Mice lacking the IL-1R-associated kinase 4 (IRAK4) are completely resistant to LPS-induced endotoxic disorder or the TLR9 agonist CpG DNA plus D-galactosamine-induced acute liver injury (ALI), whereas wild-type strains succumb. However, translational drugs against sepsis or ALI remain elusive. Lonicerae flos extract is undergoing the clinical trial phase I in LPS-injected healthy human volunteers for sepsis treatment. In the current study, chlorogenic acid (CGA), a major anti-inflammatory constituent of lonicerae flos extract, rescued endotoxic mortality of LPS-intoxicated C57BL/6 mice, as well as ameliorated ALI of LPS/D-galactosamine-challenged C57BL/6 mice. As a mechanism, CGA inhibited various TLR agonist-, IL-1 alpha-, or high-mobility group box-1-stimulated autophosphorylation (activation) of IRAK4 in peritoneal macrophages from C57BL/6 or C3H/HeJ mice via directly affecting the kinase activity of IRAK4, a proximal signal transducer in the MyD88-mediated innate immunity that enhances transcriptional activity of NF-kappa B or AP-1. CGA consequently attenuated protein or mRNA levels of NF-kappa B/AP-1 target genes encoding TNF-alpha, IL-1 alpha, IL-6, and high-mobility group box-1 in vivo under endotoxemia or ALI. Finally, this study suggests IRAK4 as a molecular target of CGA in the treatment of innate immunity-related shock and organ dysfunction following insult of various TLR pathogens from bacteria and viruses.

The purpose of this study is to investigate the effects of teenagers` viewing motivation of the mixed martial arts program on viewing attitude and viewing behavior. For this study, teenagers who have seen the mixed martial arts program in Sung-nam and Young-in City were selected as the population. Then, 336 teenagers chosen by systematic stratified cluster random sampling were analyzed. Exploratory Factor Analysis, Reliability Analysis and Multiple Regression were conducted by SPSS Window 12.0. The results were as follows:First, there were statistically significant differences in viewing attitude and viewing behavior according to the gender. Second, there were no statistically significant differences in viewing attitude and viewing behavior according to the school years. Third, there were statistically significant differences in viewing attitude and viewing behavior according to the viewing time. Fourth, there were statistically significant differences in viewing attitude and viewing behavior according to the viewing period. Fifth, it was found that viewing motivation significantly influences viewing attitude and explains 69.2%(R2=.268) of the variance. Vicarious satisfaction, entertainment pursuit motivation, time consuming motivation, and social exchange motivation which are subordinate constructs of viewing motivation have significantly a positive effect on viewing attitude in the order. Sixth, it was found that viewing motivation significantly influences viewing behavior and explains 69.3%(R2=.268) of the variance. Time consuming motivation, entertainment pursuit motivation, vicarious satisfaction, and social exchange motivation which are subordinate constructs of viewing motivation have significantly a positive effect on viewing behavior in the order. However, information pursuit motivation have significantly a negative effect on viewing behavior.

Scrub typhus is an acute febrile disease caused by the pathogenic bacterium Orientia tsutsugamushi, belonging to the Rickettsiaceae family. The shotgun proteomic analysis was performed using the sera of scrub typhus patients to identify the proteins having their origin in O. tsutsugamushi. Three different databases approaches were used for the identification of the proteomes. We identified the RsmD, an RNA methyltransferase as the commonly detected protein from all three approaches. This protein was not detected in the sera of healthy negative controls. We believe that this protein is a potential biomarker of Orientia tsutsugamushi present in the sera of scrub typhus patients.
TCCR/WSX-1 is a novel angiogenic factor in age-related macular degeneration
Sung, Ho Jin,Han, Jung Il,Lee, Ji Won,Uhm, Ki Bang,Heo, Kyun Molecular Vision 2012 Molecular vision Vol.18 No.-

Purpose

Age-related macular degeneration (AMD) is the major cause of blindness among persons aged 60 years and older. The current approved therapies for AMD are exclusively limited to inhibiting vascular endothelial growth factor. However, substantial improvement in vision occurs in only one-third of patients treated with vascular endothelial growth factor antagonists, and one-sixth of treated patients still progress to legal blindness. Therefore, more specific targets are needed to treat AMD. Our goal was to find secretory proteins that change in number in the aqueous humor and that cause exudative AMD disease.

Methods

The number of molecules changed in the aqueous humor of patients with AMD compared to the control group was determined using antibody array analysis. The levels of angiopoietin-2 and insulin-like growth factor binding protein-related protein 7 were measured using enzyme-linked immunosorbent assay. The levels of T-cell cytokine receptor (TCCR/WSX-1) were determined using western blot. Potential TCCR/WSX-1-mediated effects on tube formation as well as phosphorylation of extracellular signal-regulated kinase in human umbilical vein endothelial cells were determined.

Results

We found that the numbers of several molecules were changed in the aqueous humor of patients with AMD compared to the control group. Among them, angiopoietin-2 was reduced by 20% and TCCR/WSX-1 was increased twofold. Moreover, exogenous TCCR protein induced tube formation and phosphorylation of extracellular signal-regulated kinase in human umbilical vein endothelial cells.

Conclusions

Our study suggests that TCCR/WSX-1 is closely associated with angiogenesis and could serve as a novel therapeutic target in patients with AMD.

Content delivery service has become a major traffic load on today's Internet, and this has triggered the interest of ISPs in operating their own content delivery networks (CDNs) to optimize Internet traffic considering both content delivery caching and user-network proximity. ISPs, however, are typically regionally bound or network-domain-wise isolated; hence, their CDN gain is somewhat limited. In order to enhance the gain of ISP CDN services to the level of incumbent global CDNs, a CDN interconnection (CDNI) model is introduced by IETF, where local ISP CDN services can be extended among heterogeneous CDNs across network domains. However, despite the multiple benefits of a CDNI system, it is difficult to apply a CDNI service to the current CDN market due to the platform independence. Hence, we introduced a CDNI gateway model that is standard-capable and platform-independent. With the CDNI gateway model, we design and implement a complete CDNI system and conduct a CDNI service trial with three major ISPs in South Korea. To the best of our knowledge, this is the first CDNI service trial complying with the IETF standard achieved by a multi-ISP collaboration. According to the analysis of experimental results from the service trial, we observe that CDNI can reduce content traffic by about 40 percent at the Internet exchange link compared to a legacy CDN system.

Assembly and Densification of Nanowire Arrays via Shrinkage
Bang, Jaehoon,Choi, Jonghyun,Xia, Fan,Kwon, Sun Sang,Ashraf, Ali,Park, Won Il,Nam, SungWoo American Chemical Society 2014 NANO LETTERS Vol.14 No.6

Chemically synthesized semiconductor nanowires (NWs) have demonstrated substantial promise for nanoelectronics, nanoenergy, and nanobiotechnology, but the lack of an effective and controllable assembly process has limited the wide adoption of NWs in these areas. Here we demonstrate a facile, robust, and controllable approach to assembling and densifying a parallel array of NWs using shrinkable shape memory polymers. Using thermal-induced shrinkage of polystyrene, we were able to successfully assemble and densify NW arrays up to close-packing and, furthermore, achieve tunable density (up to ∼300% amplification of density) by controlling the shrinkage process. We also demonstrate scalable assembly and densification of NWs on a 2.5 × 6 inch scale to explore the manufacturability of the shrink-induced assembly process. Finally, we demonstrate the successful transfer of the shrink-assembled NW arrays onto various 2-dimensional and 3-dimensional substrates without compromising the integrity of NW assembly and density.

Graphic Abstract

ACS Electronic Supporting Info
