      • Meniscus-on-Demand Parallel 3D Nanoprinting

        <P>Exploiting a femtoliter liquid meniscus formed on a nanopipet is a powerful approach to spatially control mass transfer or chemical reaction at the nanoscale. However, the insufficient reliability of techniques for the meniscus formation still restricts its practical use. We report on a noncontact, programmable method to produce a femtoliter liquid meniscus that is utilized for parallel three-dimensional (3D) nanoprinting. The method based on electrohydrodynamic dispensing enables one to create an ink meniscus at a pipet-substrate gap without physical contact and positional feedback. By guiding the meniscus under rapid evaporation of solvent in air, we successfully fabricate freestanding polymer 3D nanostructures. After a quantitative characterization of the experimental conditions, we show that we can use a multibarrel pipet to achieve parallel fabrication process of clustered nanowires with precise placement. We expect this technique to advance productivity in nanoscale 3D printing.</P> [FIG OMISSION]</BR>

        <P>Light-powered wireless manipulation of small objects in fluids has been of interest for biomedical and environmental applications. Although many techniques employing UV-vis-NIR light sources have been devised, new methods that hold greater penetrating power deep into medium are still in demand. Here, we develop a method to exploit X-rays to propel half-metal-coated Janus microparticles in aqueous solution. The Janus particles are simultaneously propelled and visualized in real-time by using a full-field transmission X-ray microscope. Our real-time observation discovers that the propulsive motion follows the bubble growth enhanced by water radiolysis near the particle surface under X-ray irradiation. We also show that the propulsion speed is remotely controlled by varying the radiation dose. We expect this work to open opportunities to employ light-powered micro/nanomotors in opaque environments, potentially by combining with medical imaging or nondestructive testing.</P> [FIG OMISSION]</BR>

        Volatile Chemical and Carotenoid Profiles in Watermelons [Citrullus vulgaris (Thunb.) Schrad (Cucurbitaceae)] with Different Flesh Colors

        Twelve watermelon [Citrullus vulgaris (Thunb.)Schrad (Cucurbitaceae)] cultivars with different flesh colors were analyzed by HPLC, GC-FID, and GC-MS for their differences in carotenoid, soluble sugar, organic acid,and flavor. Results showed that all-trans violaxanthin, 9-cis-violaxanthin, and luteoxanthin were the main carotenoid esters in watermelons with yellow flesh. However,watermelons with red flesh were rich in all-trans lycopene and their cis-isomers. High concentrations of β-carotene and pro-lycopenes were found in watermelon with orangeyellow flesh. Large variations in the sucrose concentration were observed among the different watermelons. Sucrose and/or fructose were the dominant sugars, while citric acid and malic acid were the main organic acids in watermelon flesh. Limonene was detected in the watermelon flesh of all investigated genotypes. Interestingly, partial correlation analysis of the chemical concentrations revealed 2 significant (p<0.01) positive correlations between β-ionone and β-carotene, and between (E)-geranyl acetone and prolycopenes

        Favorable Immunotherapy Plus Tyrosine Kinase Inhibition Outcome of Renal Cell Carcinoma Patients with Low CDK5 Expression

        Purpose Immunotherapy (IO) plus tyrosine kinase inhibitor (TKI) has become the first-line treatment for advanced renal cell carcinoma, despite the lack of prognostic biomarkers. Cyclin-dependent kinase 5 (CDK5) affects the tumor microenvironment, which may influence the efficacy of TKI+IO. Materials and Methods Two cohorts from our center (Zhongshan Metastatic Renal Cell Carcinoma [ZS-MRCC] cohort, Zhongshan High-risk Localized Renal Cell Carcinoma [ZS-HRRCC] cohort) and one cohort from a clinical trial (JAVELIN-101) were enrolled. The expression of CDK5 of each sample was determined by RNA sequencing. Immune infiltration and T cell function were evaluated by flow cytometry and immunohistochemistry. Response and progression-free survival (PFS) were set as primary endpoints. Results Patients of low CDK5 expression showed higher objective response rate (60.0% vs. 23.3%) and longer PFS in both cohorts (ZS-MRCC cohort, p=0.014; JAVELIN-101 cohort, p=0.040). CDK5 expression was enhanced in non-responders (p < 0.05). In the ZS-HRRCC cohort, CDK5 was associated with decreased tumor-infiltrating CD8+ T cells, which was proved by immunohistochemistry (p < 0.05) and flow cytometry (Spearman’s ρ=–0.49, p < 0.001). In the high CDK5 subgroup, CD8+ T cells revealed a dysfunction phenotype with decreased granzyme B, and more regulatory T cells were identified. A predictive score was further constructed by random forest, involving CDK5 and T cell exhaustion features. The RFscore was also validated in both cohorts. By utilizing the model, more patients might be distinguished from the overall cohort. Additionally, only in the low RFscore did TKI+IO outperform TKI monotherapy. Conclusion High-CDK5 expression was associated with immunosuppression and TKI+IO resistance. RFscore based on CDK5 may be utilized as a biomarker to determine the optimal treatment strategy.
