        Impaired Gallbladder Motility and Increased Gallbladder Wall Thickness in Patients with Nonalcoholic Fatty Liver Disease

        Background/Aims Nonalcoholic fatty liver disease (NAFLD) is currently the most common chronic liver disease worldwide. Along with the increase in the incidence of NAFLD and associated obesity, an increase in gallbladder disease (GD) has been noted. This has led to the identification of a new disease entity called fatty GD. There is a gap in the literature on the dynamics of gallbladder function in patients with NAFLD. Methods An observational case-control study, a total of 50 patients with biopsy proven NAFLD without gallbladder stone/sludge and 38 healthy comparison subjects were enrolled. Fasting, postprandial gallbladder volumes (PGV), gallbladder ejection fraction (GEF), and fasting gallbladder wall thickness (FGWT) were measured by real-time 2-dimensional ultrasonography. Results Fasting gallbladder wall thickness, fasting gallbladder volumes and PGV were significantly higher in patients with NAFLD than control subjects (P < 0.001, P = 0.006, and P < 0.001, respectively). Gallbladder ejection fraction was significantly lower in the NAFLD group than the controls (P = 0.008). The presence of NAFLD was an independent predictor for GEF, PGV, and FGWT. Also, steatosis grade was an independent predictor for GEF, and GEF was significantly lower in the nonalcoholic steatohepatitis (NASH) subgroup than the controls. Conclusions Gallbladder dysfunction and increase in gallbladder wall thickness exists in asymptomatic (without stone/sludge and related symptoms) patients with NAFLD and are useful in identifying fatty GD. Measurement of these variables in NAFLD patients may be useful in identifying those at higher risk for GD. (J Neurogastroenterol Motil 2016;22:470-476)

      • Cancer Preventive Effects of Whole Cell Type Immunization against Mice Ehrlich Tumors

        Background: Effects of whole cell type immunization on mice Ehrlich tumours were evaluated. Materials and Methods: After preliminary study, mice were divided two major groups; $1{\times}1000$ and $100{\times}1000$ live Ehrlich cell transferred major groups, each divided into four subgroups (n: 10). Study groups were immunized with Ehrlich cell lysates in 0, 3, 7, $14^{th}$ days and after 30 days of last immunization, live Ehrlich cells were transferred. Mice were observed for six months and evaluated for total and cancer free days. Results: Out of $100{\times}1000$ cell transferred solid type study group, all study group mean and tumour free periods were statistically longer than control groups. All $1{\times}1000$ Ehrlich cell transferred study groups survived significantly longer than $100{\times}1000$ Ehrlich cell transferred groups. Conclusions: Ehrlich mice tumours were prevented and survival prolonged with whole cell type immunization. Effects are related to the number of transferred tumor cells.

        Genetic relationships and diversity within cultivated accessions of Salvia officinalis L. in Turkey

        Sage (Salvia officinalis L.) is not found in Turkey's natural flora; however, its cultivation has been increasing in recent years. The species is widely cultivated in many provinces of the Aegean and the Mediterranean regions. There has been no informa- tion regarding genetic characteristics and the intra-individual phylogeny of cultivated sage. The levels of genetic relatedness of 19 Salvia samples from Izmir were determined by molecular tools. The sequences from the internal transcriber spacer (ITS) region of 18S–28S nuclear ribosomal DNA (nrDNA) locus were amplified by PCR and sequenced. Approximately, a 710 bp single amplified product was obtained in all genotypes. Of the nineteen Salvia specimens, six were identified as Salvia x sylvestris L. and the remaining 13 specimens were S. officinalis. The sequences of both species shared no similar secondary structures. The ITS region of all specimens contains several indels and substitutions. In the phylogenetic dendrogram, three major clusters (I, II, and III) and two independent branches (IV and V) were observed. Ninety-seven percent of local Salvia specimens were clustered in two branches (I and II), indicating a high diversity of genetic relatedness. Sequence divergence was higher among the specimens of S. officinalis than the specimens of Salvia x sylvestris.
