      • Cortisone이 綱內系細胞의 탐식機能에 미치는 影響

        It has been claimed that the capability of the Kupffer cells of the liver to have carbon particles adhere on the surface membrane of the Kupffer cells was curtailed in experimental rats when the reticuloendothelial system of the animal was blockaded previously. With this informaton, investigation was undertaken to study the phagocytic activity and pathological changes of the subcellular organelles of the phagocytes of both liver (Kupffer cells) and spleen of the rats with prior reticuloendothelial system blockade by administration of cortisone acetate followed by intravenous injection of india ink. Twenty-four albino rats weighing 200gm. in average were divided into 3 experimental groups. Group 1. a. Two rats. untreated. b. To 6 rats, a single dose of 1.0㎖ of 3% india ink in normal saline was given intravenously through the tail vein. c. To 2 rats, a dose of 2.5㎎ of cortisone acetate was given intramuscularly for 3 consecutive days. d. To 2 rats, a dose of 25㎎ of cortisnoe acetate was given intramuscularly for 3 consecutive days. Group 2. To 6 rats was given 2.5㎎ of cortisone acetate, intramuscularly once a day for 3 consecutive days, followed by intravenous injection of 1.0㎖ of 3% india ink in normal saline on the 4th day. Group 3. To 6 rats was given a dose of 25㎎ of cortisone acetate intramuscularly, once a day for 3 consecutive days, followed by intravenous injection of 1.0㎖ of 3% india ink in normal saline on the 4th day. Three each of the experimental rats from groups 2 and 3 were sacrificed 15 and 30 minutes after injection of india ink. Control group was sacrificed accordingly. Appropriate samples were taken for both light and electron microscopic studies. The results were as follows: 1. Histologically, the phagocytic activity of the Kupffer cells of the liver and phagocytes of the spleen was markedly diminished in the group with large doses of cortisonoe acetate (group 3) whereas the phagocytic activity was considerably decreased in the group with small doses of cortisone acetate (group 2) when compared with the control group (group 1). 2. Active phagocytic activity can be demonst rated by electron microscope in the control group (b of group 1) in that the Kupffer cells and phagocytes of the spleen were seen taking up carbon particles vigorously in their cytoplasm. On the contrary, the group with cortisone treatment with resultant reticuloendothelial system blockade followed by india ink administration (group 3) failed to demonstrate this phagocytic activity at all. The carbon particles were found to be outside of the surface membrane of these phagocytic cells. In conclusion, when the reticuloendothelial system was blockaded, the surface attachment phase of the surface membrane of phagocytes was hampered which leads to an insufficient phagocytic activity of the Kupffer cells and phagocytes of the spleen.
