        Manganese dioxide-graphene nanocomposite film modified electrode as a sensitive voltammetric sensor of indomethacin detection

        Excess amount of analgesic and anti-inflammatory drug, such as indomethacin, often leads to serious gastrointestinal complications; therefore, amount of such active compound should be regulated in commercial drugs. This study proposes an efficient analytical technique to detect indomethacin selectively. We prepared and investigated electrochemical properties of a manganese dioxide-graphene nanocomposite film modified glassy carbon electrode (MnO2-Gr/GCE). The behavior of the modified electrode as electrocatalyst towards indomethacin oxidation was also examined. The cyclic voltammetric results reveal that the electrocatalytic activity for the oxidation of indomethacin can significantly be enhanced on the MnO2-Gr/GCE. Indomethacin exhibited a sensitive anodic peak at about 0.90 V at MnO2-Gr/GCE. The data obtained from differential pulse voltammetry showed that the anodic peak currents were linearly dependent on the indomethacin concentrations in the range of 1.0 × 10−7 to 2.5 × 10−5 mol/L with a detection limit of 3.2 × 10−8 mol/L (S/N = 3). Most importantly, the proposed method shows efficient and selective sensing of indomethacin in commercial pharmaceutical formulations. This is the first report of a voltammetric sensor for indomethacin using MnO2-Gr/GCE. We believe that this new method can be commercialized for routine applications in laboratories.

        Inductive expression patterns of genes related to Toll signaling pathway in silkworm (Bombyx mori) upon Beauveria bassiana infection

        Innate immunity is critical to insects and plays an important role in pathogen elimination and wound healing. Toll signaling pathway is the major signaling pathway associated with insect innate immunity mediating synthesis of anti-fungal/bacterial peptides. To better understand the Toll signaling pathwaymediated immune response in Bombyx mori against Beauveria bassiana infection, expression patterns of genes encoding sixteen putative components of Toll signaling pathway in the silkworm larvae challenged with B. bassiana, including four pattern recognition receptors (PRRs, i.e. BmβGRP 1, 2, 3, 4), eight Toll-like receptors (TLRs, i.e. Bm18w, BmToll 1, 3, 6, 9, 7, 10, 11) and four effectors (BmMoricin 1, BmGloverin 2, BmDefensin 1 and BmLysozyme 1), were analyzed using quantitative real-time PCR. At the same time, the changes in their expression by RNAi knock-down of the four PRRswere also detected.Moreover, the effects of Toll signaling pathway inhibitors on antifungal activity in larvae hemolymphwere also analyzed. The results showed that the expression levels of genes encoding sixteen putative components of Toll signaling pathway were obviously altered by the challengewith B. bassiana, but their temporal regulation mode was significantly different. Based on the expression patterns of the genes related to Toll signaling pathway, two sub-paths of immune signal recognition and transduction might be proposed in the response of silkworm larvae against B. bassiana infection. Besides, Toll signaling pathway inhibitor could significantly inhibit the antifungal activity in hemolymph and resulted in increased sensitivity of silkworm larvae to the B. bassiana infection, while the treatment with heat-inactivated B. bassiana could induce antifungal activity in the hemolymph and led to stronger resistance of the silkworm. These results implied that Toll signaling pathway played important roles in the antifungal immune response of the silkworm larvae, in which different components of Toll signaling pathway might play a specific regulatory function. These findings yield insights into the innate immune mechanisms underlying Toll signaling pathway in silkworm.

        Helmet CPAP versus Oxygen Therapy in Hypoxemic Acute Respiratory Failure: A Meta-Analysis of Randomized Controlled Trials

        Purpose: The efficacy of helmet continuous positive airway pressure (CPAP) in hypoxemic acute respiratory failure (hARF) remainsunclear. The aim of this meta-analysis was to critically review studies that investigated the effect of helmet CPAP on gas exchange,mortality, and intubation rate in comparison with standard oxygen therapy. Materials and Methods: We performed a meta-analysis of randomized controlled trials (RCTs) by searching the PubMed, Embase,Cochrane library, OVID, and CBM databases, and the bibliographies of the retrieved articles. Studies that enrolled adults with hARF who were treated with helmet CPAP and measured at least one of the following parameters were included: gas exchange,intubation rate, in-hospital mortality rate. Results: Four studies with 377 subjects met the inclusion criteria and were analyzed. Compared to the standard oxygen therapy, helmet CPAP significantly increased the PaO2/FiO2 [weighted mean difference (WMD)=73.40, 95% confidence interval (95% CI): 43.92 to 102.87, p<0.00001], and decreased the arterial carbon dioxide levels (WMD=-1.92, 95% CI: -3.21 to -0.63, p=0.003), intubationrate [relative risk (RR)=0.21, 95% CI: 0.11 to 0.40, p<0.00001], and in-hospital mortality rate (RR=0.22, 95% CI: 0.09 to 0.50, p=0.0004). Conclusion: The results of this meta-analysis suggest that helmet CPAP improves oxygenation and reduces mortality and intubationrates in hARF. However, the significant clinical and statistical heterogeneity of the literature implies that large RCTs are neededto determine the role of helmet CPAP in different hypoxemic ARF populations.
