Impact of interleukin-21 in the pathogenesis of primary Sjogren's syndrome: increased serum levels of interleukin-21 and its expression in the labial salivary glands

<P><B>Introduction</B></P><P>Interleukin (IL)-21 is a cytokine that controls the functional activity of effector T helper cells and the differentiation of Th17 cells, and promotes B-cell differentiation. To test whether IL-21 participates in the pathogenesis of primary Sjögren's syndrome (SS), serum IL-21 level was measured and IL-21 expression in the labial salivary glands (LSG) was examined.</P><P><B>Methods</B></P><P>Serum IL-21 levels in 40 primary SS, 40 rheumatoid arthritis (RA), and 38 systemic lupus erythematosus (SLE) patients and 20 healthy controls were measured. Serum IL-21 levels of SS patients were assessed for correlations with laboratory data, including anti-nuclear antibody, anti-Ro/La antibodies, globulin, immunoglobulin (Ig) class, and IgG subclass. LSGs from 16 primary SS and 4 controls with sicca symptoms were evaluated for IL-21 and IL-21 receptor (IL-21R) expression by immunohistochemistry. Confocal microscopy was performed to further characterize the IL-21 positive cells.</P><P><B>Results</B></P><P>Primary SS patients had significantly higher serum IL-21 levels than controls, and these increments correlated positively with levels of IgG, IgG1. Serum IgG1 levels correlated with anti-Ro antibody titers. Immunohistochemical analyses showed that lymphocytic foci and the periductal area of the LSGs from SS patients expressed high levels of IL-21 and lower levels of IL-21R, whereas the control LSGs showed minimal expression of both antigens. The more the lymphocyte infiltrated, IL-21expression in LSGs showed a tendency to increase. Confocal microscopic analyses revealed that IL-21 expressing infiltrating lymphocytes in the LSGs of SS patients also expressed CXCR5.</P><P><B>Conclusions</B></P><P>Primary SS is associated with high serum IL-21 levels that correlate positively with serum IgG, especially IgG1, levels. The expression of IL-21 is increased as more lymphocytes infiltrated in LSGs. These observations suggest that IL-21 may play an important role in primary SS pathogenesis.</P>

위암에서 Helicobacter pylori cagA, vacA, iceA 유전자와 숙주 Interleukin-1β및 Interleukin-1 수용체 길항제 유전자 다형성

Background: Both Helicobacter pylori (H. pylori) cagA, vacA, iceA genotype and host IL-1B/IL-1RN polymorphisms play a role in determining the clinical consequences of H. pylori infection. This study aimed to investigate whether there might be any combinations of H. pylori cagA, vacA, iceA genotype and host IL-1B/IL-1RN polymorphisms that are particularly associated with the occurrence of gastric carcinoma in Korean patients. Methods: This study population was comprised of 239 patients with H. pylori infection: 122 with gastric carcinoma and 117 with gastritis only. DNA was isolated from gastric biopsy sample and H. pylori cagA, vacA and iceA genotype were determined by PCR. IL-1B-511 polymorphisms were genotyped by PCR-RFLP and IL-1RN polymorphisms were analyzed with variable number of tandom repeat after PCR. Results: H. pylori cagA, vacA, and iceA genotype were not associated with an increased risk for gastric carcinoma. IL-1B-511*T carriers and IL-1RN*2 carriers did not show increased risk for gastric carcinoma. On combination of bacterial/host genotypes, cagA+/IL-1B-511*T carriers and cagA+/IL-1RN*2 carriers, vacA s1/IL-1B-511*T carriers, vacA s1/IL-1RN*2 carriers, vacA m1/IL-1B-511*T carriers, vacA m1/IL-1RN*2 carriers, iceA1/IL-1B-511*T carriers, iceA1/IL-1RN*2 carriers showed no increased risk of gastric carcinoma. Conclusions: Combined H. pylori cagA, vacA, iceA genotype and host IL-1B/IL-1RN polymorphisms shows no increased risk of gastric carcinoma. Therefore, it seems other endogenous or exogenous factors may play more important role in the development of gastric carcinoma in Korean.(Korean J Med 71:24-37, 2006)

Involvement of E-selectin in recruitment of endothelial progenitor cells and angiogenesis in ischemic muscle

