        하고초 추출물로부터 분리된 Danshensu의 MK-801으로 유도된 사전자극 억제 손상의 회복에 대한 작용

        Schizophrenia is a severe psychiatric disorder and characterized by positive symptom (i.e., delusions, hallucinations),negative symptom (i.e., lack of emotion, social withdrawal), and cognitive impairment. Previously, we reported that the ethanolic extract of Prunella vulgaris var. lilacina attenuated the MK-801-induced schizophrenia-like behaviors such as prepulse inhibition (PPI) deficits and cognitive impairment in mice. The aim of the present study was to investigate whether danshensu isolated from P. vulgaris var. lilacina attenuates MK-801-induced sensorimotor gating dysfunction (PPI deficits),hyperlocomotion, and memory impairment in mice. Acute administration of danshensu (1, 3, or 10 mg/kg) significantly ameliorated the MK-801-induced PPI deficits in the acoustic startle response test. We also observed that the impaired recognition memory induced by MK-801 was attenuated by danshensu (1 mg/kg) in the novel object recognition test. However, danshensu failed to reverse the MK-801-induced hyperlocomotion in the open-field test. Collectively, the present results indicate danshensu would be an active agent for treating neuropsychiatric disorders such as schizophrenia.

      • Identification of a Potential Anticancer Target of Danshensu by Inverse Docking

        Objective: To study potential targets of Danshensu via dual inverse docking. Method: PharmMapper and idTarget servers were used as tools, and the results were checked with the molecular docking program autodock vina in PyRx 0.8. Result: The disease-related target HRas was rated top, with a pharmacophore model matching well the molecular features of Danshensu. In addition, docking results indicated that the complex was also matched in terms of structure, H-bonds, and hydrophobicity. Conclusion: Dual inverse docking indicates that HRas may be a potential anticancer target of Danshensu. This approach can provide useful information for studying pharmacological effects of agents of interest.

        Evaluating Pharmacological Effects of Two Major Components of Shuangdan Oral Liquid: Role of Danshensu and Paeonol in Diabetic Nephropathy Rat

        Shuangdan oral liquid (SDO) containing radix Salviae miltiorrhizae (Chinese name Danshen) and cortex moutan (Chinese name Mudanpi) is a traditional Chinese medicine using for treating vascular diseases. Danshensu (DSS) is a main effective monomer composition derived from radix Salviae miltiorrhizae and paeonol (Pae) from cortex moutan. Although the two herbs are widely used in traditional Chinese medicine, the pharmacological functions of their active compositions were not reported. Therefore, the research of DSS and Pae in mechanisms and pharmacodynamics interaction can provide scientific evidence to support clinical application. The diabetic nephropathy (DN) rats which were induced by streptozotocin (STZ) were treated with SDO, DSS, Pae, and DSS+Pae for eight weeks. The positive effects on DN animal models were investigated by detection of physiological and biochemical indexes and oxidative stress markers, within five treatments: SDO, DSS, Pae, DSS+Pae and insulin group. Compared with the model group, the DSS+Pae group improved the renal function, blood lipid metabolism and blood viscosity, increased the vitality of T-SOD or T-AOC and decreased the level of MDA or NO after the treatment. The study was successfully showed that the DSS+Pae group could delay the process of DN, especially in the renal injury part of histopathology changes. Our results suggest that the co-administration of DSS and Pae significantly may play a protective role in DN rats through decreasing the oxidative stress and improving the blood lipid metabolism mechanisms.

        Pretreatment with Danshensu Promotes Germination of Cucumber (Cucumis sativus L.) Seeds by Regulating Reactive-Oxygen Species and Endogenous Phytohormones

        Danshensu (DSS), the aqueous extracts of Salvia miltiorrhiza has been widely used in biomedical science due to its outstanding capacity in ROS scavenger. However, studies regarding its effects on plant physiology are still scare. In the present study, cucumber seeds were soaked in different concentrations of DSS solution for 24 h before subjected to germination to test its effects on seed germination, and further to explore the metabolic and molecular mechanisms. The results shown that 0.1% DSS had significantly promoted the germination rate, radical length, as well as soluble sugar and H2O2 contents. DSS pretreatment upregulated transcript levels of genes involved in SOD, POD, and CAT biosynthesis, concurrent with promoted enzyme activities. In accordance with lower ABA, but higher GA contents, transcript levels of genes involved in ABA and GA biosynthesis were down and upregulated, respectively, in DSS pretreated seeds. DSS pretreatment also stimulated ethylene production and related genes expression levels. These positive effects of DSS on seed germination were confirmed by addition of inhibitors of GA, ABA, NADPH oxidase, and H2O2 scavenger. In conclusion, DSS stimulated cucumber seed germination by mediating the regulation of endogenous hormones with promoted ROS levels. Our results highlight the potentiality of DSS in plant science.

        중의 약재로부터 피부 미백제의 탐색

        본 연구에서는 미백 화장품 원료를 개발하기 위해 중국에서 전통적으로 사용되어 오던 47종의 천연약재 또는 복합처방 중에서 멜라닌 생성 저해 효과를 지닌 원료를 찾고자 하였다. 그 중 버섯 티로시나제 활성 실험에서는 저해효과를 보이지 않았으나, B16-F10 멜라닌 생성세포(B16-F10 melanoma cell)를 이용하여 류기노와 단삼을 선별하였으며 단삼에서 단삼소(丹蔘素, α,3,4-trihydroxybenzenepropanoic acid sodium salt)를 분리하여 B16-F10 멜라노마 세포를 이용하여 멜라닌 생성 억제에 관한 실험을 실시하였다. 류기노와 단삼소는 농도에 따라 멜라닌 생성을 억제하였으며, 류기노는 300 μg/mL의 농도에서 약 60%, 단삼소는 100 μg/mL의 농도에서는 약 50 %의 멜라닌 생성 저해 효과를 보였다. 따라서 연구 결과로써 얻어진 2종의 원료는 새로운 천연 미백 소재로 적용할 수 있을 것으로 기대된다. In order to search for new whitening cosmetic ingredients from Chinese herbal extracts including Chinese herbs and complex prescriptions from TCM (Traditional Chinese Medicine), we screened about 47 TCM extracts collected from China. We tested their inhibitory effects on melanogenesis by using in vitro tyrosinase inhibition assay and B16 melanoma cells. We selected Siphonostegia chinensis and Salvianic miltiorrhiza Bunge. We isolated Danshensu (α,3,4-trihydroxybenzenepropanoic acid sodium salt) from Salvianic miltiorrhiza Bunge extract and tested its inhibitory effect on melanin formation in B16-F10 melanoma cells. Danshensu suppressed melanin synthesis up to about 50 % at a concentration of 100 μg/mL. Siphonostegia chinensis suppressed melanin synthesis up to about 60 % at a concentration of 300 μg/mL. The results showed that these extracts could be used as new natural active ingredients for whitening cosmetics.
