        미생물,발효,생물공학 : Auxin과 항진균물질을 생산하는 식물생장촉진근권세균의 분리동정 및 특성

        This study was investigated the physiological properties of auxin-producing bacteria that have plant growth promoting activity and plant pathogen antagonistic ability. Auxin-producing bacteria were isolated from field soils of Gyeongsan, Korea. Selected strains were identified as a Pseudomonas fulva N2l and a Pantoea agglomerans K35 by morphological and physiological test, and Biolog (Microlog) system. Auxins were determined by Salkowski in vitro test and mungbean adventitious root induction bioassay. Also produced indole-3-acetic acid (IAA) was identified by TLC. During cell growth, auxin production were highest in their idiophase after log phase and 35℃ at pH 7.5.

        항독성 소간추출물의 생산을 위한 효소분해의 최적화

        소간추출물은 각종 간질환에 해독효과를 가진 것으로 알려져 있다. 항독성 소간추출물의 대량생산 공정을 개발하고자 효소의 선발 및 가수분해의 최적조건을 탐색하였고, 확립된 분해조건으로 항독성 소간추출물의 시제품을 제조하여 일반성분, 미생물, 그리고 비타민 B₁₂함량을 분석하였다. Bromelain, papain, ficin, pancreatin, 그리고 protease NP등의 효소 중 추출물의 건물량과 아미노태 질소의 수율이 양호하고 경제성이 뛰어난 papain을 최적효소로 선발하였다. Papain(1%)을 소간 분쇄물에 첨가하여 65℃에서 24시간 동안 가수분해하는 조건을 확립하였다. 분무건조한 항독성 소간추출물의 시제품은 원료 간 대비 11%의 수율을 보였고 건물량은 95%, 총질소 함량은 11.8%이었다. 또한 병원성 미생물은 검출되지 않았고 비타민 B₁₂ 함량은 4.1 ㎍/g이었다. 본 연구에서 확립한 가수분해조건은 높은 수율의 항독성 소간추출물 생산에 적용할 수 있을 것으로 생각한다. Bovine hepatic extract is recognized as possessing detoxifying activity against various liver diseases. In order to develop a process for its mass production, various enzymatic hydrolysis conditions were tested, and bovine hepatic extracts were prepared. These extracts were then examined for composition, microorganism levels, and vitamin B₁₂ content. Among the enzymes tested, papain was selected based on yields for dry residue and amino nitrogen. The other enzymes tested included bromelain, ficin, pancreatin, and protease NP. The optimal hydrolysis conditions were established at 65℃ for 24 hr, with an addition of 1%(w/w) papain to the beef liver. The prepared spray-dried bovine hepatic extract showed an 11% recovery yield on a raw beef liver basis, with 95% dry residue and 11.8% total nitrogen content. Microorganisms were not detected in the dried extract, and its vitamin B₁₂ content was 4.1 ㎍/g. In summary, the conditions established in this study could be applied for the high yield mass production of bovine hepatic extract.

        In Drosophila melanogaster the interactions of the P=M system generate germ-line aberration(e. g., sterility, mutations)found in certain interstrain hybirds. Gwacheon wild populations from Korea were examined in order to determines the distribution of chromosomal P factor and extrachromosomal cytotypes, and selection for the M cytotypes. The P factor was found to be very rare in this populations. P factor to be lost in lines take from this populations and inbred in this laboratory for many generation. A large scale search for the M cytotype, which cause susceptibility to the P factor showed that it is present in the population at only very low frequencies. Distribution of P and M strains observed 4(2.8%) and 2(1.24%) lines, and Q and M', were 100(62.11%) and 55(34.17%) lines, respectively. For the analysis of effect in selection for the M cytotype, two traits of chromosome type and cytotype resulted from P and M potentials were performed though 13 generations. Moderate P factor activity was shown in P strain at G₁stage, which GD sterility of Cross A was 16.02%, and then decreased gradually as 1.48%, at G? stage. In the Cross B for the cytotype, GD sterility increased through 13 generations from 5.06% to 9.65%. However, the change of chromosome type and cytotype were not occured in Q and M' strains. The frequency of the M cytotype could not usually be increased by artificial selection. No significant difference in distribution of P element, between each cytotype of P-M system was found.
