        Involvement of tumor necrosis factor receptor superfamily (TNFRSF) members in the pathogenesis of inflammatory diseases

        Current therapies for autoimmune diseases are notcures but merely palliatives, aimed at reducingsymptoms. For the most part, these treatmentsprovide nonspecific suppression of the immunesystem and thus do not distinguish between apathogenic autoimmune response and a protectiveimmune response. Recently emerging evidencenot only has indicated the involvement of membersof the TNF receptor/ligand superfamilies butalso has revealed exciting innovative strategies forthe treatment of autoimmune diseases and otherchronic inflammatory diseases without depressingthe immune response in general. In this review,we will discuss the regulatory mechanisms of TNFreceptor/ligand family members, such as HVEM/LIGHT, 4-1BB/4-1BBL, and GITR/GITRL that regulateT and B cell functions and participate in theprocess of inflammatory diseases. We will also discusshow intervening in the costimulatory pathwaysmediated by these molecules might havesome potential as a therapeutic approach to immunedisorders.

        Regulation of Inflammation by Bidirectional Signaling through CD137 and Its Ligand

        Although the majority of research on CD137 has been directed to T cells, it is becoming clear that this molecule has distinct functions in other lineages of cells, including nonhematopoietic cells. In particular, emerging evidence suggests that the CD137-its ligand (CD137L) network involving immune cells and non-immune cells, directly or indirectly regulates inflammation in both positive and negative manners. Bidirectional signaling through both CD137 and CD137L is critical in the evolution of inflammation: 1) CD137L signaling plays an indispensible role in the activation and recruitment of neutrophils by inducing the production of proinflammatory cytokines and chemokines in hematopoietic and non-hematopoietic cells such as macrophages, endothelial cells and epithelial cells; 2) CD137 signaling in NK cells and T cells is required for their activation and can influence other cells participating in inflammation via either their production of proinflammatory cytokines or engagement of CD137L by their cell surface CD137: 3) CD137 signaling can suppress inflammation by controlling regulatory activities of dendritic cells and regulatory T cells. As recognition grows of the role of dysregulated CD137 or CD137L stimulation in inflammatory diseases, significant efforts will be needed to develop antagonists to CD137 or CD137L.

        CD137-CD137 Ligand Interactions in Inflammation

        The main stream of CD137 studies has been directed to the function of CD137 in CD8+ T-cell immunity, including its anti- tumor activity, and paradoxically the immunosuppressive activity of CD137, which proves to be of a great therapeutic potential for animal models of a variety of autoimmune and inflammatory diseases. Recent studies, however, add complexes to the biology of CD137. Accumulating is evidence supporting that there exists a bidirectional signal transduction pathway for the CD137 receptor and its ligand (CD137L). CD137/CD137L interactions are involved in the network of hematopoietic and nonhematopoietic cells in addition to the well characterized antigen-presenting cell-T cell interactions. Signaling through CD137L plays a critical role in the differentiation of myeloid cells and their cellular activities, suggesting that CD137L signals trigger and sustain inflammation. The overall consequence might be that the amplified inflammation by CD137L enhances the T-cell activity together with CD137 signals by upregulating costimulatory molecules, MHC molecules, cell adhesion molecules, cytokines, and chemokines. Solving this outstanding issue is urgent and will have an important clinical implication.         Roles of Host Nonhematopoietic Cells in Autoimmunity and Donor Cell Engraftment in Graft-versus-host Disease

        Graft-versus-host disease (GVHD) is initiated when alloreactive donor T cells are primed by host APCs to undergo clonal expansion and maturation. Since there is a controversy regarding the role of nonhematopoietic cells in GVHD, we wanted to investigate the influence of MHC disparity on nonhematopoietic cells on the pathogenesis of GVHD in the MHC-haplomismatched C57BL/6 (H-2b) or DBA/2 (H-2d)→an unirradiated (C57BL/6×DBA/2) F1(BDF1;H-2b/d) murine model of acute GVHD (aGVHD) or chronic GVHD (cGVHD). Methods: We generated (BDF1→C57BL/6),(BDF1→DBA/2), and (BDF1→BDF1) chimeras and examined GVHD-related parameters and donor cell engraftment in those chimeras. Results: Using this experimental system,we found that 1) severe aGVHD across MHC Ag barrier depends on the expression of nonhematopoietically rather than hematopoietically derived alloAgs for maximal GVHD manifestations;2) host APCs were sufficient to break B cell tolerance to self molecules in cGVHD, whereas host APCs were insufficient to induce autoimmunity in aGVHD; 3) donor cell engraftment was greatly enhanced in the host with MHC-matched nonhematopoietic cells. Conclusion: Taken together, our results provide an insight how MHC disparity on GVHD target organs contribute to the pathogenesis of GVHD.

