Carob waste is a useful raw material for the second-generation ethanol because 50% of its dry weight is sucrose, glucose, and fructose. To optimize the process, we have studied the influence of the initial concentration of sugars on the fermentation performance of Saccharomyces cerevisiae. With initial sugar concentrations (S<sub>0</sub> ) of 20 g/l, the yeasts were derepressed and the ethanol produced during the exponential phase was consumed in a diauxic phase. The rate of ethanol consumption decreased with increasing S<sub>0</sub> and disappeared at 250 g/l when the Crabtree effect was complete and almost all the sugar consumed was transformed into ethanol with a yield factor of 0.42 g/g. Sucrose hydrolysis was delayed at high S<sub>0</sub> because of glucose repression of invertase synthesis, which was triggered at concentrations above 40 g/l. At S<sub>0</sub> higher than 250 g/l, even when glucose had been exhausted, sucrose was hydrolyzed very slowly, probably due to an inhibition at this low water activity. Although with lower metabolic rates and longer times of fermentation, 250 g/l is considered the optimal initial concentration because it avoids the diauxic consumption of ethanol and maintains enough invertase activity to consume all the sucrose, and also avoids the inhibitions due to lower water activities at higher S<sub>0</sub> .

The use of recycled materials, such as the fne recycled aggregate made from concrete waste and carbon fber (CF) product of industrial waste, for the manufacture of conductive recycled mortars (CRM), transforms the mortar base cement normally made with cement:sand in a sustainable multifunctional material, conferring satisfactory mechanical and electrical properties for non-structural uses. This action provides ecological benefts, reducing the use of natural fne aggregates from rivers and the amount of concrete waste deposited in landflls resulting from construction waste. In this investigation the efect of the addition of CF on electrical properties in hardened, wet and dry state, electric percolation in dry state and fuidity of the wet mixture of a cement based CRM was evaluated: fne recycled aggregate: graphite powder, CRM specimens with dimensions of 4×4×16 cm. were manufactured for 3, 7 and 28 days of age and sand/cement ratios=1.00, graphite/cement=1.00, water/cement=0.60 and CF=0.1, 0.3, 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0% compared to the weight of cement. The results demonstrated the efect of the addition of CF in CRM, reducing fuidity of the mixtures due to the opposition generated by its physical interaction of CF with recycled sand or recycled fne aggregate and graphite powder (GP), in its case, placing the electric percolation percolation at 0.30% and 0.45% of CF for CRM with and without GP, respectively. Increases in electrical conductivity (EC) without the presence of GP are defned by the contact between the CF and the conductive paths formed. In contrast, with the presence of GP, the EC is defned by the contact between the CF and the GP simultaneously, forming conductive routes with greater performance in its EC.
eIF5A-PEAK1 Signaling Regulates YAP1/TAZ Protein Expression and Pancreatic Cancer Cell Growth

<P>The focal adhesion kinase PEAK1 drives aggressive pancreatic cancer cell behaviors by activating a stem cell-like transcription factor program.</P><P>In pancreatic ductal adenocarcinoma (PDAC), mutant KRAS stimulates the translation initiation factor eIF5A and upregulates the focal adhesion kinase PEAK1, which transmits integrin and growth factor signals mediated by the tumor microenvironment. Although eIF5A-PEAK1 signaling contributes to multiple aggressive cancer cell phenotypes, the downstream signaling processes that mediate these responses are uncharacterized. Through proteomics and informatic analyses of PEAK1-depleted PDAC cells, we defined protein translation, cytoskeleton organization, and cell-cycle regulatory pathways as major pathways controlled by PEAK1. Biochemical and functional studies revealed that the transcription factors YAP1 and TAZ are key targets of eIF5A-PEAK1 signaling. YAP1/TAZ coimmunoprecipitated with PEAK1. Interfering with eIF5A-PEAK1 signaling in PDAC cells inhibited YAP/TAZ protein expression, decreasing expression of stem cell–associated transcription factors (STF) including Oct4, Nanog, c-Myc, and TEAD, thereby decreasing three-dimensional (3D) tumor sphere growth. Conversely, amplified eIF5A-PEAK1 signaling increased YAP1/TAZ expression, increasing expression of STF and enhancing 3D tumor sphere growth. Informatic interrogation of mRNA sequence databases revealed upregulation of the eIF5A-PEAK1-YAP1-TEAD signaling module in PDAC patients.
