        크롬 6가 투여 후 A549 세포에서의 Reactive Oxygen Species와 크롬 5가의 발생

        The production of reactive oxygen species on addition of hexavalent chromium (potassium dichromate, K₂Cr₂O_(7) to lung cells in culture was studied using flow cytometer analysis. A Coulter Epics Profile flow cytometer was used to detect the formation of reactive oxygen species after K₂Cr₂O_(7) was added to A549 cells grown to confluence. The cells were loaded with the dye, 2',7'-dichlorofluorescein diacetate, after which cellular esterases removed the acetate groups and the dye was trapped intracellularly. Reactive oxygen species oxidized the dye, with resultant fluorescence, Increased doses of Cr(Ⅵ) caused increasing fluorescence (10-fold higher than back-ground at 200 μM). Addition of Cr(Ⅲ) compounds, as the picolinate or chloride, caused no increased fluorescence. Electron paramagnetic resonance (EPR) spectroscopic studies indicated that three (as yet unidentified) spectral "signals" of the free radical type were formed on addition of 20, 50, 100 and 200 pM Cr(VI) to the A549 cells in suspension. Two other EPR "signals" with the characteristics of Cr(Ⅴ) entities were seen at field values lower than the standard free radical value.

      • 로버스트 능형회귀 추정법

        회귀분석을 위한 자료에는 회귀이상점이 포함되어 있으며, 동시에 다중공선성이 존재하는 경우가 있다. 이상점들만 포함되어 있는 경우에는 로버스트 추정법을 적용할 수 있으며, 다중공선성만 존재하는 경우에는 능형회귀 추정법을 적용할 수 있다. 그러나 회귀이상점과 다중공선성은 상호 관련을 가지므로 어느 한 문제만을 해결하는 것은 완전한 것일 수가 없다. 따라서 회귀이상점과 다중공선성이 동시에 존재하는 상황에서의 효과적인 추정 방법을 제안하고자 한다. Statisticians have suggested many methods for the problems of multicollinearity and outliers seperately. However, we usually encounter many data sets which have both problems. Thus, we propose a procedure that is called the robust ridge estimation and is very effective to deal with the problems simultaneously. The proposed procedure employs the LMS estimate to determine the bias parameter of ridge regression. In addition, it is verified by simulation studies that the proposed iterative method is more effective, in terms of the stability of estimates and squared distances, than the existing procedure based on the least squares estimates.

        급성 뇌졸중 환자의 응급실도착 소요시간과 대체의료 이용에 관한 연구

        The purpose of this study is to analyse and evaluate the characteristics of stoke patients and the causes of their spending time in arriving emergency room and the actual conditions of using alternative medicines before entering emergency room. The subject of this study four hundred seventy six patients who hospitalized in Western Medical and Chinese Herb Medicine of D Hospital in Pusan within two weeks after attack. The collected data are analysed by the analysis method of narration statistics through SAS program and one-way analysis through ANOVA. The important results of this study are as followings : The spending time in arriving emergency room shows that under 3 hours is 19.5%, under 6 hours is 29.0%, under 24 hours is 55.1%. In the aspects of using alternative medicines before entering emergency room, the patients of 60.1% had used alternative medicines. Through the examination and analysis it can be seen that the geographical-environmental features and the clinical features for the acute stroke patients have more effect than the populational-sociological features on the spending time in arriving emergency room. For the aspects of using alternative medicines before entering emergency room, however, the charactericstics of clinical features have more effect than the populational-sociological features and the geographical-environmental features.

      • Pseudomonas putida 378이 생산하는 Lipoprotein lipase의 성질

        Pseudomonas putida 378이 생산하는 LPL을 정제하여 이에 대한 몇 가지의 생물화학적 성질을 검토하여 다음과 같은 결과를 얻었다. 1. 배양액으로부터 ethanol 분획, Ammonium Sulfate 침전, DEAE-Cellulose column chromatography, CM-Sephadex column chromatography, Gel filtration를 거쳐 LPL을 정제하였다. 이때 효소는 122배 정제되었고, 회수율은 11.77%였다. 2. 정제된 효소는 SDS-PAGE에서 단일 band를 보여주어, 순수하게 정제되었음을 나타내었고, 분자량은 Gel filtration법에 의해 약 108,000으로 나타났고, SDS-PAGE에 의하여 약 107,000으로 나타났으므로, 본 효소액은 monomer로 생각된다. 3. 효소의 최적 pH는 7.0이며, 최적온도는 45℃이었으며, pH는 5.0~8.0 범위에서 안정하였고, 온도는 60℃까지 안정하였다. 4. 본 효소는 1mM Ca 에 의해 활성화 되며, 10mM Hg , Ni 에 의해 저해되었다. 5. 본 효소의 각종 지질에 대한 가수 분해율은 대조구인 olive oil에 비해 lecithin에 대해 다소 높았으며, stearic aid와 tributyrin에 대해서는 매우 낮았다. The purification and properties of the lipoprotein lipase from Pseudomonas putida 378 were investigated. It was purified by ethanol, ammonium sulfate fractionation and chromatographies on DEAE- Cellulose, CM-Sephadex and gel filtration of Sephadex G-100. The enzyme was purified about 122 fold with a yield of 11.77%. The purified enzyme showed a single protein band on SDS-PAGE. Its molecular weight was estimated to be about 108,000 and 107,000 by gel filtration and SDS-PAGE, respectively. Lipoprotein lipase had an optimum pH of 7.0, and an optimal temperature of 45℃, and the stable range of pH was 5.0 to 8.0. The enzyme was thermostable at below 60℃. The enzyme was strongly inhibited by Hg (10mM), Ni (10mM) and showed the most activity ohn Ca (1mM). This lipoprotein lipase was capable of hydrolyzing a variety of oils, showing were active on lecithin than on olive oil.

        Dynamics of phytoplankton in the lower Nakdnng river was investigated from March to December, 1993. The results obtained on this study are as follows. The changing pattern of standing crop of phytoplankton in water temperature and precipitation. It was found that when water temperature was high and the weather was droughty the standing crop of upper region (Hanam, Samrangjin) were higher than lower region (Mulgum, Kupo), and when water temperature was low and the weather was droughty the results were opposite. The phytoplankton distribution patterns of the lower Nakdong River have a close relationship with the patterns of middle stream.