        Is CD137 Ligand (CD137L) Signaling a Fine Tuner of Immune Responses?

        Now, it has been being accepted that reverse signaling through CD137 ligand (CD137L) plays an important role in vivo during hematopoiesis and in immune regulation. However, due to technical difficulty in dissecting both directional signaling events simultaneously in vivo, most biological activities caused by CD137-CD137L interactions are considered as results from signaling events of the CD137 receptor. To make the story more complex, CD137−/− and CD137L−/− mice have increased or decreased immune responses in a context-dependent manner. In this Mini review, I will try to provide a plausible explanation for how CD137L signaling is controlled during immune responses.

        수목장지 유형별 만족도 조사 분석에 따른 활성화 방안 제안

        본 논문은 수목장의 활성화 방향을 제언하기 위한 연구로서 수목장지의 유형별로 만족도를 비교 측정하는 연구를 진행하였다. 이를 위하여 수목장지를 4가지 유형으로 분류하고, 유형별 사례지를 선정하여 방문자 414명에 대하여 설문조사를 실시하였다. 수목장지 간 만족도 비교를 통해서 국민들이 공설 수목장지를 사설 수목장지에 비하여 선호하고, 그 중에서도 자연산림에 조성된 수목장지를 가장 선호함을 알 수 있었다. 이를 통해 국민들의 선호를 충족하기 위해서는 공설 수목장지 조성을 활성화 하고, 특히 자연산림에 조성하는 수목장지의 확대가 중점적으로 추진되어야 함을 제언하였다. 수목장에 대한 일반적 시민의식에 대한 문항을 통해서는 편의시설에 대한 낮은 만족도, 사설 수목장지의 이용가격과 공신력에 대한 낮은 만족도, 추모목의 안정적 성장환경에 대한 강한 수요가 나타나 이에 대한 개선대책이 필요하였다. 반면 수목장지 이용 경험자의 수목장지 추천의향과 본인이 수목장을 하겠다는 의향이 높게 나타나 수목장 확산의 밝은 전망도 확인하였다. This paper aims to analyze the satisfaction level of people for the different types of tree burial sites, in order to suggest directions for the policy to revitalize tree burials. For the purpose of this study, the tree burial sites were classified into four types, and we conducted a questionnaire, targeting 414 visitors to the different case sites. Through the comparative analysis among the case sites, this study found that people prefer the public tree burial sites to the private ones, and most people liked the type of tree burials best created in a natural forest among the public sites. Based on these results, this study suggested that the policy should be focused on the revitalization of public tree burial sites created especially in natural forests in order to satisfy the preference of people. Through the analysis of the questions regarding the general citizens` awareness of tree burials, this study found the improvement measures are required for convenience facilities, low satisfaction for usage charge, low credibility for private tree burial sites, and strong demands to create stable growth environment for memorial trees. On the other hand, this study confound the bright prospect for the spread of tree burials because people who have used tree burial sites were highly willingness to recommend tree burial sites to others and they wanted to do a tree burial for their own funerals.

        This study intends to analyze the present condition and landscape value of cultural assets relics about the buddhist temple in the northern Chung- Nam province. The result of this study will be as follows: 1. The northern Chung- Nam province has many buddist temples which was flourishing in Backje and Korea dynasty, but these have not been preserved infact because of political and cultural negligence. So, it need to support politically for excavaition and restoration which base on investigation. 2. In the northern Chung - Nam province, a special feature is many buddist ma-ae-bul relics. And it need to improve these circumferences. 3. The excavation value of BOwonsa relics in Seosan as a his torical relic is very high. So, it need to excavate extensively. 4. Cultural assets in the northern Chung-Nam province are appointed. Appointed rate of cultural assets in the northern Chung-Nam province is higher than other provinces. Especially, the appointed rate of as sets relics about the buddhist temple is 40.2% which is higher than any other assets relics. 5. The northern Chung-Nam province has relatively many in Chean-An, Seosan, Dangjin province compared to other provinces. Chung- Nam assets research materials need to ascend for nation or province designating assets.
