        A Study on the Development of Wind and Wave Model of Typhoon

        In this paper, after analyzing other models with their advantages and disadvantages, we proposed a simple parametric model for calculating wind speed & direction and wave height & direction at any location around the typhoon at sea. The proposed wind-field model of typhoon is asymmetric, and consists of a circular symmetric wind-field caused by the pressure gradient of stationary typhoon and a moving wind-field caused by the movement of typhoon. By verifying this model through observed data, we found that it is accurate enough to develop the simulation software for training students and seafarers so as to take appropriate actions while being faced with the typhoon at sea.

        On the Development of Typhoon Avoidance Simulation System with the Evaluating Method by Seakeeping Performance of Ship

        A simulation system is needed to train students and mariners in order that they can take suitable actions to evade typhoon's strike promptly and sufficiently. In order to make such kind of system, three kinds of models about the typhoon are necessary, typhoon prediction model to generate typhoon's track, wind & wave-field model to make sea conditions around the typhoon and evaluation model of trainee's action whether their actions were suitable or not during simulation. We have developed the prediction and wind & wave-field models of typhoon, but the evaluation model has not been developed yet. In this paper, after making a method for evaluating trainee's actions by seakeeping performance, we propose an typhoon avoidance simulation system for training mariners so that they can promote their abilities to evade the typhoons at sea.

        Synthesis and Evaluation of Antitumor Activity

        Fourty eight derivatives of 2-(1-oxyalkyl)-1,4-dioxy-9,10-anthraquinone were synthesized, and their antitumor activity was evaluated. On the whole, 2-(1-hydroxyalkyl)-1,4-dihydroxy-9,10-anthraquinones (DHAQ=1,4-dihydroxy-9,10-anthraquinone) showed stronger cytotoxic activity against L1210 cells than 2-(l-hydroxyalkyl)-1,4-dimethoxy-9,10-anthraquinones(DMAQ =1,4-dimethoxy-9,10-anthraquinone), implying that free hydroxy groups at C-1 and C-4 of the anthraquinone structure are necessary for the cytotoxic activity. The bioactivity of 2-(lhydroxyalkyl)-DHAQ derivatives differed according to the size of alkyl group at C-1;while the elongation of alkyl group over 7 carbon atoms failed to enhance the bioactivity, the derivatives possessing alkyl moiety of 1-6 carbon atoms showed an increase in the cytotoxicity and the antitumor activity in Sarcoma-180; 2-hydroxymethyl-DHAQ ($ED_{50}$, $15\mu\textrm{g}$/ml; T/C, 125%), 2-(1 -hydroxyethyl)-DHAQ($1.9{\mu}g/ml;139.2%)$;, 2-(1-hydroxypropyl)-DHAQ ($7.2{\mu}g$/ml; 135.1%), 2-(1-hydroxybutyl)-DHAQ ($10.2{\mu}g/ml; 125.3%)$, 2-(1-hydroxypentyl)-DHAQ ($23.7{\mu}g/ml; 110.1%$). and 2-(1-hydroxyhexyl)-DHAQ ($58{\mu}g/ml;108%$). Next, 2-(1-Hydroxyalkyl)-DHAQ derivatives were acetylated to produce 2-(1-acetoxyalkyl)-DHAQ analogues. Although the acetylation somewhat enhanced the cytotoxicity, but not the antitumor action. In addition, the presence of phenyl group at $C-1^{l}$ enhanced the cytotoxicity and the T/C value, compared to alkyl groups of same size; 2-(1-hydroxy-1-phenyl)-DHAQ ($ED_{50}$, $5.6{\mu}g$, T/C, 137%).

        A Study on the Development of Typhoon Track Forecast Model Based on the Past Track Data

        This paper is aimed to develop a mathematical model for making the forecast information of typhoon's movement such as the estimated movement direction and positions after 24 and 48 hours. The proposed model calculates such kind of information of a typhoon by similar past typhoon's track data which are selected with three similarity criteria among the database of typhoons' tracks for past fifty years. We carried out a simulation forecast with No.14 typhoon formed in 1997, and found that the results of the proposed model were reasonable and it would be suitable for a simulation system for training mariners so that they can take suitable actions to evade the typhoons.

      • Molecular-like Transformation from PhSe-Protected Au<sub>25</sub> to Au<sub>23</sub> Nanocluster and Its Application

        <P>In this work, we report a new size conversion from [Au-25(SePh)(18)](-) to [Au-23(SePh)(16)](-) nanoclusters under the reductive condition (NaBH4). This novel transformation induced by only reductant has not been reported before in the field of gold nanocluster. The conversion process is studied via MALDI mass spectrometry, and UV-vis spectroscopy. These results demonstrate that the [Au-23(SePh)(16)](-) nanocluster is directly obtained by pulling out two units of 'Au-SeR' from the [Au-25(SePh)(18)](-) nanocluster, which is similar to the 'small molecular' reaction. In order to further understand this novel conversion, DFT calculations were performed, in which, with addition of two H- in the [Au-25(SeH)(18)](-) model, two Au atoms will depart from the structure of the [Au-25(SeH)(18)](-), which is consistent with the experimental results. Furthermore, the as prepared [Au-23(SePh)(16)](-) nanoclusters can be converted into [Au-25(PET)(18)](-) nanocluster (PET = SCH2CH2Ph) with excess PET under the reductive condition, which is quite remarkable due to a stronger bond of Au-Se in comparison to Au-S of the final product. Interestingly, the number of the PET ligands on the surface of the 25-atoms nanocluster can be controlled by the addition of the reductant. Based on these results, a circularly progressive mechanism of ligand exchange is proposed. This may offer a new approach to synthesis of new gold nanoclusters and also have significant contribution for understanding and further exploration of the mechanism of ligand exchange.</P>

