Doubled haploid breeding using anther culture is potentially acclaimed for early fixation of homozygosity. We recovered 129 androgenic doubled haploid lines (DHLs) from a cross Khandagiri (drought sensitive) x Dular (drought tolerant). Fifty five DHLs with high seedling vigour showed wide variability for traits associated with drought tolerance and seed yield under drought stress. DHL 1, DHL 4, DHL 8, DHL 24, DHL 30, DHL 41, DHL 43, DHL 48 and DHL 53 showed higher degree of drought tolerance and also revealed the drought tolerant allele (126 bp) amplified by SSR marker RM 8085. DHL41, DHL 53, DHL43, DHL4 and DHL24 had high yield potential than Sahbhagidhan. However, DHL 41 being a short duration (90 days) and semi-dwarf (101.9 cm) with significantly higher number of grains/panicle, very high 1000-grain weight (28.9 g), fertility percentage (94.2%) and seed yield (46.6 q/ha); can be fitted to upland condition. Besides, the DHLs can serve as an ideal mapping population for mapping and detection of QTLs/genes of interest in upland rice.

Use of human patient simulator for apnea studies: a preliminary in vitro trial

Background: Modern human patient simulators (HPSs) could be used for researching critical scenarios such as apnea oxygenation. We aimed to study the use of a high-fidelity HPS to assess prolonged apnea using various oxygenation strategies with a simple high-flow nasal cannula (15 L/min). Methods: An experimental simulation study using an HPS (CAE Healthcare™) was conducted after obtaining approval from the Institutional Review Board. The HPS responded according to real-time physiologically modeled responses to external gases, such as oxygen (O2). Apnea experiments were performed with different physiological settings, such as shunt fraction (5%) and O2 consumption (250, 500, and 750 ml/min). The following four apnea experiments were conducted: no oxygenation (NO), apnea oxygenation alone (AO), preoxygenation alone (PO), and para-oxygenation (PAO). The time to 92%, 75%, and 50% saturation was recorded. Alveolar and arterial gas levels were recorded till 50% saturation. Results: At 250 ml/min, PO (1121 s) and PAO (1274.5 s) had a significantly longer time to 50% saturation (400% increase) compared to NO (222.5 s) and AO (239 s). A similar trend was observed for the time to 92% and 75% saturation. At higher O2 consumption rates, a shorter time to desaturation was observed. Conclusions: Apnea trends in the HPS correlated with similar prior human experiments. AO without preoxygenation was found to provide no additional benefit. Preoxygenation with high-flow O2 via nasal cannula prolonged the time to desaturation in the PAO more than PO scenario. Therefore, HPSs can be used in future studies where patient safety is a concern.

Mesoporous ZnO nanoclusters as an ultra-active photocatalyst

Morphology modulation of nanostructured materials are highly crucial for various applications including photocatalysis, drug delivery, etc. In this study, mesoporous zinc oxide (ZnO) nanoclusters (MZN) were synthesized via a simple, cost-effective, low-temperature wet chemical route and further sucessfully utilized for photodegradation of Rhodamine B (RhB). Firstly, polystyrene (PS) nanospheres (~300nm) were prepared by polymerization of styrene in aqueous solvent. Then the MZN were achieved by formation of ZnO nanoparticles-shell over the surface of PS nanospheres via esterification of zinc acetate dihydrate in isopropyl alcohol followed by toluene dissolution of PS core. The as-synthesized MZN were spherically-shaped, porous in nature with a diameter of ~ 400nm and composed of well-arranged highly-crystalline ZnO nanoparticles (~ 5nm). The MZN also exhibited a high surface area of 78.3+/-5.4m<SUP>2</SUP>g<SUP>-1</SUP> and an average pore diameter of ~26nm. Furthermore, this unique structure demonstrates an expeditious photodegradation of RhB under UV illumination, monitored by UV-visible spectroscopy at different time intervals until the dye was completely degraded to colorless end products. Fast RhB decomposition was observed with a degradation rate of ~98% within initial 40min which can be attributed to the porous nature, large specific surface area and excellent electron accepting features of the engineered nanoclusters.

<P>Six mononuclear Ir complexes (1-6) using polypyridyl-pyrazine based ligands (L-1 and L-2) and {[cp*IrCl-(mu-Cl)](2) and [(ppy)(2)Ir(mu-Cl)](2)} precursors have been synthesised and characterised. Complexes 1-5 have shown potent anticancer activity against various human cancer cell lines (MCF-7, LNCap, Ishikawa, DU145, PC3 and SKOV3) while complex 6 is found to be inactive. Flow cytometry studies have established that cellular accumulation of the complexes lies in the order 2 > 1 > 5 > 4 > 3 > 6 which is in accordance with their observed cytotoxicity. No changes in the expression of the proteins like PARP, caspase 9 and beclin-1, Atg12 discard apoptosis and autophagy, respectively. Overexpression of CHOP, activation of MAPKs (P38, JNK, and ERK) and massive cytoplasmic vacuolisation collectively suggest a paraptotic mode of cell death induced by proteasomal dysfunction as well as endoplasmic reticulum and mitochondrial stress. An intimate relationship between p53, ROS production and extent of cell death has also been established using p53 wild, null and mutant type cancer cells.</P>
