        Refolding Behavior of Urea-Induced Denaturation Collagen

        Exploration of the denaturation and refolding of natural collagen is important for the application of collagen and its denatured products. In this study, using urea as a denaturant, we prepared a denatured natural collagen product and analyzed its structural changes. The denaturation treatment severely destroyed the triple helix conformation of collagen, but had no significant effect on the primary structure of its α chains or the covalent cross-linking between α chains. Next, we observed the refolding behavior of the denatured collagen by removing urea through dialysis. We found that the denatured collagen products from different sources (grass carp skin, bovine tendon) all showed a reconstruction of the triple helix conformation up to 60-75% of the value of natural collagen during the refolding process. The telopeptide did not significantly promote triple helix reconstruction. In conclusion, the reconstruction of the α chains did not perfectly occur in a “head-to-head, tail-to-tail” manner in refolded collagen, as each α chain was participating in the reconstruction of multiple triple helix domains. The refolded collagen still had weak self-assembly ability and formed a unique network-like structure containing small interlaced and closely combined fibers, which shows favorable cell compatibility and potential applications.

        Corroded pipeline failure analysis using artificial neural network scheme

        <P><B>Abstract</B></P> <P>Corrosion defects occur very often on the internal and external surfaces of pipelines, which may result in a serious threat to the integrity of the pipelines. Numerous studies investigated failure behavior of corroded pipelines with single corrosion defects. However, few studies focus on interacting corrosion defects. Interacting defects are defined as defects with certain proximity that interact to reduce the overall strength of a pipeline. In the present study, the failure behavior of pipelines with interacting corrosion defects was studied using a finite element method, and then a solution was proposed to predict burst pressure using an artificial neural network. The solution was validated by experimental results in previous studies and compared with other existing assessment solutions to prove its applicability and efficiency.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The failure behavior of pipelines with interacting corrosion defects was studied using FE method. </LI> <LI> A series of models were created for the sensitive study of the various parameters. </LI> <LI> A solution was proposed to predict burst pressure using an artificial neural network (ANN). </LI> <LI> The solution was validated by comparing with experimental results and existing codes. </LI> </UL> </P>

      • Expression and Significance of Microsomal Prostaglandin Synthase-1 (mPGES-1) and Beclin-1 in the Development of Prostate Cancer

        The aim of this study was to investigate the expression and significance of microsomal prostaglandin synthase-1 (mPGES-1) and Beclin-1 in the development of prostate cancer (PCa). Immunohistochemistry was performed on paraffin-embedded sections with rabbit polyclonal against mPGES-1 and Beclin-1 in 40 PCa, 40 benign prostatic hyperplasia (BPH) and 10 normal prostate specimens for this purpose. Quantitative real-time polymerase chain reaction (qRT-PCR) was applied for mRNA expression of mPGES-1 and Beclin-1, while MTT assays were used to ascertain the best working concentration of the mPGES-1 inhibitor (CAY10526). The effect of CAY10526 treatment on expression of Beclin-1 in DU-145 cells was studied using Western blot analysis. Localization of Beclin-1 and mPGES-1 was in endochylema. Significant differences in expression was noted among PCa, BPH and normal issues (P<0.05). Beclin-1 expression inversely correlated with mPGES-1 expression in PCa tissue (P<0.05). CAY10526 could significantly block mPGES-1 expression and the proliferation of DU-145 cells (P<0.05), while increasing Beclin-1 levels (P<0.05). Overexpression of mPGES-1 could decrease the autophagic PCa cell death. Inhibiting the expression of mPGES-1 may lead to DU-145 cell death and up-regulation of Beclin-1. The results suggest that inhibition of mPGES-1 may have therapeutic potential for PCa in the future.

      • Colorectal Cancer Incidence and Mortality in China, 2010

        Background: The National Central Cancer Registry of China (NCCR) affiliated to the Bureau of Disease Control, National Health and Family Planning Commission of China is responsible for cancer surveillance in the entire country. Cancer registration data from each local registry located in each province are collected by NCCR annually to be analyzed and published to provide useful information for policy makers and cancer researchers. Materials and Methods: Until 1st June, 2013, 219 population-based cancer registries submitted data of 2010 to the National Central Cancer Registry of China covering about 207,229,403 population, and 145 cancer registries were selected after quality evaluation for this study. Colorectal cancer cases were selected from the database according to ICD-10 coded as "C18-C20". We calculated the crude incidence and mortality rates by sex, age groups and location (urban/rural). The China population in 2000 and Segi's population were used as standardized populations for the calculation of age-standardized rates. The 6th National Population Census data of China was used to combined with the cancer registries' data to estimate the colorectal cancer burden in China in 2010. Results: Colorectal cancer was the sixth most common cancer in China. It was estimated that there were 274,841 new cases diagnosed in 2010 (157,355 in males and 117,486 in females), with the crude incidence rate of 20.1/100,000, highest in males in urban areas. Age-standardized rates by China standard population of 2000 (ASRcn) and World standard population (Segi's population, ASRwld) for incidence were 16.1/100,000 and 15.9/100,000 respectively. There were 132,110 cases estimated to have died from colorectal cancer in China in 2010 (76,646 men and 55,464 women) with the crude mortality rate of 10.1/100,000. The ASRcn and ASRwld for mortality were 7.55/100,000 and 7.44/100,000 respectively, higher in males and urban areas than in females and rural areas. The incidence and mortality rates increased with age, reaching peaksin the 80-84 year old, and oldest age groups, respectively. Conclusions: Colorectal cancer is one of the most common incident cancers and cause of cancer death in China. Primary and secondary prevention, with attention to a health lifestyle, physical activity and screening should be enhanced in the general population.

