A Study on changes in ingredients and activity according to the manufacturing methods of Hwangryunhaedok-tang formulation Choi Jaeeon Advisor: Prof. Lee, Dong-Sung Dept. of Food and Drug Graduate School of Chosun University Objective : To conduct a study on the change in the content and activity of herbal medicine according to the change in the preparation method, Hwangryun-haedoktang was used. The index components of yellow lotus, gold, and gradenia contained in the Hwangryun Haedoktang were selected and analyzed using HPLC, and based on this, the change in the content between the individual and mixed hot water solutions was compared. Methods : The content of Baicalin, Berberine, and Geniposide in Hwangryunhaedoktang was quantified using HPLC with an LC-40 system (Shimadzu, Kyoto, Japan). Separation was achieved using a Capcell Pak C18 column (150 × 4.6 mm, 5 µm, 100 Å, Osaka Soda Co., Ltd., Osaka, Japan) and a YMC Pack ODS column (250 × 4.6 mm I.D., 5 µm, 12 nm, YMC Korea Co., Ltd., Seongnam, Korea). Detection was performed at wavelengths of 250 nm and 254 nm, with the column maintained at 35°C. The mobile phase was composed of 0.01% phosphoric acid (PA) in water (A) and acetonitrile (B). The analysis was performed with a flow rate of 1.0 mL/min using a gradient elution method. [iii] Results : Comparison of the contents of the excipients in Hwangryun-haedoktang and the excipients in the extracted showed that Berberine, Baicalin, Geniposide had a higher individual excipients mixtures than content in the extracted with all yewllo lotus, amur cork, gold, gardenia. Conclusion : The comparison of the ingredients between the mixture and the hot water extract after individual extraction showed no significant differences. It is more efficient to produce herbal medicines based on the specific needs and circumstances of pharmaceutical companies.
