<P>Highly effective and minimally toxic antimicrobial agents have been prepared by immobilizing glucose oxidase (GOx) onto biocompatible chitosan nanoparticles (CS-NPs). CS-NPs were prepared via ionotropic gelation and used for the immobilization of GOx via approaches of covalent attachment (CA), enzyme coating (EC), enzyme precipitate coating (EPC), and magnetic nanoparticle-incorporated EPC (Mag-EPC). EPC represents an approach consisting of enzyme covalent attachment, precipitation, and cross-linking, with CA and EC being control samples while Mag-EPC was prepared by mixing magnetic nanoparticles (Mag) with enzymes during the preparation of EPC. The GOx activities of CA, EC, EPC, and Mag-EPC were 8.57, 17.7, 219, and 247 units/mg CS-NPs, respectively, representing 26 and 12 times higher activity of EPC than those of CA and EC, respectively. EPC improved the activity and stability of GOx and led to good dispersion of CS-NPs, while Mag-EPC enabled facile magnetic separation. To demonstrate the expandability of the EPC approach to other enzymes, bovine carbonic anhydrase was also employed to prepare EPC and Mag-EPC samples for their characterizations. In the presence of glucose, EPC of GOx generated H2O2 in situ, which effectively inhibited the proliferation of <I>Staphylococcus aureus</I> in both suspended cultures and biofilms, thereby demonstrating the potential of EPC-GOx as environmentally friendly and highly effective antimicrobial materials.</P> [FIG OMISSION]</BR>

<P>In a previous study, we found that the short isoform of DNAJB6 (DNAJB6(S)) had been decreased in the striatum of a mouse model of Parkinson's disease (PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). DNAJB6, one of the heat shock proteins, has been implicated in the pathogenesis of PD. In this study, we explored the cytoprotective effect of DNAJB6(S) against 1-methyl-4-phenylpyridinium ion- (MPP+-) induced apoptosis and the underlying molecular mechanisms in cultured LN18 cells from astrocytic tumors. We observed that MPP+ significantly reduced the cell viability and induced apoptosis in LN18 glioblastoma cells. DNAJB6(S) protected LN18 cells against MPP+-induced apoptosis not only by suppressing Bax cleavage but also by inhibiting a series of apoptotic events including loss of mitochondrial membrane potential, increase in intracellular reactive oxygen species, and activation of caspase-9. These observations suggest that the cytoprotective effects of DNAJB6(S) may be mediated, at least in part, by the mitochondrial pathway of apoptosis.</P>
Long-term follow-up may be needed for pancreaticobiliary reflux in healthy adults
Sung-Pil Yun,Jee Yeon Lee,Hong Jae Jo,Hyun Sung Kim,Dae Hwan Kim,Jae Hun Kim,Sung Jin Park,Do Yoon Park,Hyung-Il Seo 대한외과학회 2013 Annals of Surgical Treatment and Research(ASRT) Vol.84 No.2
Purpose: The reflux of pancreatic enzymes into the biliary tract is associated with chronic inflammation and increases cellular proliferation of the biliary epithelium, leading to biliary carcinoma. The aim of this study is to detect the incidence of occult pancreaticobiliary reflux (OPBR) in patients who underwent elective cholecystectomy. Methods: Forty-seven patients with symptomatic gallstones who underwent cholecystectomy were recruited for this study. The gallbladder bile samples were obtained from the specimen of gallbladder and the amylase level was measured. The immunohistochemistry of p53, SMAD4 and Ki-67 were performed for the detection of metaplasia and dysplasia. Results: Biliary amylase was higher than the serum amylase in 10 patients (group A, 15,402.66 ± 33,592.43 IU/L; group B, 13.06 ± 18.12 IU/L). The mean age was 67.2 years in group A and 51.2 in group B (P 〈 0.01). The ratio of male to female was 1:2.3 and 1:1.8 in group A and B, respectively (P = 0.297). Eight patients in group A and thirteen patients in group B had inflammation (P = 0.014). The positive results of the Ki-67 test were exhibited in five cases in each group (P = 0.024). Conclusion: Results from the study indicate that the age was older, degree of inflammation and positive rate of Ki-67 were higher when OPBR was suspected. In conclusion, the patients with OPBR would need long-term follow-up, because the OPBR can cause dysplasia and the reflux of pancreatic juice may be considered as a risk factor for extrahepatic bile duct carcinoma.

