        Dynamic expression of early responsible genes to acute left-ventricular ischemia in a time-dependent pattern

        Acute myocardial infarction remains one of the leading causes of death and disability worldwide. Themechanisms underlying myocardial infarction involve a complex of signaling molecules, such as tumornecrosis factor α (TNFα), interleukin-6 (IL6), C-Myc, atria natriuretic peptide (ANP), superoxide dismutase1 (SOD1), and so on. The aim of this study is to understand the time-dependent expressional pattern of theseearly responsible genes following acute myocardial ischemia established by left anterior descending (LAD)coronary artery ligation. After LAD ligation, a collection of genes was detected using real-time polymerasechain reaction (PCR). The expression of inflammation-related genes, such as TNFα and IL6, was immediatelyupregulated at 2 h, reached to the highest point at 12 h, and then decreased to nearly basis level at 24 h afterligation, suggesting inflammation appeared and disappeared rapidly after acute ischemia. C-Myc, an importanttranscription factor, was significantly upregulated at 2 h, and thereafter persisted at high level to 24 h. Thesecretary peptide, ANP, was consistently upregulated from 2 to 24 h, reached to 40-folds at 24 h. The calciumregulatedgene, FK506-binding protein 12.6, was not significantly altered after ischemia. SOD1 was not alteredat the first 4 h, and began to downregulate at 12 and 24 h. These results indicate that several genes weredynamically and transiently regulated after acute myocardial infarction (AMI) in a time-dependent pattern,suggesting that there is an immediate molecular response to acute myocardial ischemia, which might provideus a new insight to understand molecular mechanisms of AMI.

        A Lightweight Three-Party Privacy-preserving Authentication Key Exchange Protocol Using Smart Card

        How to make people keep both the confidentiality of the sensitive data and the privacy of their real identity in communication networks has been a hot topic in recent years. Researchers proposed privacy-preserving authenticated key exchange protocols (PPAKE) to answer this question. However, lots of PPAKE protocols need users to remember long secrets which are inconvenient for them. In this paper we propose a lightweight three-party privacy-preserving authentication key exchange (3PPAKE) protocol using smart card to address the problem. The advantages of the new 3PPAKE protocol are: 1. The only secrets that the users need to remember in the authentication are their short passwords; 2. Both of the users can negotiate a common key and keep their identity privacy, i.e., providing anonymity for both users in the communication; 3. It enjoys better performance in terms of computation cost and security. The security of the scheme is given in the random oracle model. To the best of our knowledge, the new protocol is the first provably secure authentication protocol which provides anonymity for both users in the three-party setting.

        A New Roaming Authentication Framework For Wireless Communication

        Roaming authentication protocol is widely used in wireless network which can enable a seamless service for the mobile users. However, the classical approach requires the home server`s participation during the authentication between the mobile user and the foreign server. So the more the roaming requests are performed the heavier burden will be on the home server. In this paper, we propose a new roaming authentication framework for wireless communication without the home server`s participation. The new roaming authentication protocol in the new framework takes advantage of the ID-based cryptography and provides user anonymity. It has good performance compared with the roaming authentication protocols whose authentication do not need the home server`s participation in terms of security and computation costs. Moreover, a new User-to-User authentication protocol in the new framework is also present. All the authentications proposed in this paper can be regarded as a common construction and can be applied to various kinds of wireless networks such as Cellular Networks, Wireless Mesh Networks and Vehicle Networks.

      • Synthesis and evaluation of arylpiperazine-reverse amides as biased dopamine D3 receptor ligands

        The dopamine D3 receptor (D3R) preferential ligands have been universally adopted as a strategy for the treatment of drug addiction and other neuropsychiatric disorders due to fewer side effects. However, the high sequence homology between D3R and the D2 receptor (D2R) challenges the development of D3R-biased compounds. Herein. we design and synthesize a novel series of reverse amide-piperazine hybrid ligands and evaluate their biological affinities in vitro. Compound 4d was found to be the most potent D3R-selective ligand among these hybrid derivatives. Molecular modeling revealed that extracellular loop 1 (EL1) and loop 2 (EL2) of D3R together likely contribute to D3R selectivity over D2R. In particular. Gly94 in EL1 of D3R may act as a molecular determinant for D3R specificity.