<B>Abstract</B><P>E-selectin plays critical roles in tethering leukocytes to endothelial cells (ECs). We studied the role of E-selectin in endothelial progenitor cell (EPC) homing and vasculogenesis. After ischemia, the expression of E-selectin on ECs peaked 6 to 12 hours and returned to baseline at 24 hours, whereas the level of soluble E-selectin (sE-selectin) in serum increased over 24 hours and remained high at day 7. Mouse bone marrow-derived EPCs expressed not only E-selectin but also its ligand. Homing of circulating EPCs to ischemic limb was significantly impaired in E-selectin knock-out mice, as well as wild-type mice pretreated with blocking antibody against E-selectin, which was rescued by local sE-selectin injection. Mechanism for this is that sE-selectin stimulated not only ECs to express ICAM-1, but also EPCs to secrete interleukin-8 (IL-8), leading to enhanced migration and incorporation to ECs capillary formation. In therapeutic aspect, local treatment with sE-selectin enhanced efficacy of EPC transplantation for vasculogenesis and salvage of ischemic limb. Conversely, when E-selectin was knocked down by E-selectin small interfering RNA, blood flow recovery after EPC transplantation was significantly impaired. But this impaired vasculogenesis was rescued by sE-selectin. In conclusion, these data demonstrate E-selectin is a pivotal molecule for EPCs' homing to ischemic limb and vasculogenesis.</P>

가시오가피가 흰쥐의 Collagen 유발 관절염 발생 및 치료에 미치는 영향

Objectives : This study was carried out to investigated the anti-pathogenetic and curative effects of Radix Cortex Acanthopanacis Senticosis (CAS) on collagen-induced arthritis in rats. Methods : In experiment Ⅰ, rats(n=30) were divided into normal group, porcine type II collagen immunized control group which taken with normal saline, and collagen immunized treated group which taken with extract of CAS. Body weight, paw edema volume, and thickness of ankle joint were measured at 0, 10, 15 days after immunization. At 15 days after immunization, serum TNF-α, IL-1β and anti-collagen IgG level were analysed, and histopathological examination was performed on the ankle joint. In experiment Ⅱ, rats(n=13) that were shown the gross lesions of collagen-induced arthritis, were divided into control group and treated group which taken with extract of CAS for 15 days after onset of arthritis. Items and methods of measurement were the same as experiment Ⅰ. Results : In experiment I, the results were as follows ; Arthritic score, paw edema volume, thickness of ankle joint, TNF-α, anti-collagen IgG of treated group were significantly decreased. In experiment Ⅱ, the results were as follows ; Paw edema volume, TNF-α, IL-1β, anti-collagen IgG of treated group were significantly decreased. Conclusions : These results indicated that CAS has the inhibitory effects on the incidence and progression of collagen-induced arthritis in rats.

삼황사심탕(三黃瀉心湯)이 난황 알부민으로 유도된 알레르기 Mouse모델에서 항알레르기 효과

Objectives In this study, we investigated the inhibitory effects of Samhwangsasim-tang (S.H) on the allergic response caused by ovalbumin(OVA) sensitization and challenge in BALB/c mice. Methods The experimental animals were divided into five groups; 1) normal as negative control, 2) OVA-sensitized mice, 3) OVA-sensitized mice treated with 200 mg/kg of S.H 200, 4) OVA-sensitized mice treated with 400 mg/kg of S.H 400, and 5) OVA-sensitized mice treated with 5 mg/kg of Dexamethasone (Dex). Antigen sensitization for allergic mouse model was performed with twice injection of OVA for 2 weeks. After secondary injection, S.H was administrated orally into mice every day for 13 days and the inhibitory effect of S.H on allergic responses was evaluated. Results Treatment of S.H into allergic mice reduced significantly ear edema and infiltration of immune cells in ear tissues induced with OVA challenge in a dose-dependent manner. S.H reduced significantly the serum levels of Total Immunoglobulin(Ig)G and IgE, and particularly inhibited the production of OVA-specific IgE, but not OVA-specific IgG. The serum level of proinflammatory cytokine TNF-α and Th2-associated cytokine IL-4 also were significantly decreased by S.H adminstration in a dose denpendent manner. S.H attenuated OVA-induced secretion of IFN-γ, but not IL-12 which is a cytokine inducing the development of Th1 cells. It also reduced significantly the secretion of IL-4, which is a cytokine inducing the development of Th2 cells, after splenocytes were stimulated with OVA. However the secretion of IL-5 and IL-13 was influenced weakly or a little. Conclusions These results indicate that S.H could reduce the allergic response through inhibition of antigen-specific IgE and Th2-inducing cytokines. It suggest that S.H may be available clinically for the treatment of allergic patients. (J Korean Med Rehab 2014;24(3): 71-85)

