As part of the Cultural Heritage Administration project, we have been evaluating microbial diversity in Jeju volcanic caves. It is aiming to provide microbial diversity of the caves, about which little is known. Samples were collected from Manjang (MJ) Cave and Yongcheon (YC) Cave, while they may play roles in the subsurface ecosystem. It is to analyze microbiome found in lava tube from two different types of lava tubes whether entrance of lava tube is closed or open type. The management of the caves should be careful, above all, the latter is extremely fragile with values. These studies will result in a multitude of new insights into the dynamics between microbes and the cave environments and will have the potential to elucidate development of colorful wall mats. Direct cave sampling led to the characterization of members of a microbial community in cave wall mats of YC Lava Tube and MJ Lava Tube at Jeju Island. Samples were mainly taken from white or yellow mats forming on the surface of lava tubes. After extracting DNA from the samples, metagenomic Next-Generation Sequencing (mNGS) will be performed using the single molecule, real-time (SMRT) sequencing that allows us whole metagenomic shotgun sequencing (Metagenomics). Further analyses of these data will provide insights into the ecological roles they play, and how microbes inhabit in such lava tube environments.