        Inter‑turn short‑circuit fault detection with high‑frequency signal injection for inverter‑fed PMSM systems

        This paper develops an online inter-turn short-circuit fault (ISF) detection method for inverter-fed permanent magnet synchronous machine (PMSM) systems. A high-frequency (HF) mathematical model of a PMSM with an ISF is established. Based on the presented HF model, the HF responses of the PMSM with various severity of ISF under HF voltage injectionare analyzed in detail, where both the short-circuit ratio and the fault resistance are considered to evaluate the fault severity. In the proposed fault diagnosis method, the amplitude of the HF negative-sequence component is used to determine the fault severity, and the initial phase is used to locate the position of the faulty phase. The simulation and experimental results verify the effectiveness of the proposed method.

      • Luciferase Assay to Screen Tumour-specific Promoters in Lung Cancer

        Objective: Specific promoters could improve efficiency and ensure the safety of gene therapy. The aim of our study was to screen examples for lung cancer. Methods: The firefly luciferase gene was used as a reporter, and promoters based on serum markers of lung cancer were cloned. The activity and specificity of seven promoters, comprising CEACAM5 (carcinoembryonic antigen, CEA), GRP (Gastrin-Releasing Peptide), KRT19 (cytokeratin 19, KRT), SFTPB (surfactant protein B, SP-B), SERPINB3 (Squamous Cell Carcinoma Antigen, SCCA), SELP (Selectin P, Granule Membrane Protein 140kDa, Antigen CD62, GMP) and DKK1 (Dickkopf-1) promoters were compared in lung cancer cells to obtain cancer-specific examples with strong activity. Results: The CEACAM5, DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB promoters were cloned. Furthermore, we successfully constructed recombinant vector pGL-CEACAM5 (DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB) contained the target gene. After cells were transfectedwith recombinant plasmids, we found that the order of promoter activity from high to low was SERPINB3, DKK1, SFTPB, KRT19, CEACAM5, SELP and GRP and the order for promoters regarding specificity and high potential were SERPINB3, DKK1, SELP, SFTPB, CEACAM5, KRT19 and GRP. Conclusion: The approach adopted is feasible to screen for new tumour specific promoters with biomarkers. In addition, the screened lung-specific promoters might have potential for use in lung cancer targeted gene therapy research.

      • Pu-erh Tea Powder Preventive Effects on Cisplatin-Induced Liver Oxidative Damage in Wistar Rats

        Background: Chemotherapy is one of the major means for control of malignancies, with cisplatin (CDDP) as one of the main agents, widely used for the treatment of various malignant solid tumors. However, prevention of hepatotoxicity from cisplatin is one of the urgent issues in cancer chemotherapy. In this study, we aimed to investigate the effects of pu-erh tea on hepatotoxicity through body weight and tissue antioxidant parameters like, liver coefficient, serum alanine aminotransferase (ALT), serum aspartate aminotransferase (AST), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), malondialdehyde(MDA) and glutathione (GSH) levels, and light microscopic evaluation by histological findings. Materials and Methods: The rats were randomly divided into five groups: Control (n=10), cisplatin (3 mg/kg p.i., n=10), cisplatin+pu-erh (0.32 g/kg/day i.g., n=10), cisplatin+pu-erh (0.8 g/kg/day i.g., n=10) and cisplatin+pu-erh (1.6 g/kg/day i.g., n=10). Pu-erh tea powder was administrated for 31 consecutive days. The rats were sacrificed at the end on the second day after a single dose of cisplatin treatment for measuring indices. Results: Pu-erh tea powder exhibited a protective effect by decreasing MDA and GSH and increasing the SOD and GSH-PX levels and GSH-PX/MDA ratio in camparison with the control group. Besides, pu-erh tea was also able to alleviate the pathological damage to some extent. Conclusion: Pu-erh tea powder is protective against cisplatin-induced liver oxidative damages, especially at the medium dosage (0.8 g/kg/d).