In this study to investigated the changes of properties of the reactor-materials with respect to content of irradiation by neutrons we prepare samples of the reactor-materials irradiated by the fast neutrons and try industrial utilization of the Mossbauer spectroscopic method, similar attempts of which have been rare, comparing with several other possibilities in the researched about the reactor-materials SA 508 cl.3. In addition, the determination for the annealing temperature of the material which is possible means of the Mossbauer spectroscopy is very important and necessary for inhibition of the rise in remaining stress by neutron irradiated reactors-materials. The data the annealing temperatures obtained by means of the method avail us to trace the behavior of ??? isotope in the samples of the materials.

<P>To fully understand the influence of kinetic inhibitor, poly-N-vinylcaprolactam (PVCap), on methane hydrate formation, gas uptake measurement (macroscopic point of view) and in situ Raman spectroscopic analysis (microscopic point of view) were carried out simultaneously in a semibatch stirred tank reactor at constant temperature (<I>T</I>) and pressure (<I>P</I>). The capturing the behavior of guest molecules (large to small cavity ratio or cage variation) is one of the most important properties of gas hydrate studies. However the real-time properties of the cage variation under constant <I>T</I> and <I>P</I> conditions in an agitation system have not been reported previously. In this study, we measured this property (i.e., the large to small cavity ratio) from the in situ Raman spectra during hydrate formation in an agitation system instead of a static system, which provided valuable information on the time-dependent hydrate kinetic behavior. The study reveals that the presence of PVCap prevents the rate of large cavity encapsulation at an early stage of hydrate formation. The influence of PVCap from microscopic and macroscopic points of view is also presented.</P>
Risk factors for parastomal hernia: based on radiological definition
Sung Yeon Hong,Seung Yeop Oh,Jae Hee Lee,Do Yoon Kim,Kwang Wook Suh 대한외과학회 2013 Annals of Surgical Treatment and Research(ASRT) Vol.84 No.1
Purpose: The aim of this study was to investigate the clinical and radiological incidence of parastomal hernia and to analyze the risk factors for parastomal hernia. Methods: We reviewed retrospectively 108 patients with end colostomy from January 2003 to June 2010. Age, sex, surgical procedure type, body mass index (kg/m2), stoma size, and respiratory comorbidity were documented. Results: There were 61 males (56.5%) and 47 females (43.5%). During an overall median follow-up of 25 months (range, 6 to 73 months), 36 patients (33.3%) developed a radiological parastomal hernia postoperatively and 29 patients (26.9%) presented with a clinical parastomal hernia. In multivariate analysis, gender (odds ratio [OR], 6.087; P = 0.008), age (OR, 1.109; P = 0.009) and aperture size (OR, 6.907; P 〈 0.001) proved to be significant and independent risk factors after logistic regression analysis. Conclusion: This study showed that the incidence of radiological parastomal hernia is higher than clinical parastomal hernia. Risk factors for parastomal hernia proved to be female, age, and aperture size.

In general, the application of HP contributes to improvement of functionalities of meat protein. In the current study, HP allowed the lower thermal processing of restructured meat product. Low thermal processing allowed fresh-like meat color which is one of problems in hot-set restructuring, while they showed slight discolorization induced by HP. In addition, HP processing combined with thermal processing could be achieved the palatable binding strength in restructured meat product. The addition of non-meat protein had an effect in binding strength. However, they showed no effect on water binding properties at 200 MPa, especially in milk proteins such as casein and whey protein. This is probably due to protein aggregation or to increase in surface hydrophobicity under HP. This result indicates that the application of HP on meat restructuring is more significant than the addition of binders. Therefore, the application of HP has apotential benefit in restructured meat product, and further investigations are needed.
