        Constrained High Accuracy Stereo Reconstruction Method for Surgical Instruments Positioning

        In this paper, a high accuracy stereo reconstruction method for surgery instruments positioning is proposed. Usually, the problem of surgical instruments reconstruction is considered as a basic task in computer vision to estimate the 3-D position of each marker on a surgery instrument from three pairs of image points. However, the existing methods considered the 3-D reconstruction of the points separately thus ignore the structure information. Meanwhile, the errors from light variation, imaging noise and quantization still affect the reconstruction accuracy. This paper proposes a method which takes the structure information of surgical instruments as constraints, and reconstructs the whole markers on one surgical instrument together. Firstly, we calibrate the instruments before navigation to get the structure parameters. The structure parameters consist of markers` number, distances between each markers and a linearity sign of each instrument. Then, the structure constraints are added to stereo reconstruction. Finally, weighted filter is used to reduce the jitter. Experiments conducted on surgery navigation system showed that our method not only improve accuracy effectively but also reduce the jitter of surgical instrument greatly.

        Center of Gravity Position Estimation of Counterbalanced Forklift Truck Based on Multi Model Data Fusion

        The center of gravity of a forklift truck, a crucial parameter for vehicle stability, changes with different loads during operation. We propose an estimation algorithm for the center of gravity position suitable for a counterbalanced forklift truck. By installing sensors on the fork, we use an inclinable platform and propose a static joint center of gravity measurement method. For straight-line driving, we establish a longitudinal dynamics model and propose a nonlinear H∞ estimation algorithm. For steering conditions, we establish a roll dynamics model and propose a forgetting factor recursive least square estimation algorithm. A data fusion algorithm for the forklift truck’s center of gravity position under various working conditions is proposed. The fusion of these estimation results yields the best estimated center of gravity height. We validate the algorithm’s effectiveness using a hardware-in-the-loop simulation platform under different working conditions. The experiments demonstrate the algorithm’s fast parameter fitting, wide applicability, and accurate position control within a 5 % error range.

        SIRT3-mediated mitochondrial unfolded protein response weakens breast cancer sensitivity to cisplatin

        Background Mitochondrial unfolded protein response plays an important role in the occurrence and development of breast cancer. However, the role of mitochondrial unfolded protein response (UPRmt) in the sensitivity of breast cancer to cisplatin chemotherapy has not yet been cleared. Objectives The purpose of this study is to explore the role of mitochondrial unfolded protein response in breast cancer sensitivity to cisplatin. Methods In this study, qRT-PCR, Western blotting, Immunofuorescence, CCK-8, Colony formation, Transwell assay and TUNEL staining assay were used to confrm the role of UPRmt in breast cancer cells treated with cisplatin. Results Cisplatin increased the levels of UPRmt including CLPP, HSP60, LONP1 in MCF7 and MDA-MB-231 cells. UPRmt inducer Nicotinamide ribose (NR) could promote the proliferation and invasion of breast cancer cells treated with cisplatin. Importantly, SIRT3 was discovered to increase UPRmt in breast cancer cells and silencing of SIRT3 could inhibit the efect of NR in breast cancer. Conclusions UPRmt regulated by SIRT3 could protect breast cancer cell from cisplatin. Controlling SIRT3-induced UPR may be a potential therapeutic target to increase the sensitivity of breast cancer chemotherapy.

      • EA-D p45-IgG as a Potential Biomarker for Nasopharyngeal Carcinoma Diagnosis

        Aim: To identify new biomarkers for NPC diagnosis with an anti-EBV Western blot test kit. Methods: Serum samples from 64 NPC patients and healthy subjects with four specific VCA-IgA/EA-IgA profiles were tested with an anti-EBV Western blot test kit from EUROIMMUN AG. Proteins were quantified with scores of intensity visually assigned to the protein bands. The markers which showed statistical differences between the NPC and non-NPC subjects were further evaluated in another 32 NPC patients and 32 controls in comparison with established biomarkers including VCA-IgA, EA-IgA, EBV-related protein IgG, and EBV DNA. Results: Among the markers screened, EA-D p45-IgG showed a statistically significant difference (p < 0.05) between NPC and non-NPC subjects with VCA-IgA positivy. In 32 VCA-IgA positive NPC patients and 32 control subjects, the diagnostic accuracy of EA-D p45-IgG was 78.1% with a positive predictive value of 77.8% and a negative predictive value of 78.6%. In the verification experiment, the specificity and sensitivity of EA-D p45-IgG were 75.0% and 90.6 %, respectively. Conclusions: EA-D p45-IgG might be a potential biomarker for NPC diagnosis, especially among VCA-IgA positive subjects.
