      • ADPRT Val762Ala and XRCC1 Arg194Trp Polymorphisms and Risk of Gastric Cancer in Sichuan of China

        Objective: Gastric cancer remains a major health problem in China. We hypothesized that XRCC1 Arg194Trp and ADPRT Val762Ala may be associated with risk. Methods: We designed a multicenter 1:1 matched case-control study of 307 pairs of gastric cancers and controls between October 2010 and August 2011. XRCC1 Arg194Trp and ADPRT Val762Ala were sequenced, and demographic data as well as lifestyle factors were collected using a self-designed questionnaire. Results: Individuals carrying XRCC1 Trp/Trp or Arg/Trp variant genotype had a significantly increased risk of gastric cancer (OR, 1.718; 95% CI, 1.190-2.479), while the OR for ADPRT Val762Ala variant genotype (Ala/Ala or Val/Ala) was 1.175 (95% CI, 0.796-1.737). No gene-gene or gene-environment interactions were found. In addition, family history of cancer and drinkers proportion were higher among cases than among controls (P<0.05). Conclusions: XRCC1 194 Arg/Trp or Trp/Trp genotype, family history of cancer, and drinking are suspected risk factors of gastric cancer from our study. Our findings may offer insight into further similar large gene-environment and gene-gene studies in this region.

      • Silencing of COX-2 by RNAi Modulates Epithelial-Mesenchymal Transition in Breast Cancer Cells Partially Dependent on the PGE<sub>2</sub> Cascade

        In order to prove whether downregulation of COX-2 (Cyclooxygenase-2) could modulate the epithelial-mesenchymal transition (EMT) of breast cancer, celecoxib and siRNA were respectively used to inhibit COX-2 function and expression in MDA-MB-231 cells. The EMT reversal effect in the RNAi treated group was better than that of the celecoxib group while there were no obvious differences in the medium $PGE_2$ levels between the two groups. The results show that COX-2 pathways may contribute considerably to EMT of breast cancer cells, partially dependent on the PGE2 cascade. Akt2, ZEB2 and Snail were measured to clarify the underlying mechanisms of COX-2 on EMT; COX-2 may modulate EMT of breast cancer by regulating these factors. This finding may be helpful to elucidate the mechanisms of selective COX-2 inhibitor action in EMT modulation in breast cancer.

        Meeting a Learning Century: Formation and Modernization Of Lifelong Learning Cities in China

        The purpose of the study was to examinee how lifelong learning cities are developing in China. To accomplish this goal, the study first reviewed theoretical framework and practical achievements in other countries to anchor China in an international context. Since Shanghai first declared to ‘build Shanghai into a lifelong learning city to meet the challenges of the new era’in September, 1999, many cities followed the suit. Until now, by a rough estimate, there were altogether 61 lifelong learning cities in China, which occupied 10% of the total number of cities. China’s experiences to build lifelong learning cities showed distinctive features from other countries on the sense of purposes, ways of promotion and other aspects. Meanwhile, some problems were identified and needed to be solved. Therefore, the study provided several suggestions for future development in the end. 중국 평생학습도시의 형성과 현대화최은수, 여문연본 논문은 우선 평생학습도시의 이론적 발전 및 몇 개 대표적 도시에서의 실제 추진 상황을 요약하고자 한다. 중국의 평생학습도시건설의 이론 및 실제를 국제 궤도에 접목시키는데 그 목적을 두고 있다. 1999년 9월, 중국 상해시가 선두적으로 "학습형도시를 건설하여 새로운 세기의 도전에 응하자" 라는 구호를 제기하자 기타 도시들에서도 뒤질세라 상응한 목표를 제기하였다. 불완전한 통계에 따르면 현재 중국에는 61개에 달하는 학습형도시가 건설되었으며 이는 전국 도시 총수의 10%에 달한다. 중국은 학습형 도시건설의 목적, 추진 방식 및 기타 방면에서 자체의 특징을 나타내고 있는 동시에 일련의 문제점과 부족한 점이 존재하고 있다. 따라서 본 논문의 결론부분에서 몇 가지 건설적인 의견과 미래의 발전 방향에 대해 제시하였다. 핵심되는 말 : 학습형 도시, 학습형 사회구역, 정책 및 법률, 상해

        Positive Association of TEAD1 With Schizophrenia in a Northeast Chinese Han Population

        Objective Schizophrenia is a complex and devastating psychiatric disorder with a strong genetic background. However, much uncertainty still exists about the role of genetic susceptibility in the pathophysiology of schizophrenia. TEA domain transcription factor 1 (TEAD1) is a transcription factor associated with neurodevelopment and has modulating effects on various nervous system diseases. In the current study, we performed a case–control association study in a Northeast Chinese Han population to explore the characteristics of pathogenic <i>TEAD1</i> polymorphisms and potential association with schizophrenia.Methods We recruited a total of 721 schizophrenia patients and 1,195 healthy controls in this study. The 9 single nucleotide polymorphisms (SNPs) in the gene region of <i>TEAD1</i> were selected and genotyped.Results The genetic association analyses showed that five SNPs (rs12289262, rs6485989, rs4415740, rs7113256, and rs1866709) were significantly different between schizophrenia patients and healthy controls in allele or/and genotype frequencies. After Bonferroni correction, the association of three SNPs (rs4415740, rs7113256, and rs1866709) with schizophrenia were still evident. Haplotype analysis revealed that two strong linkage disequilibrium blocks (rs6485989-rs4415740-rs7113256 and rs16911710-rs12364619-rs1866709) were globally associated with schizophrenia. Four haplotypes (C-C-C and T-T-T, rs6485989-rs4415740-rs7113256; G-T-A and G-T-G, rs16911710-rs12364619-rs1866709) were significantly different between schizophrenia patients and healthy controls.Conclusion The current findings indicated that the human <i>TEAD1</i> gene has a genetic association with schizophrenia in the Chinese Han population and may act as a susceptibility gene for schizophrenia.
