Objective: The aim of this study was to reveal the role and regulatory mechanism of miR-188-5p in the proliferation and differentiation of goat muscle satellite cells. Methods: Goat skeletal muscle satellite cells isolated in the pre-laboratory were used as the test material. First, the expression of miR-188-5p in goat muscle tissues at different developmental stages was detected by quantitative reverse transcription polymerase chain reaction (qRT-PCR). In addition, miR-188-5p was transfected into goat skeletal muscle satellite cells by constructing mimics and inhibitors of miR-188-5p, respectively. The changes of differentiation marker gene expression were detected by qPCR method. Results: It was highly expressed in adult goat latissimus dorsi and leg muscles, goat fetal skeletal muscle, and at the differentiation stage of muscle satellite cells. Overexpression and interference of miR-188-5p showed that miR-188-5p inhibited the proliferation and promoted the differentiation of goat muscle satellite cells. Target gene prediction and dual luciferase assays showed that miR-188-5p could target the 3'untranslated region of the calcium/calmodulin dependent protein kinase II beta (CAMK2B) gene and inhibit luciferase activity. Further functional studies revealed that CAMK2B promoted the proliferation and inhibited the differentiation of goat muscle satellite cells, whereas si-CAMK2B restored the function of miR-188-5p inhibitor. Conclusion: These results suggest that miR-188-5p inhibits the proliferation and promotes the differentiation of goat muscle satellite cells by targeting CAMK2B. This study will provide a theoretical reference for future studies on the molecular mechanisms of skeletal muscle development in goats.

Background: The effect and possible mechanism of traditional Chinese medicine, baicalin, on the PI3K/Akt signaling pathway in drug-resistant human myeloid leukemia HL-60/ADR cells have been investigated in this current study. Methods: HL-60/ADR cells were treated by 20, 40, $80\;{\mu}mol/L$ baicalin followed by cell cycle analysis at 24h. The mRNA expression level of the apoptosis related gene, Bcl-2 and bad, were measured by RT-PCR on cells treated with $80\;{\mu}mol/L$ baicalin at 12, 24 and 48hr. Western blot was performed to detect the changes in the expression of the proteins related to HL-60/ADR cell apoptosis and the signaling pathway before and after baicalin treatment, including Bcl-2, PARP, Bad, Caspase 3, Akt, p-Akt, NF-${\kappa}B$, p-NF-${\kappa}B$, mTOR and p-mTOR. Results: Sub-G1 peak of HL-60/ADR cells appeared 24 h after $20\;{\mu}mol/L$ baicalin treatment, and the ratio increased as baicalin concentration increased. Cell cycle analysis showed 44.9% G0/G1 phase cells 24 h after baicalin treatment compared to 39.6% in the control group. Cells treated with $80\;{\mu}mol/L$ baicalin displayed a trend in decreasing of Bcl-2 mRNA expression over time. Expression level of the Bcl-2 and PARP proteins decreased significantly while that of the PARP, Caspase-3, and Bad proteins gradually increased. No significant difference in Akt expression was observed between treated and the control groups. However, the expression levels of p-Akt, NF-${\kappa}B$, p-NF-${\kappa}B$, mTOR and p-mTOR decreased significantly in a time-dependent manner. Conclusions: We conclude that baicalin may induce HL-60/ADR cell apoptosis through the PI3K/AKT signaling pathway.
Gene Expression Profiles of HeLa Cells Impacted by Hepatitis C Virus Non-structural Protein NS4B

By a cDNA array representing 2308 signal transduction related genes, we studied the expression profiles of HeLa cells stably transfected by Hepatitis C virus nonstructural protein 4B (HCV-NS4B). The alterations of the expression of four genes were confirmed by real-time quantitative RT-PCR; and the aldo-keto reductase family 1, member C1 (AKR1C1) enzyme activity was detected in HCV-NS4B transiently transfected HeLa cells and Huh-7, a human hepatoma cell line. Of the 2,308 genes we examined, 34 were up-regulated and 56 were down-regulated. These 90 genes involved oncogenes, tumor suppressors, cell receptors, complements, adhesions, transcription and translation, cytoskeletion and cellular stress. The expression profiling suggested that multiple regulatory pathways were affected by HCV-NS4B directly or indirectly. And since these genes are related to carcinogenesis, host defense system and cell homeostatic mechanism, we can conclude that HCV-NS4B could play some important roles in the pathogenesis mechanism of HCV.