      • Suppression of Cellular Apoptosis Susceptibility (CSE1L) Inhibits Proliferation and Induces Apoptosis in Colorectal Cancer Cells

        The cellular apoptosis susceptibility (CSE1L) gene has been demonstrated to regulate multiple cellular mechanisms including the mitotic spindle check point as well as proliferation and apoptosis. However, the importance of CSE1L in human colon cancer is largely unknown. In the present study, we examined expression levels of CSE1L mRNA by semiquantitative RT-PCR. A lentivirus-mediated small interfering RNA (siRNA) was used to knock down CSE1L expression in the human colon cancer cell line RKO. Changes in CSE1L target gene expression were determined by RT-PCR. Cell proliferation was examined by a high content screening assay. In vitro tumorigenesis was measured by colony-formation assay. Cell cycle distribution and apoptosis were detected by flow cytometric analysis. We found CSE1L mRNA to be expressed in human colon cancer cells. Using a lentivirus based RNAi approach, CSE1L expression was significantly inhibited in RKO cells, causing cell cycle arrest in the G2/M and S phases and a delay in cell proliferation, as well as induction of apoptosis and an inhibition of colony growth capacity. Collectively, the results suggest that silencing of CSE1L may be a potential therapeutic approach for colon cancer.

        Cat NPC2, a Newly Identified Allergen, With High Cross-Reactivity to Can f 7

        Purpose: Pet-derived allergens are the common indoor inhalant allergens. Among them, cat and dog allergens constitute more than 80% of animal allergic patients, which greatly affect the quality-of-life of patients and increase the burden of social health care. The aim of this study was to identify Cat-Niemann pick type C2 (NPC2) protein, a homologue of Can f 7, as a new allergen. Methods: Cat-NPC2 complementary DNA (cDNA) was cloned and optimized for amplification and expression in Escherichia coli. Then, recombinant Cat-NPC2 (rCat-NPC2) was purified by Ni2+ affinity chromatography. The allergenicity was assessed by enzyme-linked immunosorbent assay (ELISA), western blot and basophil activation test (BAT). Based on the sequence similarity, the cross-reactivity between Cat-NPC2 and Can f 7 was investigated by inhibition ELISA. Circular dichroism spectroscopy and homology modeling were used to characterize the structure of Cat-NPC2. Results: The cDNA sequence of Cat-NPC2 was cloned with a 450-bp open reading frame coding for 149 amino acids (GenBank MN_737596). The condon-optimized NPC2 gene was subcloned and expressed in E. coli with a molecular weight of 18.9 kDa. The native Cat-NPC2 was detected in cat dander extracts. The allergenicity determined by ELISA, western blot and BAT suggested at least 14.5% cat-allergic patients displayed high specific immunoglobulin E (IgE) recognition of Cat-NPC2. The predicted structure of Cat-NPC2 was found to consist of 7 β-strands arranged in 2 β-sheets. An ELISA based assay showed that rCat-NPC2 bound to cholesterol in a dose dependent manner. Based on the structure and sequence similarities, IgE cross-reactivity was demonstrated between Cat-NPC2 and Can f 7/Der f 2. Conclusions: In the study, a novel cat allergen, belonging to the NPC2 protein family, was identified and characterized at both molecular and immunological levels. The study will offer a deeper understanding of cat allergens and improve a component-resolved diagnosis in pet allergy.

      • Comparison Different Methods of Intraoperative and Intraperitoneal Chemotherapy for Patients with Gastric Cancer: A Meta-analysis

        Purpose: To investigate the efficacy and safety of intraperitoneal chemotherapy (IPC) for patients with gastric cancer and to compare effects between different regimens of IPC. Method: Randomized controlled trials comparing the effects of surgery plus intraperitoneal chemotherapy with surgery alone or comparing the efficacy between different regimens of intraperitoneal chemotherapy were searched for in Medline, Embase, Pubmed, the Cochrane Library and the Chinese BioMedical Disc and so on by two independent reviewers. After quality assessment and data extraction, data were pooled for meta-analysis using RevMan5.16 software. Tests of interaction were used to test for differences of effects among subgroups grouped according to different IPC regimens. Results: Fifteen RCTs with a total of 1713 patients with gastric cancer were included for quality assessment and data extraction. Ten studies were judged to be of fair quality and entered into meta-analysis. Hyperthermic intraoperative intraperitoneal chemotherapy (HR=0.60, P<0.01), hyperthermic intraoperative intraperitoneal chemotherapy plus postoperative intraperitoneal chemotherapy (HR=0.47, P<0.01) and normothermic intraoperative intraperitoneal chemotherapy (HR=0.70, P=0.01) were associated with a significant improvement in overall survival. Tests of interaction showed that hyperthermia and additional postoperative intraperitoneal chemotherapy did not impact on its effect. Further analysis revealed that intraperitoneal chemotherapy remarkably decrease the rate of postoperative hepatic metastasis by 73% (OR=0.27, 95% CI=0.12 to 0.67, P<0.01). However, intraperitoneal chemotherapy increased risks of marrow depression (OR=5.74, P<0.01), fever (OR=3.67, P=0.02) and intra-abdominal abscess (OR=3.57, P<0.01). Conclusion: The present meta-analysis demonstrates that hyperthermic intraoperative intraperitoneal chemotherapy and normothermic intraoperative intraperitoneal chemotherapy should be recommended to treat patients with gastric cancer because of improvement in overall survival. However, it is noteworthy that intraperitoneal chemotherapy can increase the risks of marrow depression, intra-abdominal abscesses, and fever.
