days. Also, a suitable period of Alizarin-red solution staining is 14 days.

        보조 전원을 이용한 12 펄스 다이오드 정류기를 사용하는 추진시스템의 고조파 및 토크 리플 저감

        The input current of three phase rectifier which is mainly used in the propulsion system of the electric propulsion ship includes a variety of low order harmonics. To reduce these harmonics, the power conversion system, used in the large vessels which high power is required, is currently used the rectifiers of 12-pulse output, but it still has a problem that occurs $12{\pm}1$ harmonics. Also, in the case of the direct torque control technique which is widely used for the speed and torque control, the torque ripple is severe and the input current of motor has greatly included harmonics by the switching of the inverter. In order to reduce harmonics and improve the performance of torque control, this paper presents that the auxiliary supply assisted into the 12-pulse rectifier of the electric propulsion system using direct torque control technique. We confirm the validity of the proposed method through the simulation under the environment of a real vessel system. 전기추진선박의 추진시스템에 주로 사용되는 3상 정류기의 입력전류는 다양한 저차 고조파를 포함하고 있다. 이러한 고조파 저감을 위해서 대전력이 필요한 대형 선박에 사용되는 전력변환장치는 12펄스 출력의 정류기가 현재 사용되고 있지만 여전히 $12{\pm}1$차의 고조파가 발생되는 문제점이 있다. 또한, 속도 및 토크 제어기법으로 널리 사용되고 있는 직접토크제어기법의 경우에는 토크 리플이 심하고 전동기의 입력전류는 인버터의 스위칭에 의해 고조파를 크게 포함하고 있다. 본 연구에서는 직접토크제어기법을 이용하는 전기추진시스템의 12펄스 정류기에 보조 전원을 공급하여 고조파를 저감하고 토크 제어성능을 향상시켰으며 실선시스템 환경에서 시뮬레이션을 통해 그 유효성을 입증하였다.

        마우스에서 diazinon, toxaphene 과 endrin 단독 혹은 그 혼합물 독성의 대사

        The effects of mixtures of diazinon(DA;5mg/kg), toxaphene(TOX;40mg/kg) and/or endrin(END; 5mg/kg) on the hepatic mixed-function oxygenase(MFO) system were stuided in ICR mice(18~22g) by oral intubation daily for 7 days. In general, TOX and TOX-containing mixtures were found to induced the metabolism of aminopyrine(22~60%), aniline(42~85%), phenacetin(145~194%) and benzo [a]pyrene(158~210%), and pentobaribtal biotransformation in the 9,000g liver supernatants and to increased the hepatic cytochrome p-450 contents(47~89%). Results of these may be, at least in part, associated with the MFO system. TOX pretreatment increased the aliesterase activity in the serum and liver homogenates and supernatants by 23~145%. The toxicity of TOX and TOX-containing mixtures would be lower than that of diazinon because of TOX-induced increase in the metabolism of diazinon(DA) or diazioxon(DO) and capability of TOX to stimulate the metabolism of diazinon and diazioxon and provide a pool of non-critical enzymes. These results suggest that this information might be helpful in the evaluation of the potential hazard due to occupational and/or environmental exposures to pesticides and their mixtures.

        The present study has been carried out to investigate the optimal condition on lymphocyte blastogenesis of rabbit lymphocytes, whole blood culture and microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the In vitro optimal condition of rabbit lymphocytes. Data were presented to show many variables that are involved in studying the phytohemagglutinin(PHA) and lipopolysaccharide(LPS) response of rabbit lymphocyte in a microculture system. Analysis indicated that the conditions for optimal PHA as measured by incorporation of $^3H$-TdR include: (1) use of RPMI-1640 as culture medium. (2) use of $6{\mu}g$ of PHA, per culture. (3) 48-hours culture period. Conditions for optimal stimulation with LPS mitogen were similar to those used for PHA.
