        Listeria monocytogenes YM-7의 생리적 특성

        리스테리아 증세로 의심되는 환자의 혈액으로부터 분리한 균은 저온에서의 운동성 시험, 용혈능 검정시험, 당 이용성, CAMP 시험의 결과 전형적인 L. monocytogenes의 특성을 보여 L. monocytogenes YM-7로 동정하고 명명하였다. L. monocytogenes YM-7의 세균학적 특성과 이 균이 생산하는 용혈독소의 이화학적 특성은 다음과 같다. L. monocytogenes YM-7 균주는 Gram양성 간균으로, 운동성 측정 결과 반고체 배지에서 우산 모양의 층이 생겼고, 최적증식조건은 $37^{\circ}C$, 식염 농도 $0^{\circ}C$, pH 8.0이었고, 이 균의 생육 가능범위는 $8-40^{\circ}C$, 식염 농도 $0.7\%$, pH 5.0-10.0였다. L. monocytogenes YM-7 균주가 생산하는 용혈독소는균 생육곡선의 정지기 부근에서 최대 활성을 나타내었다. 이 용혈독소는 비교적 열에 불안정하였으나, $37^{\circ}C$에서 180분 후에도 용혈활성이 남아 있었다. 그리고 용혈독소는 pH 6.0-8.0에서 안정하였으나 그 외의 pH 영역에서는 매우 불안정 하였다 Listeria monocrogenes has been increasingly recognized as an important food poisoning and pathogenic bacterium which is Gram positive, non spore forming and facultative anaerobic rod shape. Bacteriological and physiological characterization of L. monocytogenes YM-7 isolated from a patient was performed. Optimum growth condition of 1. monocytogenes YM-7 was at $37^{\circ}$, pH 8.0 and $0\%$ sodium chloride in tryptic soy broth, and then it grew pretty well in the range of $8-40^{\circ}C,\;pH 5.0-10.0\;and\;up\;to\;7\%$ of sodium chloride in the medium. The highest hemolysin activity of hemolysin produced by L. monocytogenes YM-7 was shown in the stationary phase of its growth. Hemolysin produced by the isolated strain was stable at $4^{\circ}C\;and\;pH\;6.0-8.0, while it was gradually unstable by increasing the storage temperature.

        갯벌에서 분리한 3,4-Dichloroaniline 분해 미생물의 특성

        3.4-dichloroaniline (DCA)를 함유한 최소배지에서의 집식배양과 배양 후 HPLC에 의한 잔류분석을 통해 3,4-DCA의 분해 능력이 우수한 균주 Arthrobacter sp. YM-14를 여천석유화학공단 인근의 갯벌에서 분리하였다. 분리균 YM-14는 1/10 LB 배지에 함유된 50 ppm의 3,4-DCA를 12 시간 만에 완전히 제거하였다. 이외에도 분리균 YM-14는 3-chloroaniline(CA), 2,5-DCA 및 3,5-DCA의 분해 활성을 나타내었으나 2-CA, 4-CA와 2,4-DCA에 대한 분해활성을 가지고 있지는 않았다. 또한, 분리균 YM-14에서 3,4-DCA의 유도에 의한 catechol 1,2-dioxygenase 활성의 증가가 관찰되었다. 이러한 결과는 catechol 1,2-dioxygenase이 3,4-DCA 분해에 관여하는 중요한 효소군중의 하나로 생각된다. The compound 3,4-dichloroaniline (DCA) is an aromatic amine used as an intermediate product in the synthesis of herbicides, azo-dyes and harmaceuticals. It is also a degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as active agent in the cosmetic industry. 3,4-DCA, however, is considered potential pollutants due to their toxic and recalcitrant properties to humans and other species. A bacterium capable of growth on 3,4-DCA was isolated by dilution method from 3,4-DCA-containing enrichment culture. Finally, a strain, YM-14, capable of degrading efficiently 3,4-DCA was isolated from a sandbank. The isolated strain, YM-14 was identified to be Arthrobacter sp.. Fifty ppm 3,4-DCA in 1/10 LB media was completely degraded by the growth of Arthrobacter sp. YM-14 for 12 h at $30^{\circ}C$. The isolated strain is capable of growth on 3,4-DCA as sole carbon source and also able to degrade other chloroaniline compounds. Also, the isolated strain showed high level of catechol 1,2-dioxygenase activity by 3,4-DCA exposure. The catechol 1,2-dioxygenase was supposed to be ones of the important factors for 3,4-DCA degradation.

        Studying the Genetic Diversity and Phenetic Relationships of Porphyra yezoensis Populations in Korea Using Random Amplified Polymorphic DNA (RAPD)

