        Clinical significance of delta neutrophil index in the differential diagnosis between septic arthritis and acute gout attack within 24 hours after hospitalization

        <▼1><P>Supplemental Digital Content is available in the text</P></▼1><▼2><P><B>Abstract</B></P><P>The most important differential diagnoses of acute monoarticular arthritis are septic arthritis and acute gout attack. Identifying infection is crucial in preventing the devastating outcome of septic arthritis. The delta neutrophil index (DNI) is a value that corresponds to the fraction of circulating immature granulocytes. As DNI reflects the burden of infection, we evaluated this index as a differentiating marker between septic arthritis and acute gout attack.</P><P>The medical records of 149 patients with septic arthritis and 194 patients with acute gout attack were reviewed. A specific cell analyzer, ADVIA 2120, was used to measure DNI. Clinical and laboratory markers associated with predicting septic arthritis were assessed by using logistic regression.</P><P>Patients with septic arthritis showed higher levels of DNI than those with acute gout attack (3.3 vs 0.6%, <I>P</I> < .001). Similar results were observed in patients without monosodium urate (MSU) crystal confirmation or those with normouricemia (3.3 vs 0.5 and 3.1 vs 0.7%, respectively; <I>P</I> < .001 for both). A DNI level of 1.9% was determined as the cutoff value for predicting septic arthritis. In the multivariate analysis, DNI was the most powerful independent value for predicting septic arthritis (odds ratio 14.003).</P><P>This study showed the possibility of using DNI as a differentiating marker between septic arthritis and acute gout attack at the crucial early phase. DNI showed its relevance regardless of confirmation of MSU crystal deposition or serum level of uric acid.</P></▼2>

        Exosomes: A new delivery system for tumor antigens in cancer immunotherapy

        <P>Exosomes are small membrane vesicles that are released into the extracellular environment during fusion of multivesicular bodies with plasma membrane. Exosomes are secreted by various cell types including hematopoietic cells, normal epithelial cells and even some tumor cells. They are known to carry MHC class I, various costimulatory molecules and some tetraspanins. Recent studies have shown the potential of using native exosomes as immunologic stimulants. Here, we demonstrate a novel means of using exosomes engineered to express a specific tumor antigen to generate an immune response against tumors. We expressed a target tumor antigen, human MUC1 (hMUC1), in 2 MHC type-distinct mouse cell lines, CT26 and TA3HA. Analysis of exosomes purified from these cells revealed that exosomes contained the target MUC1 antigen on their surfaces as well as other well-described exosomal proteins, including Hsc70 and MHC class I molecules. In addition, both autologous and allogenic exosomes were able to stimulate the activation of immune cells and suppress hMUC1-expressing tumor growth in a MUC1-specific and dose-related manner. Therefore, these data suggest that exosomes can be engineered from tumor cell lines to deliver a target immunogen capable of inducing an effective immune response and that they may represent a new cell-free tumor vaccine. © 2004 Wiley-Liss, Inc.</P>

        센서 네트워크의 데이터 스트림 마이닝을 위한 온톨로지 기반의 전처리 기법

        By a number of sensors and sensor networks, we can collect environmental information from a certain sensor space. To discover more useful information and knowledge, we want to employ data mining methodologies to sensor data stream from such sensor spaces. In this paper, we present a novel data preprocessing scheme to improve the performances of the data mining algorithms. Especially, ontologies are applied to represent meanings of the sensor data. For evaluating the proposed method, we have collected sensor streams for about 30 days, and simulated them to compare with other approaches.

        온톨로지 기반 분산 정보시스템 환경에서는 시스템들 간의 정보 공유을 위해 전문가에 의한 명시적 온톨로지 매핑(Explicit Mapping)을 통해 의미적 이질성(Semantic Heterogeneity)을 해결하고 있다. 하지만, 온톨로지 매핑의 고비용성 때문에 모든 정보시스템들 간의 매핑이 이루어지기 힘들다. 따라서, 본 논문에서는 이미 존재하는 온톨로지 매핑 정보들의 재사용을 통해 존재하지 않은 온톨로지 매핑 정보를 묵시적으로 예측하고자 한다. 본 논문에서는 이와 같은 분산 환경에서의 쿼리 전송을 통한 지식 검색에 있어서의 온톨로지 매핑을 기반으로 한 적절한 쿼리 변환 방법론을 소개하고자 한다.

      • Fine mapping of suppressor of the fruit shape gene OVATE using SNP markers in tomato

        The fruit shape is an important character in tomato. OVATE is one of genes controlling fruit elongation in tomato. Two loci suppress the ovate mutation, sov1 and sov2, on chromosome 10 and chromosome 11 respectively. sov1 appears to control neck constriction in the fruits (Rodriguez et al, 2013). We sequenced the genomes of Gold Ball Livingston and Yellow Pear using the Illumina Hiseq 2000 generating 101 PE reads and developed molecular markers tightly linked to sov1. The locus was confirmed by fruit shape index analysis, marker genotyping and progeny testing of recombinants. We find mapped sov1 to a 145 kb interval corresponding to a region comprising two candidate genes. One of the candidate genes for sov1 is SlOFP20 another member of the Ovate Family Protein class. Although there is no difference expression of SlOFP20 in the parents at anthesis, when the gene is expressed very high, the mutation appears to be a 34 kb promoter deletion of SlOFP20 in Yellow Pear, conferring a pear shaped and neck-constricted fruit.

