        Improved oral bioavailability of capsaicin via liposomal nanoformulation: preparation, in vitro drug release and pharmacokinetics in rats

        This study innovatively prepared an effectivecapsaicin-loaded liposome, a nanoformulation with fewerirritants, for oral administration. The in vitro and in vivoproperties of the liposomal encapsulation were investigatedand the potential possibility of oral administration evaluated. The liposomal agent composed of phospholipid, cholesterol,sodium cholate and isopropyl myristate was prepared usingfilm-dispersion method. A level A in vitro–in vivo correlation(IVIVC) was established for the first time, which demonstratedan excellent IVIVC of both formulated and freecapsaicin in oral administration. Physicochemical characterizationsincluding mean particle size, zeta (f) potentialand average encapsulation efficiency of capsaicin-loadedliposome were found to be 52.2 ± 1.3 nm, -41.5 ±2.71 mv and 81.9 ± 2.43 %, respectively. In vivo, liposomalencapsulation allowed a 3.34-fold increase in relativebioavailability compared to free capsaicin. The gastricmucosa irritation studies indicated that the liposomal systemwas a safe carrier for oral administration. These resultssupport the fact that capsaicin, an effective drug for thetreatment of neuropathic pain, could be encapsulated inliposome for improved oral bioavailability. The excellentIVIVC of capsaicin-loaded liposome could also be a promisingtool in liposomal formulation development with anadded advantage of reduced animal testing.

        중국 체육학 전공대학생의 진로자기조절과 진로탐색행동의 관계

        The purpose of this study was to verify the relationship between career self-regulation and career exploration of Chinese physical education students majoring in physical education. To achieve the purpose of the research, collecting samples from students who are majoring in Physical Education in Shandong province(N=557) with convenience sampling among the non-arbitrary sampling method. Collected data were analyzed by reliability test, exploratory factor analysis and multiple linear regression analysis with IBM SPSS 24.0 statistical program. The results of the study are as follows. First, it was verified that positive career thinking, career feedback, career environment creation, and career commitment willingness influence self-exploration. Second, it was verified that career feedback and career commitment willingness influence job search. Third, it is verified that career feedback affects career opportunity exploration, and the higher the career self-regulation, the more active career exploration activities are, which is an important variable influencing career exploration behavior of physical education college students.

        A Novel Zero-Crossing Compensation Scheme for Fixed Off-Time Controlled High Power Factor AC-DC LED Drivers

        A fixed off-time controlled high power factor ac-dc LED driver is proposed in this paper, which employs a novel zero-crossing-compensation (ZCC) circuit based on a fixed off-time controlled scheme. Due to the parasitic parameters of the system, the practical waveforms have a dead region. By detecting the zero-crossing boundary, the proposed ZCC circuit compensates the control signal VCOMP within the dead region, and is invalid above this region. With further optimization of the parameters KR and Kτ of the ZCC circuit, the dead zone can be eliminated and lower THD is achieved. Finally, the chip is implemented in HHNEC 0.5㎛ 5V/40V HVCMOS process, and a prototype circuit, delivering 7~12W of power to several 3-W LED loads, is tested under AC input voltage ranging from 85V to 265V. The test results indicate that the average total harmonic distortion (THD) of the entire system is approximately 10%, with a minimum of 5.5%, and that the power factor is above 0.955, with a maximum of 0.999.

        Tensile Behavior of High-Strength Stainless Steel Wire Rope (HSSSWR)-Reinforced ECC

        Engineered cementitious composites (ECC) show the distinguished characteristics of high post-cracking resistance and ductility. High-strength stainless steel wire rope (HSSSWR) has been successfully used for restoring or strengthening of existing structures. By combining the advantages of these two materials, a new composite system formed by embedding HSSSWR into ECC was proposed and expected to be a promising engineering material for repair or strengthening of structures. To investigate the tensile failure mechanism and mechanical properties of HSSSWR-reinforced ECC, an experimental study on 27 HSSSWR-reinforced ECC plates was conducted considering the effects of the reinforcement ratio of longitudinal HSSSWRs, formula of ECC and width of the plate. Test results revealed that HSSSWR-reinforced ECC exhibit superior post-cracking resistance, deformation capacity and crack-width control capacity. Increasing the reinforcement ratio of longitudinal HSSSWRs can effectively enhance the tensile strength, crack-width control capacity, deformation capacity and tensile toughness of HSSSWR-reinforced ECC. Adding thickener in ECC can significantly improve the crack-width control capacity and deformation capacity of HSSSWR-reinforced ECC due to enhancing uniform distribution of polyvinyl alcohol fibers, but would slightly reduce the cracking stress and maximum tensile stress by bringing small bubbles in the matrix. The tensile properties of HSSSWR-reinforced ECC plates are almost not affected by varying the plate width. Besides, a tensile constitutive model was developed for charactering the stress-strain relationship of HSSSWR-reinforced ECC in tension. Based on mechanical theories and failure characteristics of HSSSWR-reinforced ECC, the model parameters were determined, and calculation equations of cracking stress and tensile strength were proposed. The accuracy of the developed model and calculation equations was verified by test results.

        Ethanol enhances cucurbitacin B-induced apoptosis by inhibiting cucurbitacin B-induced autophagy in LO2 hepatocytes

        Ethanol is a common risk factor for liver injury. Cucurbitacin B (CuB) is a natural product with potent cytotoxic activities mediated by inducing apoptosis. This study investigated the effect of ethanol on CuB-induced cytotoxicity in LO2 hepatocytes. Low concentration of ethanol alone showed no significant cytotoxic effect on LO2 cells. CuB dose-dependently decreased cell viability. However, ethanol co-treatment significantly enhanced CuB-induced cytotoxicity. CuB-induced mitochondria membrane potential (ΔΨ) depolarization was further decreased by ethanol. Furthermore, CuB-induced apoptosis was augmented by ethanol, as evidenced by DNA fragmentation, Annexin V staining, and apoptotic protein expression. Ethanol inhibited CuB-induced autophagy, as determined by MDC staining, autophagic protein expression, and transmission electron microscopy. Therefore, the present data suggested that ethanol enhanced CuB cytotoxicity in LO2 hepatocytes, which was mediated by inhibiting autophagy and augmenting apoptosis.
