The purpose of this study was to observe the histologic alteration of Langerhans cells in the lining epithelium of apical periodontal cysts and any statistical relationships between the status of inflammatory cell infiltration, the sex, the sites of lesion, and the ages.
For this study 39 biopsy specimens referred to the Department of Oral Pathology, Infirmary School of Dentistry, Chosun University under the diagnosis of apical periodontal cysts were used.
Taken specimens were embedded in paraffin sectioned 6-8u in thickness, and the Langerhan cells were detected using anti S-100 protein antibody(AHP 385 Bioproducts Co), and histochemically processed with Avidin Biotin Complex(ABC AP for rabbit IgG, AK 5010, Vector Lab) method.
The Langerhans cells were counted in each section at 400 magnification field and standard deviation, probability test between each group were evaluated using statistical analysis system(S.A.S.) program.
For the electron microscopic study, taken tissues measured in 1㎣, fixed in Karnovsky solution(pH, 7.4, 0.1M cacodylate buffer, 4℃) for 24 hours and in osmium solution(pH 7.4, 0.lM, cacodylate buffer)for 2 hours, washed with cacodylate buffer, dehydrated with ethyle alcohol solution, immersed with propylene oxide. And embedded in epon mixture according to Luft's method.
Semithin specimens, 1 um in thickness were stained with 1% toluidine blue stain, examined under light microscope. Ultrathin specimens were double stained with uranyle acetate and lead citrate stain method.
And examined with transmitted electron microscope(J.E.M 100 CX1l type 80KV).
Following results were obtained;
1. The prevalence rate of apical periodontal cysts was the highest at the upper anterior, and upper premolar,. lower molar, upper molar, and lower anterior region were followed(P<0.0l).
2. Langerhans cells in lining epithelium of apical periodontal cyst on inflammatory cell infiltration group were decreased compare to that on the control group but the intensity of inflammation does not influnced to the distribution of Langerhans cells on each inflammatory cell grades (P<0.05).
3. Langerhans cells were higher in female group than in male, but the number was decreased in both group compare to that in control gorup(P<0.05).
4. In electron micorscopic study, it is noted that Langerhans cells had clear cytoplasm, with lobulated, indentated nucleus and rod, curved or tennis racket shaped Birbeck granules.
