Toxicological Studies on Aucubin (Ⅰ) : Acute Toxicities and Effects on Blood Serum Enzymes

Aucubin, an iridoid glucoside which was previously reported to exhibit liver-protective activities against CCl₄ and α-amanitin induced liver damages, was subject to toxicological studies. To measure the lethal dose, the doses of 100 ㎎/㎏, 300 ㎎/㎏, 600 ㎎/㎏ and 900 ㎎/㎏ were administered intraperitoneally to experimental mice. No death was observed 24 hrs later, but serum GOT and alkaline phosphatase activities were decrased slightly at the doses of 300 ㎎ to 900 ㎎/㎏, and the triglyceride contents were slightly increased. To investigate acute toxioity of aucubin itself, multiple dosages (20 ㎎/㎏, 40 ㎎/㎏ and 80 ㎎/㎏ for four times a week) were injected intraperitoneally into mice, then serum enzymes activities and chemistries were assayed; no significant change of the enzyme activities of alkaline phosphatase, GPT, GOT in the test groups were observed in comparison with those of the control group, and the contents of triglyceride, glucose, urea nitrogen and total proteins in the test group serums appeared to be almost same levels as those of the control group were. Histological examiation on liver biopsy samples indicated no gross changes between the control group and the test group were noted. Therefore, aucubin appears to be apparantly low toxic substance and its minimum lethal dose in mouse seems to be more than 0.9 g.

Methanol extracts of the roots of Atractylodes japonica (alba) (Compositae) and Gentiana scabra (Gentianaceae) were further fractionated with n-butanol, chloroform, n-hexane and water. Then each fraction was p.o. administered to an animal model of hepatitis caused by CCl₄ intoxication. Measurements of sleeping time of mice induced by hexobarbital administration indicated that n-butanol and chloroform fractions of Atractylodes japonica (alba), and chloroform and n-hexane fractions of Gentiana scabra showed significant liver protective activities. Measurements of the activities of serum transaminase, S-GOT and S-GPT appeared to support these results.

Water fractions prepared from Plantaginis Semen and Plantaginis Folium exhibited both significant liver-protective activities against CCl₄ intoxication. Aucubin, an iridoid glycoside, isolated from both leaves and seeds of Plantago asiatica(Plantaginaceae) also showed potent liver protective activities against the animal model of hepatitis induced by CCl₄ intoxication.

보간성 생약, 차전자의 보간효과를 나타내는 분획인 물분획으로부터 C<sub>12</sub>H_(20)O_(10) m.p 186°의 disaccharide를 분리하였으며 그 구조는 6-0-(3`,6`-anhydro-glucosyl)-glucose로 추정하였다.

β-Sitosteryl-3-O-β-D-glucopyranoside was isolated from the seeds of Plantago asiatica(Plantaginaceae). The assignments of <sup>13</sup>C NMR spectra of β-sitosterol and β-sitosteryl-3-O-β-D-glucopyranoside were made by comparing with <sup>13</sup>C NMR spectra of cholesterol and cholesteryl-3-O-β-D-glucopyranoside. Our data indicate that the revision of previous ^(l3)C NMR spectral assignment is needed.

Total methanol extract of Plantago asiatica seeds showed significant hepatotonic activities against animal model of hepatitis. The methanol extract was further fractionated with petroleum ether, chloroform, n-butanol and water. Among these fractions, methanol extract and water and chloroform fractions exhibited hepatonic activities, whereas petroleum ether and n-butanol fractions appeared to give rise a rather toxicity. Measurements of serum GOT, GPT and duration of sleeping time induced by hexobarbital after CCl₄ intoxication indicated that water fraction showed most potent activities.

Total fourty-four medicinal plants described in old literatures to be hepatotonic were evaluated their potential hepatotonic activities against an animal model of hepatitis. Tweenty-seven plants extracts appeared to be significant hepatotonic activities, whereas twelve plants extracts exhibited rather some toxicity. And five plants extracts showed no significant hapatotonic activities and toxicity.

Alisol A monoacetate, alisol B monoacetate, alisol C monoacetate and alisol B were isolated from Alismatis Rhizoma, which is a herbal drug used frequently in the oriental prescriptions. Potential liver-protective activities of the isolated alisol compounds were evaluated against CCl₄-induced liver damage. The results obtained from liver microsomal enzyme assay, measurement of serum glutamic pyruvic transaminase (EC 2.6.1.2) and serum triglyceride content indicated that alisol A,B and C monoacetates showed significant liver-protective activities against CCl₄ poisoning. Alisol B monoacetate exhibited slightly higher activity than that of alisol B.

Several medical surveys including W.H.O. report indicate that exceptionally high incidence rate of liver diseases such as hepatitis and cirrhosin occurrs to Korean peoples. For the purpose of treating liver illness or protecting liver damages from various factors such as virus and toxic chemicals, the present study is aimed to investigate potential hepatotonic substances and/or therapeutic agents originated from natural products like medicinal plants. Total methanol-water extracts were prepared with 44 species of plants obtained in Korea. Then these extracts were evaluated against the animal model of hepatitis. Among them, 6 species of plants extracts showed significant hepatotonic activities, and 21 extracts exhibited median activities, whereas 12 extracts appeared to be rather toxic.
Inhibition of Ribonucleic Acid Biosynthesis in Leukemia SN 36 Cells in vitro by Aucubin

Aucubin, an iridoid gludoside, was isolated from Aucuba japonica(Cornaeae). One of biological activities of aucubin was found to inhibit RNA biosynthesis in leukemia SN 36 tumor cells in vitro. Aucubin appeared not to possess antitumor activities against leukemia SN 36 murine tumor. The combination therapy with aucubin plus actinomycin D did not show any significant additive or synergistic antitumor activities in comparison with that of actinomycin D alone treatment. Iridoid glucoside인 aucubin을 식나무, Aucuba japonica(Cornaceae)로 부터 순수분리하였다. 이 물질의 약리작용을 밝히기 위하여 동물암 세포인 leukemia SN 36 세포의 RNA 생합성에 미치는 영향을 조사한 결과, 농도 0.5 mM 및 1.0 mM에서 각각 30% 및 46% 정도의 RNA 생합성 억제작용을 aucubin은 갖고 있음을 알 수 있었다. Aucubin 자체는 동물암인 leukemia SN 36에 큰 항암작용을 나타내지는 못하였고, Actinomycin D와 병용투여시에도 additive 또는 synergistic 효과 역시 나타내지는 않았다.