This research aims to suggest a desirable way of organizing Korea's Art Textbooks through the comparison and analysis between Korea and Japan's Art Textbook which are being used in high school. Both texts having been divided into three-dimension and plane, mainly the plane section is analyzed by field. The analysis on illustrations and commentaries are classified by country, and I analyze the common and difference by subtitle, focusing on the artists who appear in the both countries texts and others,. From the research I have reached the following conclusions. The first, art education should be aimed to encourage students to have creative, experiential activities, and to use the artistic experience as a means of actieving self-realization and appreciating art works. The content of the textbook , therefore, has to be focused on the procedure of experience in order to cause intrinsic motivation to the students. The second, it is required to add more explanations to the contents of textbooks and their art works and to describe more easily to help students' personal understanding on their expression and appreciation activities. The third the textbooks needs to include different trends of works and genuine works of students' which high school students c deal with. The fourth, it is essential to emphasize the current point of view on modern art and mention information on modern art trend, since the attitudes toward beauty changes with time. As a result, it is necessary to be aware of the current trend in art. The last, apart from only highlighting masterpieces of paintings, there should be a portion which deals with the preparation for the present trend as modern life has become complex and the needs of design and other departments are being stressed. I hope that the above outcomes can be useful for the benefit of Art Education in the Seventh Curriculum, which is going to be applied to high school from next year. Nowadays Art education in Korean is given less attention than other subjects, and there should be improvement in the structures and contents of the textbook. In the current curriculum the art textbooks are organized according to 'Art and Life', 'Painting', 'Sculpture', 'Design', 'Calligraphy', and 'Appreciation'. However, from the Seventh curriculum the texts are described according to 'Aesthetical Experience', 'Expression', and 'Appreciation'. Therefore, the existence of the textbooks is necessary which satisfy the need of students and appeal to them.

The space group of nalidixic acid(1-ethyl-1, 4-dihydro-7-methyl-4-oxo-1, 8-naphthyridine-3-carboxylic acid, C_12H_12N_2O_3), a clinically important antibiotics, has been determined by single crystal X-ray photographic data collected three-dimensionally using Weissenberg method with Cu Kα radiation (λ=1.5418 Å), as the structural data is required and important for the elucidation of the molecular mechanism of the inhibitory action of the antibiotics on DNA synthesis. The space group is P2_1/c with four molecules in the unit cell of dimensions, a=8.98±0.15, b=13.30±0.03, c=9.50±0.16Å, and β=99.5±0.1°.

이족로봇의 경사면 균형 유지와 보행에 관한 연구

This paper aims to provide a way to improve dynamic stability of biped robots against undesirable disturbances and in a slope. By using an angular velocity sensor and an acceleration sensor on its waist, we can make a medium-sized biped robot walk stably in a slope against impulsive disturbances. In addition, it is possible for the robot to walk stably in an unknown slope. The measured signals from the sensor are used for compensating the reference angles of ankle, knee, and pelvis joints. Some experiments show that the stability of the robot is much enhanced by using cheap sensors and a simple algorithm. This work helps biped robots walk more stably in real environments.

The space group os p-toluenesulfonanlide, C_13 H_13 NO_2S, has been determined by single crystal X-ray photographic data collected three-dimensionally using Weissenberg and precession methods with Cu Kα radiation (λ=1.5418 Å). The crystal data are monoclinic, space group p 1 2_1/n 1 with cell dimensions a=15.369±0.423Å, b=9.806±0.009Å, c=8.99±0.03Å, β=98.53±0.18°and z=4. The precise a and b parameters were obtained by finding a proper extrapolation function and using the least-squares refinement of eight 2θ values measured on (hk0) and (h0ℓ) Weissenberg photographs, and because of insufficient number of diffraction-spots along the c^* axis the c parameter and the β angle were measured from oscillation and Weissenberg photographs respectively. Four molecules in a unit cell appear to be related by screw diads parallel to the b axis and n-glide planes perpendicular to it.
