        Molecular Characterisation of the Mafriwal Dairy Cattle of Malaysia Using Microsatellite Markers

        The Mafriwal dairy cattle was developed to meet the demands of the Malaysian dairy Industry. Although there are reports on its production and reproductive performance, there has been no work on its molecular characterization. This study was conducted to characterize the Mafriwal dairy cattle using microsatellite markers. Fifty two microsatellite loci were analysed for forty Mafriwal dairy cows kept at Institut Haiwan Kluang, Malaysia. The study showed two microsatellite loci to be monomorphic. Allele frequencies for the polymorphic loci ranged from 0.01 to 0.31. Genotype frequencies ranged from 0.03 to 0.33. The mean overall heterozygosity was 0.79. All polymorphic microsatellite loci deviated significantly (p<0.01) from Hardy-Weinberg equilibrium. The Mafriwal dairy cattle showed high genetic variability despite being a nucleus herd and artificial insemination being practiced.

        Reproductive Performance of Three Iranian Sheep Breeds

        To evaluate the ewe reproductive performances of three Iranian local sheep breeds, namely Moghani, Chal and Zel, 27 rams and 473 ewes were used as the foundation flock in a crossbreeding program. The three breeds were crossed amongst themselves reciprocally. Ram breed and ewe breed showed a two-way interaction on apparent fertility. The Zel ewes showed higher fertility than the Chal and Moghani ewes (94.4 vs 77.3 and 84.6%). Prolificacy among main and individual mating groups were not significantly different (p>0.05). Zel ewes were 22.3 and 14.8% more reproductive (p<0.05) than Chal and Moghani ewes, respectively. Zel and Moghani ewes showed higher (p<0.05) ewe productivity than Chal ewes. Ram breed had a significant (p<0.05) effect on ewe efficiency. The Zel rams were not completely successful to mate with Chal ewes, this caused a to way-interaction between ram and ewe breeds. The Zel ewes mated with Chal and Moghani rams showed 26.1 and 28.5% more efficiency than those mated with Zel rams. This observation strongly supports the use of crossbreeding to improve the efficiency of the Zel ewes. With decreasing lamb mortality, through supplemented feeding of lambs in pre-weaning period, and increasing litter weight, through crossbreeding, it should be possible to improve the efficiency of Zel ewes even better than Chal and Moghani ewes. Small body size of Zel ewes that needs the lower maintenance requirements would be a well advantageous to using Zel ewes as the dam herd to produce commercial lambs in north area of Iran.

        RAPD Analysis of Three Deer Species in Malaysia

        The genetic variability within and among three deer species in Malaysia, namely Cervus nippon (sika), Cervus timorensis (rusa) and Cervus unicolor (sambar), were evaluated using the RAPD technique. The DNA extracted from the buffy coat of 34 sika, 38 rusa and 9 sambar were analysed using ten primers that gave bands which showed good resolution. The primers generated 164 RAPD markers in total, and these ranged in size from 150 to 900 bp. The percent of polymorphism of the bands generated per primer ranged from 66.66-93.33% for rusa, 36.84-61.14% for sambar and 52.38-100% for sika. The overall percent polymorphism observed for the 164 RAPD markers was 99.39%. The results revealed five exclusive, monomorphic markers for sambar and one exclusive, monomorphic marker for sika; none was observed for rusa. However, these cannot be declared as markers for the identification of the species without analysis of more samples, populations and species. The means of within population genetic distances, based on Dice's and Jaccard's similarity indices, were similar for the rusa (0.383 and 0.542, respectively) and sika (0.397 and 0.558, respectively) populations with the sambar population being the least variable (0.194 and 0.323, respectively). The Dice based genetic distances within the species ranged from 0.194 to 0.397 and the genetic distances among the species were 0.791-0.911. The genetic distances based on Dice's and Jaccard's similarity indices between the rusa and sambar were 0.556 and 0.713, between the rusa and sika populations were 0.552 and 0.710, and between sambar and sika were 0.622 and 0.766, respectively.

        ASSOCIATION OF RED CELL X-PROTEIN, NADH-DIAPHORASE 1, MALIC ENZYME AND SERUM ARYLESTERASE PHENOTYPES WITH ECONOMIC TRAITS IN SHEEP

        The red cell X-protein, NADH-diaphorase 1, malic enzyme and serum arylesterase phenotypes of 50 Thai Longtail and 53 Cameroon X Thai Longtail ($F_1$) crossbred sheep were determined by horizontal starch gel electrophoresis. None of the economic traits was influenced by DIA1, ME and EsA phenotypes. However, XP phenotypes showed a highly significant association with body weight, body height, heart girth and back girth, with mean values of XP+ve phenotype greater than XP-ve. The $XP^+$ allele was associated with greater body weight, body height, heart girth and back girth.

        BIOCHEMICAL POLYMORPHISM STUDIES IN BREEDS OF WOOL-SHEEP, HAIR-SHEEP AND THEIR HYBRIDS IN MALAYSIA

        A biochemical genetic study on blood enzyme/protein systems in some breeds/crosses of sheep in Malaysia was carried out using horizontal starch gel electrophoresis. Blood samples were collected from 435 sheep, representing 8 breeds/crosses. These included 5 wool sheep breeds (Thai Longtail, wiltshire, Suffolk, Dorsimal and cMBLx), 1 hair sheep breed (Barbados Blackbelly) and 2 hybrids between wool sheep and hair sheep (Cameroon ${\times}$ Thai Longtail and Bali Bali ${\times}$ Malin). Twenty loci systems were examined. Of these, ten ($HB{\beta}$, ALB, TF, XP, CAT, DIA1, EsA, GPI, ME and NP) exhibited genetic variation whereas the other ten (AAT, CA, DIA2, ${\alpha}GLO$, ${\alpha}GLU$, LDH, MDH, PEP[leu-gly-gly], 6PGD and SOD) were monomorphic. The allelic frequencies which were obtained in 10 polymorphic markers are assessed and compared with the results obtained by previous workers. The estimations of inbreeding coefficient, intrabreed variation and breed relationships have been critically discussed and are used to reveal some important recommendations.
