        육미지황탕(六味地黃湯),택사탕(澤瀉湯), 실비음(實脾飮)이 메산지움세포 증식, Fibronectin 합성 및 MHC-class II 발현에 미치는 영향(影響)

        Objective : To analyze the effects of Yukmijihwang-tang, Taeksa-tang, Silbi-um on mesangial cell proliferation, fibronectin synthesis and MHC-class II expression. Methods : Laboratory studies were performed with the method of surface enzyme immunoassays or flow cytometry after addition of peripheral blood mononuclear cells(PBMC) supernatants treated with medications using the cultured human mesangial cells. Results : 1. Silbi-um produces more suppressive effect than control group and hydrocortisone group on the mesangial cell proliferation. In Yukmijihwang-tang, Taeksa-tang and Silbi-um, mesangial cell proliferation significantly decreased than in hydrocortisone group 2. In the 'without fetal bovine serum' study, Yukmijihwang-tang take more suppressive effect than Control group on the fibronectin synthesis. In the 'with fetal bovine serum' study, Yukmijihwang-tang, Taeksa-tang, Silbi-um all have suppressive effect, but it hasn' t any statistical significance. 3. Yukmijihwang-tang, Taeksa-tang, Silbi-um all have a suppressive effect on the MHC-class II expression. Conclusions : Herb medicine generally show a suppressive effect on the suppression of the mesangial cell proliferation, fibronectin synthesis and MHC-class II expression.

        실비음(實脾飮)이 Zucker rat의 비만 및 항산화에 미치는 영향

        Objective : This study was carried out to examine the effect of Silbi-um(Shipi-yin, SBU) on obesity and antioxidation.Methods and Materials : SBU was investigated effects of obesity and antioxidation for DPPH radical ability, nitrate reductase activity, total body weight and fed ratio, serum glucose content, serum triglyceride and total cholesterol content, serum GPT and GOT content, serum leptin content, catalase activity, superoxide dismutase activity, lipidperoxidation and glutathione activity. In this experiment rats were divided into 4 group, Lean control; non-treated, Lean A; SBU, Fat; control: non treated, Fat A; SBU, Each value is the mean ± S.E for 3 separate experiments. Result and Conclusions : In vitro, DPPH radical ability and nitrate reductase activity were increased according to the increasing of SBU concentrations. In vivo, compared with Fat control group, the serum glucose content, the serum triglyceride content, the serum total cholesterol content the serum GPT and the serum GOT content were decreased in Fat A group. The serum leptin content was increased, too. Catalase activity and superoxide dismutase activity were increased and lipidperoxidation was decreased in Fat A group compared with Fat control group. Glutathione activity was not increased significantly. According to the above results, the obesity and the antioxidant activities of SBU were proved in experiment and it should be studied and developed more throughly.

        흰쥐의 알코올 유발성 간손상에 실비음(實脾飮)이 미치는 보호 효과

        Objectives: The objective of this study is to investigate the effects of Silbi-um (SBU) extract on the alcoholic fatty liver induced by EtOH administration for 8 weeks. Methods: Male Sprague Dawley rats were used. All animals were randomly divided into 3 groups; Normal, EtOH and EtOH+SBU. The rats of EtOH group were daily treated with ethanol of 25% (v/v) for 8 weeks (n=10). EtOH+SBU group was orally treated with SBU water extract after ethanol administration (n=10). The rats of Normal group were treated with saline (n=10). After 8 weeks, the mean body weight, liver weight, and liver-body weight ratio were calculated. The serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) of all groups were measured. The morphological alterations were observed using hematoxylin and eosin (H&E) and Oil Red O staining. Moreover, the alteration of tumor necrosis factor-α (TNF-α) levels were analyzed immunohistochemistrically. Results: The histological data showed that liver sections from EtOH group displayed severe steatosis. SBU extract significantly inhibited the progression of the alcoholic liver injury. The increased serum level of ALT and AST induced by ethanol administration were decreased by SBU extract. Furthermore, SBU extract significantly decreased the liver concentrations of TNF-α. Conclusions: SBU water extract attenuated the alcohol induced fatty liver by improving hepatic lipid metabolism via suppression of TNF-α protein. SBU could be effective in protecting the liver from alcoholic fatty liver.
