1,4-Dihydroxy-2-naphthoic acid (DHNA), a precursor of menaquinone (vitamin K2), has an effect on growth stimulation of bifidobacteria and prevention of osteoporosis, making it a promising functional food material. Therefore, we tried to clone the menB gene encoding DHNA synthase from Leuconostoc mesenteroides CJNU 0147. Based on the genome sequence of Leu. mesenteroides ATCC 8293 (GenBank accession no., CP000414), a primer set (Leu_menBfull_F and Leu_menBfull_R) was designed for the PCR amplification of menB gene of CJNU 0147. A DNA fragment (1,190 bp), including the menB gene, was amplified, cloned into pGEM-T Easy vector, and sequenced. The deduced amino acid sequence of MenB (DHNA synthase) protein of CJNU 0147 had a 98% similarity to the corresponding protein of ATCC 8293. The menB gene was subcloned into pCW4, a lactic acid bacteria - E. coli shuttle vector, and transferred to CJNU 0147. The transcription of menB gene of CJNU 0147 (pCW4::menB) was increased, when compared with those of CJNU 0147 (pCW4) and CJNU 0147 (-). The DHNA was produced from it at a detectable level, indicating that the cloned menB gene of CJNU 0147 encoded a DHNA synthase which is responsible for the production of DHNA, resulting in an increase of bifidogenic growth stimulation activity.
The Antithrombotic and Fibrinolytic Effect of Natto in Hypercholesterolemia Rats

Antithrombotic and fibrinolytic activity of natto was evaluated on platelet aggregation in vitro and in vivo. Natto showed inhibitory effects on platelet aggregation induced by adenosine 5’diphosphate (ADP) and collagen. Orally administered natto also showed fibrinolytic activity in hypercholesterolemia rats. Normal levels of natto, when administered for four weeks, shortened euglobulin clot lysis time (ECLT) and prolonged partial thromboplastin time (PATT) significantly compared to non-treated group. In addition, the natto treatment decreased total cholesterol in serum. These results showed that intake of normal levels of natto can elicit antithrombotic and fibrinolytic effects, suggesting its consumption may improve blood circulation.

Previous studies have confirmed that fermented whey produced by Leuconostoc mesenteroides CJNU 0147 or Lactobacillus casei CJNU 0588 display bifidogenic growth stimulator (BGS) activity. The present study sought to determine if the strain itself can improve the growth of bifidobacteria in co-cultures. In reinforced clostridial medium (RCM), both strains stimulated the growth of a Bifidobacterium lactis strain during the exponential phase and also stimulated the growth during almost all growth phases in whey broth. Fermented whey containing viable Leu. mesenteroides CJNU 0147 and L. casei CJNU 0588 cells maintained viability of the B. lactis strain stored at 10℃ in MRS broth. Viable cell count of the B. lactis strain without the fermented whey was decreased to 5.6 log cfu/mL after 15 days, whereas that of the strain with the fermented whey was slightly increased to 7.1 log cfu/mL as compared with initial viable cell count of 6.9 log cfu/mL.

Probiotics comprise living microorganisms that have health benefits for the host when the proper amount is administered and also have a great influence on the immune regulation of the body, for instance, improvement of allergic diseases. Atopic dermatitis (AD) is a major chronic allergic disease. Thus we investigated the anti-allergic effect and associated molecular mechanism of Pediococcus pentosaceus KF159 (PPKF159) in a mouse AD model induced by house dust mite. Our ex vivo studies revealed that PPKF159 treatment decreased levels of Th2 cytokines. Oral administration of PPKF159 either alleviated clinical symptoms of AD or suppressed levels of IgE in the serum. Furthermore, PPKF159 increased the levels of Th1 cytokine IFNγ while decreased IL6, and Th2 cytokines such as IL4, IL5, and IL13 levels in the ear tissue. Our in vivo and ex vivo studies identified that PPKF159 induced anti-inflammatory cytokine IL10 production and increased the proportion of CD4+Foxp3+Treg. Collectively, our studies suggested that PKF159 suppresses AD-like clinical symptoms through the regulation of Th1/Th2 immune balance, inhibition of IgE production, and enhancing Treg and IL10 production.
Heterologous Production of Pediocin PA-1 in Lactobacillus reuteri

The recombinant DNA pLR5cat_PSAB, in which pediocin PA-1 structural and immunity genes (pedAB) fused with the promoter and deduced signal sequence of an α-amylase gene from a bifidobacterial strain were inserted in Escherichia coli-lactobacilli shuttle vector pLR5cat, was transferred to Lactobacillus reuteri KCTC 3679 and the transformant presented bacteriocin activity. The recombinant L. reuteri KCTC 3679 transformed with the shortened pLR5cat(S)_PSAB, where a nonessential region for the lactobacilli replicon was removed, also showed bacteriocin activity. The molecular mass of the secreted pediocin PA-1 from the recombinant bacteria was the same as that of native pediocin PA-1 (~4.6 kDa) from Pediococcus acidilactici K10 on a sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gel. In cocultures with Listeria monocytogenes, the recombinant L. reuteri KCTC 3679 effectively reduced the viable cell count of the pathogenic bacterium by a 3 log scale compared with a control where L. monocytogenes was incubated alone.
Detection of 1,4-Dihydroxy-2-Naphthoic Acid from Commercial Makgeolli Products

To support beneficial effects of makgeolli for human health, we investigated for the presence of 1,4-dihydroxy-2-naphthoic acid (DHNA), a bifidogenic growth stimulator (BGS), from commercial makgeolli products. Among eleven makgeolli products (A~K), four showed positive peaks for DHNA in high performance liquid chromatography analysis. Makgeolli product A in particular contained the highest concentration of DHNA (0.44 ppm), as confirmed by liquid chromatography-mass spectrometry. Furthermore, BGS activity of the makgeolli product A was higher than those of products in which DHNA was not detected. These results indicate that makgeolli can be a good source for DHNA and that DHNA-enriched makgeolli could be developed by modifying manufacturing procedures and controlling its microbiota.
Leuconostoc mesenteroides Producing Bifidogenic Growth Stimulator via Whey Fermentation

Two whey culture supernatants of CJNU 0147and CJNU 0400 were found to effectively enhance the growth of Bifidobacterium longum FI10564 by 1.58 fold compared to non-fermented whey medium. The 2 isolates were identified to be Leuconostoc mesenteroides (99%identity) by 16S rRNA gene sequence analysis. To determine whether the whey culture supernatant of CJNU 0147 selectively stimulate the growth of bifidobacteria, the growth rates of Escherichia coli DH5α, Enterococcus faecalis KFRI 675, Listeria monocytogenes ATCC 19111,and Staphylococcus aureus ATCC 14458 with the supernatant were measured. In these experiments, the supernatant slightly inhibited the growths of bacteria except for E. coli,indicating that the whey culture supernatant had very little influence on the growth of these bacterial strains.