A series of Fe2O3 and TiO2 co-doped graphene aerogel (Fe2O3–TiO2–GA) nanocomposites with a unique 3D architecture are assembled by a one-pot hydrothermal method combined with freeze-drying. The one with 30 wt% Fe2O3 exhibits the highest adsorptivity (95.0%) and the another with 25 wt% Fe2O3 displays the highest total efficiency (97.7%) including adsorption and visible light-driven photocatalytic degradation for rhodamine B dyes (20.0 mg L−1, 25 mL) within 1 h over 5.0 mg of the nanocomposites. The materials also display satisfactory cyclic stability and the removal efficiency can be 81.8% even after four runs. Additionally, a related photocatalytic mechanism is proposed.

Existing literature has pointed to the dilemmas encountered by many women in the current Chinese marriage market, which mainly result from the conflict between the elevated status of women and the engrained patriarchal gender ideology. However, less is known about women’s mate selection concerns which have been shaped by this context. This article contributes to the field in two aspects. First, it demystifies gan jue (feelings), which is a central discourse in contemporary Chinese young people’s mate choice criteria and, yet, a discourse that has received inadequate attention in existing literature on courtship and marriage in current Chinese society. Second, from the perspective of gender reflexivity, the present study elaborates on how the emphasis on gan jue in young women’s mate selection concerns questions gender normativity and indicates the emergence of a new generational pattern of intimacy in China.

<P>IMPORTANCE Saccharomyces boulardii is a probiotic yeast strain capable of preventing and treating diarrheal diseases. However, the genetics and metabolism of this yeast are largely unexplored. In particular, molecular mechanisms underlying the inefficient galactose metabolism of S. boulardii remain unknown. Our study reports that a point mutation in PGM2, which codes for phosphoglucomutase, is responsible for inferior galactose utilization by S. boulardii. After correction of the mutated PGM2 via genome editing, the resulting strain was able to use galactose faster than a parental strain. While the PGM2 mutation made the yeast use galactose slowly, investigation of the genomic sequencing data of other S. boulardii strains revealed that the PGM2 mutation is evolutionarily conserved. Interestingly, the PGM2 mutation was beneficial for growth at a higher temperature on glucose. We speculate that the PGM2 mutation was enriched due to selection of S. boulardii in the natural habitat (sugar-rich fruits in tropical areas).</P>

Biochar-based composites have been intensively applied in wastewater treatment. To boost theadsorption-photocatalytic capacity of biochar for dyes elimination, S-doped biochar was commodifiedby NiS and MgO (NiS-Mg/S-BC) via the one-pot sulfurization at high temperature. The functional groupsin NiS-Mg/S-BC provided the abundant vacant sites for RhB adsorption and photocatalysis, and the junctioninterface among NiS, MgO, and S-doped biochar was likely to form the channels for the separationand transfer of photo-induced e-/h+ pairs. NiS-Mg/S-BC presented the better adsorption-photocatalyticcapacity of RhB than S-doped biochar, NiS and MgO modified biochar. Langmuir and second-order modelswell described the adsorption behaviors of NiS-Mg/S-BC. The removal efficiency of optimal 2b-NiS-Mg/SBCfor 400 mg/L RhB solution was 96.98% within 120 min, and its adsorption capacity based on secondordermodel was 802.49 mg g1. The photo-corrosion induced to the declined removal efficiency of 2b-NiS-Mg/S-BC to 94.13% after five cycles.
The Analysis on the Development of South Korean Cruise Tourism of Chinese Tourist Market

As the World Tourism Organization points out that cruise tourism is the best tour product for people to enjoy the diverse experience within the shortest terms. With the constant development of cruise tourism through the years, it has gained the attention of the South Korea. This study analyzes the general information of the cruise ports in South Korea, Chinese cruise tourists to South Korea. To solve problems in the development of cruise tourism of South Korea, this study brings forward to the countermeasures including the developing specialized cruise ports, increasing variety of tour products and strengthening train programs.