        김(Porphyra yezoensis)은 김속의 홍조류이다. RAPD (random amplified polymorphic DNA) 마커를 이용하여 한국 내 네 집단의 표현형과 유전적 다양성을 조사하였다. 전체적으로 20 시발체로 김에서 55분절이 관찰되었다. 이들 밴드 중 30개(54.5%)는 다형성을 나타내었다. OPA-18-02 밴드는 낙동 김 집단에서만 증폭되었다. OPA-20-02 밴드는 서천 김 집단에서만 증폭되었다. 이 두 밴드는 특별한 집단을 구별해주는 특이밴드로 판정되었다. 대립유전자좌위의 수(Ae)는 1.161에서 1.293로 평균은 1.366였다. 서천 김 집단이 가장 높은 다형성을 나타내었다(0.163). 다른 집단과 격리되고 조간대에 위치한 낙동 김 집단은 가장 낮은 다형성을 나타내었다(0.092). 샤논의 표현형 다양성(I)은 서천 김 집단이 가장 높았다(0.238). 전체 유전적 다양도(HT)는 0.132(OPA-02)에서 0.420(OPA-19)로 나타났다. 대립유전자좌위에서 유전적 다양성(HS)은 0.059(OPA-18)에서 0.339(OPA-19)였다. 대립유전자좌위에 근거에서 전체 유전적 다양도에서 집단 간 차이(GST)는 0.012(OPA-11)에서 0.762(OPA-18)이였으며 평균은 0.415였다. 이는 전체 변이의 약 42%는 집단 간에서 발견된다는 것을 의미한다. 종 내 다양도의 58.5%는 집단 내에 있었다. 유전자 흐름(Nm)은 0.705로 낮았다. Porphyra yezoensis is a red algal species in the genus Porphyra. The phenetics and genetic diversity of four populations of P. yezoensis in Korea were reconstructed using random amplified polymorphic DNA (RAPD) markers. Overall, 55 fragments were generated among the tested P. yezoensis array with 20 OPERON primers. A total of 30(54.5%) of these bands were polymorphic. The OPA-18-02 band was amplified in the samples of Nakdong population and absent in them of other three populations. The OPA-20-02 band was only amplified in the Seocheon population. Both bands exhibited distinctive patterns in specific populations. The effective number of alleles per locus (Ae) ranged from 1.161 to 1.293 with a mean of 1.366. The Seocheon population had a high expected diversity (0.163). The Nakdong population was an isolated endemic and intertidal zone. Thus the narrow distributed Nakdong population had a low expected diversity (0.092). Shannon’s index of phenotypic diversity (I) of the Seocheon population (0.238) was the highest among all populations. Total genetic diversity (HT) varied between 0.132 for OPA-02 and 0.420 for OPA-19. The interlocus variation of genetic diversity (HS) was 0.059 for OPA-18 and 0.339 for OPA-19. On a per locus basis, the proportion of total genetic variation due to differences among populations (GST) ranged from 0.012 for OPA-11 to 0.762 for OPA-18 with a mean of 0.415, indicating that 42% of the total variation was found among these populations. In an assessment of the proportion of diversity present within this species, 58.5% (100%-41.5%) of genetic variation resided within the populations studied. The Nm was estimated to be low (0.705).

        송어양식장의 위해요소 관리를 위한 수질 모니터링

        Water quality has been considered to be one of sanitation standard operating procedures (SSOP) for hazard analysis critical control points (HACCP) application in aquaculture farms. This study was conducted to evaluate a hazard caused by water used in aquaculture farm of rainbow trout. The water quality was analyzed to investigate both physiochemical and bacteriological level in water samples collected from aquaculture farm of rainbow trout, Oncorhynchus mykiss. No significant difference were observed on water temperature and pH from season to season. However, the levels of dissolved oxygen were decreased as the outside temperature was increased, even if the levels were adequate for aquaculture. Also, other physiochemical analysis including biochemical oxygen demand (BOD), chemical oxygen demand (COD) and suspended solid (SS) revealed that the waters for aquaculture analyzed in this study was suitable for rainbow trout aquaculture. The bacterial analyses were also revealed that the waters for aquaculture were met to both coliform group (<18 MPN/100mL) and viable cell count (<100 CFU/mL). However, some of waste waters from aquaculture farms showed higher levels of BOD and COD than those of waste water standard (<2 ppm), suggesting that regular cleaning of fish tank and precipitation tank is needed.

        양식산 무지개송어에서의 항생제와 말라카이트 그린 잔류량 조사

        Products from aquaculture have sometimes been focused on the problems caused by the contamination of chemical agents as the use of chemical agents in aquaculture has been annually increased. The risk of contamination of products by chemical agents is greater in freshwater than in seawater. In order to evaluate the food safety of a fish grown in freshwater, we investigated the residues of antibiotics (tetracycline, oxolinic acid and ciplofloxacin) and malachite green in cultured rainbow trout, Oncorhynchus mykiss. Malachite green, which was prohibited in the application of aquaculture, was not detected in samples tested in this study. The residual content of tetracycline was determined to be less than the permissible amount, <0.2 mg/kg. The contents of ciplofloxacin was also less than the permissible amount, <0.1 ㎎/㎏. However, in case of oxolinic acid, one of samples was only exhibited higher content than the permissible amount (<0.1 ㎎/㎏). The results obtained in this study suggested that the control and regulation of chemical agents such as antibiotics was important to maintain a safe and worry-free seafood supply.

        본 논문에서는 진단 검사 의학 분야에서 통계적 지식이나 도구사용능력이 부족한 임상의학자들을 위해 개발된 통계적 데이터 분석 시스템을 소개한다. 개발된 시스템은 질병진단에 필요한 다중 바이오마커 데이터를 Luminex장비를 통하여 측정한 후, 분석이 용이한 형태로 변환하는 시각화 기술과 데이터에 내재된 패턴을 찾아내어 유용한 지식을 생성하는 데이터 마이닝 기술을 포함한다. 주요 모듈로는 마커선택, 데이터 시각화, 마커 평가 모듈로 구성되며 MATLAB을 기반으로 개발되었다.
