        Combinatorial nanococktails via selfassembling lipid prodrugs for synergistically overcoming drug resistance and effective cancer therapy

        Background: Circular RNAs (circRNAs) have important functions in many fields of cancer biology. In particular, we previously reported that the oncogenic circRNA, circPRMT5, has a major role in bladder cancer progression. Therapy based on circRNAs have good prospects as anticancer strategies. While anti-circRNAs are emerging as therapeutics, the specific in vivo delivery of anti-circRNAs into cancer cells has not been reported and remains challenging. Methods: Synthesized chrysotile nanotubes (SCNTs) with a relatively uniform length (~ 200 nm) have been designed to deliver an siRNA against the oncogenic circPRMT5 (si-circPRMT5) inhibit circPRMT5. In addition, the antitumor effects and safety evaluation of SCNTs/si-circPRMT5 was assessed with a series of in vitro and in vivo assays. Results: The results showed that SCNTs/si-circPRMT5 nanomaterials prolong si-circPRMT5’s half-life in circulation, enhance its specific uptake by tumor cells, and maximize the silencing efficiency of circPRMT5. In vitro, SCNTs encapsulating si-circPRMT5 could inhibit bladder cancer cell growth and progression. In vivo, SCNTs/si-circPRMT5 inhibited growth and metastasis in three bladder tumor models (a subcutaneous model, a tail vein injection lung metastatic model, and an in situ model) without obvious toxicities. Mechanistic study showed that SCNTs/sicircPRMT5 regulated the miR-30c/SNAIL1/E-cadherin axis, inhibiting bladder cancer growth and progression. Conclusion: The results highlight the potential therapeutic utility of SCNTs/si-circPRMT5 to deliver si-circPRMT5 to treat bladder cancer.       • Application Study of Fuzzy Reasoning Method for Domain Ontology

        For the representation and reasoning of fuzzy knowledge for domain ontology, the paper presents the reasoning method based on fuzzy ontology. On a basis of the study of civil aviation emergency domain ontology, it gives fuzzy extension model of domain ontology based on fuzzy description logic from the perspectives of fuzzy modifiers and fuzzy concrete domains, and designs fuzzy rules by introducing weight concept and gives the representation and construction process of fuzzy rules on a basis of f-SWRL in connection with the inference application of situation analysis for civil aviation events. The experimental results show that the fuzzy extension method of domain ontology can make up for the issue that the existing domain ontology is inadequate in terms of fuzzy knowledge representation, and provide a good methodological support for making domain ontology perfect and inference application with the reasoning implementation based on fuzzy rules.

        Density measurement and equal density temperature of CO2+brine from Dagang - formation from 313 to 363 K

        Densities of CO2+Dagang - formation brine solution were measured by a magnetic suspension balance(MSB) in the pressure range from (10 to 18) MPa, at the temperatures from (313.15 to 363.15) K and CO2 mass frac-tions at 0, 0.0101, 0.0198 and 0.0299. The experimental results revealed that the solution densities increased linearlywith the increasing pressure and CO2 concentration, while decreasing with the increasing temperatures in the experi-mental range. When the temperature increased from (313.15 to 363.15) K, the slopes of the densities versus (vs.) CO2mass fractions decreased from (0.193 to 0.106) g·cm−3. A correlation equation was developed based on thermody-namic theory and experimental data. The absolute average deviation between the correlation equation and the experi-mental data was 0.05%, and the maximum deviation was 0.37% for the density of CO2+water/brine solution in com-mon geological storage conditions. According to the density of CO2 - free brine and apparent molar volume of CO2 inbrine, the equal density temperature (Te) of CO2+Dagang brine solution was obtained at 464.67 K when pressure is10MPa, which means that the density of brine dissolved with CO2 will be less than that of CO2-free brine when thetemperature is higher than 464.67 K at 10MPa. In this work the formation temperature of the Dagang oilfield reser-voir is from 313.15 K to 363.15 K, which is lower than the equal density temperature. Therefore, the safety of CO2 stor-age in Dagang oilfield reservoir can be guaranteed.

        Biosynthesis of Three Chalcone β-D-glucosides by Glycosyltransferase from Bacillus subtilis ATCC 6633

        Chalcones exhibit multiple biological activities. Various studies have attempted to modify the structure of chalcones with a special focus on the addition of substituents to the benzene rings. However, these chemical modifications did not improve the water solubility and bioavailability of chalcones. Glycosylation can markedly affect the physical and chemical properties of hydrophobic compounds. Here, we evaluated the ability of a highly promiscuous glycosyltransferase (GT) BsGT1 from Bacillus subtilis ATCC 6633 to biosynthesize chalcone glucosides. Purified BsGT1 catalyzed the conversion of 4'-hydroxychalcone (compound 1), 4'-hydroxy-4-methylchalcone (compound 2), and 4-hydroxy-4'-methoxychalcone (compound 3), into chalcone 4'-O-β-D-glucoside (compound 1a), 4-methylchalcone 4'-O-β-D-glucoside (compound 2a), and 4'- methoxychalcone 4-O-β-D-glucoside (compound 3a), respectively. To avoid the addition of expensive uridine diphosphate glucose (UDP-Glc), a whole-cell biotransformation system was employed to provide a natural intracellular environment for in situ co-factor regeneration. The yields of compounds 1a, 2a, and 3a were as high as 90.38%, 100% and 74.79%, respectively. The successful co-expression of BsGT1 with phosphoglucomutase (PGM) and UDP-Glc pyrophosphorylase (GalU), which are involved in the biosynthetic pathway of UDP-Glc, further improved the conversion rates of chalcones (the yields of compounds 1a and 3a increased by approximately 10%). In conclusion, we demonstrated an effective whole-cell biocatalytic system for the enzymatic biosynthesis of chalcone β-D-glucoside derivatives.

        Concealment of iris features based on artificial noises

        Although iris recognition verification is considered to be the safest method of biometric verification, studies have shown that iris features may be illegally used. To protect iris features and further improve the security of iris recognition and verification, this study applies the Gaussian and Laplacian mechanisms and to hide iris features by differentiating privacy. The efficiency of the algorithm and evaluation of the image quality by the image hashing algorithm are selected as indicators to evaluate these mechanisms. The experimental results indicate that the security of an iris image can be significantly improved using differential privacy protection.
