        The Evaluation of Exogenous Melatonin Administration in Supraspinatus Overuse Tendinopathy in an Experimental Rat Model

        Background: Increased oxidative stress and inflammation play a critical role in the etiopathogenesis of chronic tendinopathy. Melatonin is an endogenous molecule that exhibits antioxidant and anti-inflammatory activity. The aim of this study was to evaluate the biochemi-cal and histopathological effects of exogenous melatonin administrations in supraspinatus overuse tendinopathy.Methods: Fifty rats were divided into the following four groups: cage activity, melatonin treatment, corticosteriod therapy, and control. Melatonin (10 mg/kg, intraperitoneal; twice a day) and triamcinolone (0.3 mg/kg, subacromial; weekly) were administered to the treat-ment groups after the overuse period. Biochemical and histopathological evaluations were performed on serum samples and biopsies obtained from rats. Plasma inducible nitric oxide synthase (iNOS), vascular endothelial growth factor (VEGF), total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI) levels were evaluated biochemically. Results: The TAS, TOS, OSI, iNOS, and VEGF values were significantly lower than the pre-treatment levels in rats receiving exogenous melatonin treatment (3 or 6 weeks) (p<0.05). TOS, iNOS, VEGF, and OSI values after 3 weeks of triamcinolone administration, and TOS, VEGF, and OSI levels after 6 weeks of triamcinolone application, were significantly lower than the pre-treatment levels (p<0.05). Conclusions: Exogenous melatonin application in overuse tendinopathy reduces oxidative stress and inflammation. Melatonin might be an alternative potential molecule to corticosteroids in the treatment of chronic tendinopathy.

        Design Principles for Wind Turbine Earthquake and Wind Load Combinations

        With the ongoing changes in the modern world, the usage of renewable energy sources is increasing. The negative impact of fossil fuels on global warming has led to the search for clean energy sources. Wind energy, which is the most widely used of those sources, is discussed in this study. Numerous studies have been undertaken in this regard, but seismic eff ects are only newly being considered. In addition to having strong wind zones, Turkey is located in a geographical position through which active fault lines pass. The presence of high wind zones overlapping with these fault lines necessitates seismic analysis for turbines planned to be built in the country. Analysis of wind and seismic load simulation is diffi cult with traditional structural design programs. In this study, coeffi cients including wind loads are suggested for civil engineers who plan to analyze wind turbines with only seismic eff ects. For this purpose, a horizontal axis steel wind turbine with a 5 MW scale is analyzed in this work considering a series of wind and seismic loads in the area around Gelibolu. These environmental eff ects are evaluated under diff erent operating conditions of the turbine, including normal operation with no earthquake loads, park condition with earthquake loads, idling condition with wind loads, normal operation with earthquake loads, and idling condition with earthquake and wind loads. The full system model of the turbine is developed with the FAST fi nite element program employing a special code for wind turbines developed by the National Renewable Energy Laboratory. As a result of the analysis, it is concluded that seismic loads acting with the wind signifi cantly change the internal forces. Damping eff ects occur when seismic and wind loads act at the same time. For this reason, wind loads need to be reduced by a certain coeffi cient in some cases. Coeffi cients are proposed here for application in initial designs and load combinations for certain wind and earthquake conditions.

        Investigation of antimicrobial activity and entA and entB genes in Enterococcus faecium and Enterococcus faecalis strains isolated from naturally fermented Turkish white cheeses

        In this research, the antagonistic effects of Enterococcus faecalis and E. faecium bacteria isolated from naturally fermented Turkish white cheeses, produced without starter culture, against Listeria monocytogenes, L. innocua, L. ivanovii, Staphylococcus aureus, and E. faecalis were evaluated. The presence of entA and entB genes was also detected in the isolates, which had antimicrobial activity. Total 71 strains of E. faecalis and 7 strains of E. faecium were tested; 20 of E. faecalis and none of E. faecium strains showed antimicrobial activity against the tested bacteria using agar spot method. Among E. faecalis strains, which had antimicrobial activity, three strains contained both entA and entB genes, two strains carried only entA gene, and five strains had only entB gene. These cheese-sourced enterococcal strains or their enterocins should be considered to be used for food preservation especially in the dairy industry.
