Low back pain (LBP) is a major musculoskeletal disorder and the socioeconomic problem with a high prevalence that mainly involves intervertebral disc (IVD) degeneration, characterized by progressive nucleus pulposus (NP) cell death and the development of an inflammatory microenvironment in NP tissue. Excessively accumulated cytosolic DNA acts as a damage-associated molecular pattern (DAMP) that is monitored by the cGAS-STING axis to trigger the immune response in many degenerative diseases. NLRP3 inflammasome-dependent pyroptosis is a type of inflammatory programmed death that promotes a chronic inflammatory response and tissue degeneration. However, the relationship between the cGAS-STING axis and NLRP3 inflammasome-induced pyroptosis in the pathogenesis of IVD degeneration remains unclear. Here, we used magnetic resonance imaging (MRI) and histopathology to demonstrate that cGAS, STING, and NLRP3 are associated with the degree of IVD degeneration. Oxidative stress induced cGAS-STING axis activation and NLRP3 inflammasome-mediated pyroptosis in a STING-dependent manner in human NP cells. Interestingly, the canonical morphological and functional characteristics of mitochondrial permeability transition pore (mPTP) opening with the cytosolic escape of mitochondrial DNA (mtDNA) were observed in human NP cells under oxidative stress. Furthermore, the administration of a specific pharmacological inhibitor of mPTP and self-mtDNA cytosolic leakage effectively reduced NLRP3 inflammasome-mediated pyroptotic NP cell death and microenvironmental inflammation in vitro and degenerative progression in a rat disc needle puncture model. Collectively, these data highlight the critical roles of the cGAS-STING-NLRP3 axis and pyroptosis in the progression of IVD degeneration and provide promising therapeutic approaches for discogenic LBP.

The objectives of this study were to identify polymorphisms of insulin-like growth factor I (IGF-I) gene and to investigate their association with growth traits in Nanjiang Huang goats. Five hundred and ninety-two animals were used to detect the polymorphisms in the complete coding sequence, part of introns and the 5'-regulatory region of the IGF-I gene by means of PCR-SSCP. A new single nucleotide polymorphism (G to C transversion) was identified at intron 4 of the IGF-I gene in the goats. Two alleles and three genotypes were observed in this group. The frequency of G and C alleles was 54.6 and 45.4%, respectively. The statistical analysis showed that polymorphism of the IGF-I gene had a significant association (p<0.05) with birth weight (BW), body weight at 6 months (W6) and at 12 months (W12), heart girth at 2 months (G2), body length at 6 months (L6), wither height at 6 months (H6) and at 12 months (H12) and heart girth at 12 months (G12). The goats with genotype CC had significantly higher BW, W6, W12, G2, L6, H6, H12 and G12 than those with genotype GC and had significantly higher W12, H6, H12 and G12 than those with genotype GG. Therefore, genotype CC may be the most advantageous for growth traits in the Nanjiang Huang goat. However, no significant association between SNP genotypes and other growth traits was observed. These results indicated that the SNP marker of the IGF-I gene may be a potential molecular marker for growth traits in Nanjiang Huang goats.
A Systematic Treat Model for Software-Defined Networking

Software-Defined Networking (SDN) has three key features: separation of control and forwarding, centralized control, and network programmability. While improving network management flexibility, SDN has many security issues. This paper systemizes the security threats of SDN using spoofing, tampering, repudiation, information disclosure, denial of service, and elevation of privilege (STRIDE) model to understand the current security status of SDN. First, we introduce the network architecture and data flow of SDN. Second, we analyze security threats of the six types given in the STRIDE model, aiming to reveal the vulnerability mechanisms and assess the attack surface. Then, we briefly describe the corresponding defense technologies. Finally, we summarize the work of this paper and discuss the trends of SDN security research.

In order to solve the problem that the measured axis trace is polluted by noise and can not reflect the real fault type of the turbine generator rotor, the adaptive local iterative filtering is applied to the purification of the axis trace of the generator rotor. As an adaptive mode decomposition method, adaptive local iterative filtering can adaptively decompose a complex original signal into a series of single components reflecting the wave nature of the signal, then, by calculating the correlation coefficient between the autocorrelation function of each decomposition component and the autocorrelation function of the original signal, the real intrinsic mode function is extracted, finally, the extracted intrinsic mode function is used to synthesize the signal to purify the axis trace. The simulation analysis and the purification results of the measured axis trace show that the adaptive local iterative filter can effectively avoid the false components in the decomposition process and is more favorable for extracting the signal features by introducing the Fokker-Planck equation to design the filter, compared with ensemble empirical mode decomposition, this method has better purification performance.

Edge Effect Investigation of DP980 Steel Sheet in Multiple Laser Scanning Process

Due to free spring back, laser forming is an effective non-contact forming technique for hard forming materials. A desired shape is obtained by residual plastic strain which is induced by thermal stresses depending on multiple laser scanning. In this study, the combination of experimental and numerical analysis is employed to investigate the temperature distribution and bending angle of the dual phase (DP980) high strength steel sheet. The edge effect of six cases of laser scanning process involving 1, 2, 3, 4, 5 and 10-time laser scanning are investigated by analyzing their bending angle relative variations and transverse plastic strains. It is found that the edge effect at the middle part of laser scanning line reduces with the increasing number of laser scanning cycles. It is attributed to the fact that the relative variation of the residual plastic strain reduces with the increase of laser scanning cycles. To efficiently reduce the edge effect of DP980 steel sheet, a locally repeated scanning strategy is proposed. The numerical simulation results indicate that the edge effect reduces significantly in the 10-time laser scanning process and the average bending angle simultaneously increases around 20% under the new laser scanning strategy.

Under proper conditions, we prove the existence and multiplicity of the nontrivial solutions to the perturbed system.

In recent years, the deoxycytidine analogue gemcitabine (2′,2′,-difluorodeoxycytidine) has become the first-line chemotherapeutic agent for patients with pancreatic cancer. However, due to the intrinsic resistance of pancreatic cancer cells, gemcitabine-based chemotherapy yields limited disease control, with >85% disease progression at 6 months from diagnosis. Therefore, elucidating the mechanisms of chemoresistance is a critical step in improving cancer therapy, especially for the treatment of pancreatic cancer. We show PROM2, a transmembrane glycoprotein, is ubiquitously upregulated in pancreatic cancer cell. We also found higher PROM2 expression is associated with shortened overall and disease-free survival times in patients diagnosed with pancreatic cancer. We provide evidence that PROM2 promotes chemoresistance to gemcitabine both in vivo and in vitro. Mechanistically, we demonstrate that PROM2 could directly interacted with Akt and activates the Akt signaling pathway, which thus inhibiting gemcitabineinduced apoptosis. As further evidence, we show PROM2 expression and Akt phosphorylation both promote gemcitabine chemoresistance, and cause poorer survival in clinical samples with pancreatic cancer. Combining gemcitabine with the Akt inhibitor MK-2206 facilitated significant tumor shrinkage and dramatically elevated the survival status in mice xenografted with pancreatic cancer cells. Our findings not only establish PROM2 as a novel positive regulator of the Akt signaling pathway and a candidate prognostic indicator of gemcitabine response, but also provide a neo-therapeutic approach for patients resistant to gemcitabine treatment.
