Angiogenesis is an essential event in a variety of physiological and pathological processes. Therefore, effective inhibition of event is a promising strategy for treating angiogenesis-related diseases, including cancer. The current study investigated two unique bafilomycin-type macrolide inhibitors of angiogenesis, PC-766B' (1) and PC-766B (2). The strain RK97-56 which produced the inhibitors was identified as Nocardia sp. by chemotaxonomic analyses, and the purification of the inhibitors was guided by their anti-angiogenic actives. PC-766B' (1) and PC-766B (2) exhibited potent inhibitory activities towards endothelial cell migration stimulated by the vascular endothelial growth factor(VEGF).

Objective: Suvorexant is a novel hypnotic drug that does not interact with the conventional γ-aminobutyric acid (GABA)-A receptor. We investigated the method by which suvorexant was introduced in insomnia patients who were taking benzodiazepine receptor agonists (BzRA). Methods: This was a retrospective study. We extracted clinical data for patients who were prescribed suvorexant and were already using BzRA. The patients were assigned to two groups, the switching and add-on groups. We assessed the suvorexant discontinuation rate at one month after the prescription of the drug. Results: One hundred and nineteen patients were assigned to the switching group, and 109 were assigned to the add-on group. The add-on group exhibited a significantly higher all-cause discontinuation rate than the switching group (odds ratio, 2.7; 95% confidence interval, 1.5 to 5.0; adjusted p<0.001). Intolerability was a significantly stronger risk factor for suvorexant discontinuation in the add-on group (22.0% vs. 7.6%, p<0.002), and the most common adverse effect was oversedation. Conclusion: Our results show that the add-on of suvorexant increases the frequency of oversedation compared with switching in insomnia patients that are taking BzRA. However, this was only a preliminary retrospective study, and further studies will be required to confirm our findings.
236U accelerator mass spectrometry with a time-of-flight and energy detection system

A time-of-flight and energy (TOF-E) detection system for the measurement of 236U accelerator mass spectrometry (AMS) has been developed to improve the 236U/238U sensitivity at Micro Analysis Laboratory, Tandem accelerator (MALT), The University of Tokyo. With observing TOF distribution of 235U, 236U and 238U, this TOF-E detection system has clearly separated 236U from the interference of 235U and 238U when measuring three kinds of uranium standards. In addition, we have developed a novel method combining kernel-based density estimation method and multi-Gaussian fitting method to estimate the 236U/238U sensitivity of the TOF-E detection system. Using this new estimation method, 3.4 1012 of 236U/238U sensitivity and 1.9 ns of time resolution are obtained. 236U/238U sensitivity of TOF-E detection system has improved two orders of magnitude better than that of previous gas ionization chamber. Moreover, unknown species other than uranium isotopes were also observed in the measurement of a surface soil sample, which has demonstrated that TOF-E detection system has a higher sensitivity in particle identification. With its high sensibility in mass determination, this TOF-E detection system could also be used in other heavy isotope AMS

Based on past experiences of natural disasters and fires in Japan, it is stipulated by law that fire-resistant buildings larger than a certain size should be unique in the world. Recent interest in global environmental issues has led to the active introduction of wooden buildings also in Japan, and it is expected that wooden buildings will become larger and higher in size. This paper introduces the background of the development of fire-resistant laminated timber with a "Self-Charring-Stop layer", the contents of this development including other related developments, and the application of these technologies. In addition, towards the realization of much larger and higher buildings in the future, the current problems and issues to be solved are set and the necessity of the future technological development is described. Finally, a conceptual model of wooden high-rise building is proposed, which will be able to be constructed in 2025 by the further technological development.
Identification of glyco-biomarker candidates for lung cancer using novel glyco-technologies

Lung cancer is the leading cause of cancer death worldwide. Currently, lung cancer is classified into two major types, small-cell lung cancer carcinoma (SCLC) and non-small-cell lung carcinoma (NSCLC), based on the histological appearance. The histological classification has important implications in the clinical practice guideline and the prediction of the patient prognosis. However, conventional serum markers used in clinical tests are insufficient for clinical demands due to the low sensitivity and the low specificity to distinguish them. We have identified a number of glyco-biomarker candidate molecules from lung cancer cell lines using our developed glycoproteomics technologies such as lectin microarray and LC/MS-based protein analysis. On the validation studies, we found out that the selected molecules showed characteristic lectin biding profiles depending on either SCLC or NSCLC. Therefore, combination of these glyco-biomarkers could be expected to improve the diagnostic accuracy for histological classification in lung cancer compared to protein expression alone.