        SUMOylation of pontin chromatin-remodeling complex reveals a signal integration code in prostate cancer cells.

        <P>Posttranslational modification by small ubiquitin-like modifier (SUMO) controls diverse cellular functions of transcription factors and coregulators and participates in various cellular processes including signal transduction and transcriptional regulation. Here, we report that pontin, a component of chromatin-remodeling complexes, is SUMO-modified, and that SUMOylation of pontin is an active control mechanism for the transcriptional regulation of pontin on androgen-receptor target genes in prostate cancer cells. Biochemical purification of pontin-containing complexes revealed the presence of the Ubc9 SUMO-conjugating enzyme that underlies its function as an activator. Intriguingly, 5alpha-dihydroxytestosterone treatments significantly increased the SUMOylation of pontin, and SUMOylated pontin showed further activation of a subset of nuclear receptor-dependent transcription and led to an increase in proliferation and growth of prostate cancer cells. These data clearly define a functional model and provide a link between SUMO modification and prostate cancer progression.</P>

        분산 환경에서의 쿼리 변환을 위한 온톨로지 매핑 결합

        Semantic heterogeneity should be overcome to support automated information sharing process between information systems in ontology-based distributed environments. To do so, traditional approaches have been based on explicit mapping between ontologies from human experts of the domain. However, the manual tasks are very expensive, so that it is difficult to obtain ontology mappings between all possible pairs of information systems. Thereby, in this paper, we propose a system to make the existing mapping information sharable and exchangeable. It means that the proposed system can collect the existing mapping information and aggregate them. Consequently, we can estimate the ontology mappings in an indirect manner. In particular, this paper focuses on query propagation on the distributed networks. Once we have the indirect mapping between systems, the queries can be efficiently transformed to automatically exchange knowledge between heterogeneous information systems.

      • Short-term weight loss attenuates local tissue inflammation and improves insulin sensitivity without affecting adipose inflammation in obese mice

        <P>Obesity is a major cause of insulin resistance, and weight loss is shown to improve glucose homeostasis. But the underlying mechanism and the role of inflammation remain unclear. Male C57BL/6 mice were fed a high-fat diet (HFD) for 12 wk. After HFD, weight loss was induced by changing to a low-fat diet (LFD) or exercise with continuous HFD. The weight loss effects on energy balance and insulin sensitivity were determined using metabolic cages and hyperinsulinemic euglycemic clamps in awake mice. Diet and exercise intervention for 3 wk caused a modest weight loss and improved glucose homeostasis. Weight loss dramatically reduced local inflammation in skeletal muscle, liver, and heart but not in adipose tissue. Exercise-mediated weight loss increased muscle glucose metabolism without affecting Akt phosphorylation or lipid levels. LFD-mediated weight loss reduced lipid levels and improved insulin sensitivity selectively in liver. Both weight loss interventions improved cardiac glucose metabolism. These results demonstrate that a short-term weight loss with exercise or diet intervention attenuates obesity-induced local inflammation and selectively improves insulin sensitivity in skeletal muscle and liver. Our findings suggest that local factors, not adipose tissue inflammation, are involved in the beneficial effects of weight loss on glucose homeostasis.</P>

      • Effect of surface pre-oxidation on laser assisted joining of acrylonitrile butadiene styrene (ABS) and zinc-coated steel

        <P><B>Abstract</B></P> <P>Surface oxidation pre-treatment of zinc-coated steel was performed to verify its effect on enhancing the joint strength in the laser assisted joining of acrylonitrile butadiene styrene (ABS) and zinc-coated steel. Tensile shear tests indicated that the joint strength between pre-oxidized zinc-coated steel and ABS was considerably enhanced in comparison with that of untreated joint. The zinc oxide layer was generated in the zinc layer by pre-treatment on zinc-coated steel and the thickness of the zinc oxide layer is proportional to the joint strength of joining as long as the strength of zinc oxide layer is higher than that of the jointed layer. When the thicker zinc oxide layer on the zinc-coated steel is reacted with carbon on the ABS, it increases the potential for chemical bonding between the zinc oxide and carbon (ZnOC). Using scanning electron microscopy (SEM) and coupled with energy dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) measurements, the strongly jointed areas were evaluated at atomic or molecular levels to investigate the physical and chemical bonding between the reacted carbon layer and zinc oxide layer.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Effect of pre-oxidation on the laser joining of EGI and ABS was examined. </LI> <LI> Joint strength between pre-oxidized EGI and ABS was considerably enhanced. </LI> <LI> ZnO was one potential contributor to the chemical bonding between the ABS and the Zn layer. </LI> <LI> The thickness of ZnO layer is proportional to the joint strength of LAMP joining. </LI> <LI> Specimen by 400°C oxidation process had a thicker zinc oxide layer and higher joint strength. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
