Prospect of plant molecular cytogenetics in the 21st century

The genomes of Arabidopsis and rice have been fully sequenced. Genomic sequencing provides global information about genome structure and organization. A comprehensive research account of our recent studies conducted on genome painting, comparative genomics and genome fusion is provided in order to project the prospects of plant cytogenetic research in post-genomics era. Genome analysis by GISH using genome painting is demonstrated as an excellent means suitable for visualization of a whole genome, since total genomic DNA representing the overall molecular composition of the genome is used as a probe. FISH on extended DNA fibers has been developed for high-resolution FISH and has contributed to determining the copy number and order of genes. We have also mapped a number of genes involving starch synthesis on wheat chromosomes by FISH and compared the position of these genes on linkage map of rice. Macro synteny between wheat and rice can be observed by comparing the location of these genes in spite of the fact that the size of DNA per chromosome differs by 20 fold in two. Moreover, to approach our goal towards making bread and udon noodles from rice flour in future by incorporating bread making and the noodle qualities in rice, we have been successful in introducing large genomic DNA fragments containing agronomically important genes of wheat into a rice by successive introduction of large insert BAC clones, there by expanding genetic variability in rice. We call this method genome fusion.

A physical map of the 5S and 18S-26S rRNA genes was determined using bi-color fluorescence in situ hybridization technique in A. victorialis var. platyphyllum. 5S rRNA genes were positioned in the intercalary regions of the short arms in homologous chromosomes 6. Two major loci of the 18S-26S rRNA genes were detected in the secondary constrictions flanking with a pair of satellite and terminal region of short arm in chromosome 4. And two additional minor loci were heterotype, representing one signal on the terminal region of the short arm in one homolog of chromsome 2, and other on one homolog of chromosome 6 with linked 5S rRNA loci. In addition chromomycin A₃ (CMA₃) fluorescent banding method was used to identify the relation between Nucleolus Organizer Region (NOR) sites and CMA, positive heterochromatin sites. In homologous chromosome 4 showing 18S-26S rDNA hybridization signals revealed also distinct CMA₃ positive band.

Development of Simulator of Allophone of Motors for Automobiles

Recently reduction of allophone of motors for automobiles has received a great deal of attention. For this background, the final goal of our research is to develop a simulator for transfer characteristics of allophone of motors in automobiles. In this paper at the preparation phase for the development of a simulator, we investigated the applicability of the transfer function synthesis method (TFSM) experimentally. Additionally, we examined some problems to be solved for practical use of the TFSM by using the finite element method (FEM). As a result, it was found that the derivation of the transfer functions H22 and H33 at the connecting point was difficult and that the modeling accuracy of H22 and H33 had a large influence on the modeling accuracy of the total transfer function H41_sim. Moreover, the method to improve the modeling accuracy was proposed on the basis of the results by FEM analysis. It was confirmed that we could apply the TFSM to the simple test body with the contact area by using the proposed method and the adjustment for damping coefficient of the transfer function obtained by the curve fitting technique (CFT).