        CD44 Antibody-Conjugated Gold Nanostars as SERS Probes for Distinguishing Cancer Cells (A549 Cells, H1229 Cells) from Normal Cells (ATII Cells)

        In this paper, we report a novel antibody conjugated gold nanostars (AuNSs) as surface-enhanced Raman spectroscopy (SERS) probes for distinguishing cancer cells (A549 cells, H1229 cells) from normal cells (ATII cells). In such a probe, bovine serum albumin (BSA) was served as the protective agent and stabilizing agent, 4-mercaptobenzoic acid (4-MBA) was used as the Raman reporter to generate SERS signals as well as the conjugation agent for attaching CD44 antibody (anti-CD44) molecules to AuNSs, where anti-CD44 could effectively target to CD44 protein overexpressed cells. All the results of the experiments con¯rmed that more SERS probes have been targeted to cancer cells (A549 cells and H1229 cells) than that of normal cells (ATII cells) under the same condition. The anti-CD44 SERS probes hold a potential application in distinguishing cancer cells from normal cells with high sensitivity and good biocompatibility.

        Preliminary numerical study on hydrogen distribution characteristics in the process that flow regime transits from jet to buoyancy plume in time and space

        Hydrogen-steam gas mixture may be injected into containment with flow regime varying both spatiallyand transiently due to wall effect and pressure difference between primary loop and containment insevere accidents induced by loss of coolant accident. Preliminary CFD analysis is conducted to gain informationabout the helium flow regime transition process from jet to buoyancy plume for forthcomingexperimental study. Physical models of impinging jet and wall condensation are validated using separatedeffect experimental data, firstly. Then helium transportation is analyzed with the effect of jetmomentum, buoyancy and wall cooling discussed. Result shows that helium distribution is totallydominated by impinging jet in the beginning, high concentration appears near gas source and wallwhere jet momentum is strong. With the jet weakening, stable light gas layer without recirculating eddyis established by buoyancy. Transient reversed helium distribution appears due to natural convectionresulted from wall cooling, which delays the stratification. It is necessary to concern about hydrogenaccumulation in lower space under the containment external cooling strategy. From the perspective ofexperiment design, measurement point should be set at the height of connecting pipe and near the wallfor stratification stability criterion and impinging jet modelling validation.

        Molecular and cytological analyses of A and C genomes at meiosis in synthetic allotriploid Brassica hybrids (ACC) between B.napus (AACC) and B.oleracea (CC)

        Success of interspecific hybridization relies mostly on the adequate similarity between the implicated genomes to ensure synapsis, pairing and recombination between appropriate chromosomes during meiosis in allopolyploid species. Allotetraploid Brassica napus (AACC) is a model of natural hybridization between Brassica rapa (AA) and Brassica oleracea (CC), which are originally derived from a common ancestor, but genomic constitution of the same chromosomes probably varied among these species through time after establishment, giving rise to cytogenetic difference in the synthetic hybrids. Herein we investigated meiotic behaviors of A and C chromosomes of synthetic allotriploid Brassica hybrids (ACC) at molecular and cytological levels, which result from the interspecific cross between natural B. napus (AACC) and B.oleracea (CC), and the results showed that meiosis course was significantly aberrant in allotriploid Brassica hybrids, and chromosomes aligned chaotically at metaphase I, chromosome bridges and lags were frequently observed from later metaphase I to anaphase II during meiosis. Simultaneously, we also noticed that meiosis-related genes were abruptly down-regulated in allotriploid Brassica hybrids, which likely accounted for irregular scenario of meiosis observed in these synthetic hybrids. Therefore, these results indicated that inter-genomic exchanges of A and C chromosomes could occur frequently in synthetic Brassica hybrids, and provided an efficient approach for genetic changes of homeologous chromosomes during meiosis in polyploid B.napus breeding program.
