        <i>Dendropanax morbifera</i> Prevents Cardiomyocyte Hypertrophy by Inhibiting the Sp1/GATA4 Pathway

        <P>An extract of <I>Dendropanax morbifera</I> branch exerts antioxidant, anti-inflammatory, antithrombotic, and anticancer activities. The purpose of this study was to investigate the effect of the extract in isoproterenol-induced cardiac hypertrophy. Phalloidin staining showed that treatment with the extract dramatically prevents isoproterenol-induced H9c2 cell enlargement and the expression of cardiac hypertrophic marker genes, including atrial natriuretic peptide (ANP) and B-type brain natriuretic peptide (BNP). Further, pretreatment with the extract decreased isoproterenol-induced GATA4 and Sp1 expression in H9c2 cells. Overexpression of Sp1 induced the expression of GATA4. The forced expression of Sp1 or its downstream target GATA4, as well as the co-transfection of Sp1 and GATA4 increased the expression of ANP, which was decreased by treatment with the extract. To further elucidate the regulation of the Sp1/GATA4-mediated expression of ANP, knockdown experiments were performed. Transfection with small interfering RNAs (siRNAs) for Sp1 or GATA4 decreased ANP expression. The extract did not further inhibit the expression of ANP reduced by the transfection of GATA4 siRNA. Sp1 knockdown did not affect the expression of ANP that was induced by the overexpression of GATA4; however, GATA4 knockdown abolished the expression of ANP that had been induced by Sp1 overexpression. The extract treatment also attenuated the isoproterenol-induced activation of p38 MAPK, ERK1/2, and JNK1. Hesperidin, catechin, 2,5-dihydroxybenzoic acid, and salicylic acid are the main phenolic compounds present in the extract as observed by high performance liquid chromatography. Hesperidin and 2,5-dihydroxybenzoic acid attenuated isoproterenol-induced cardiac hypertrophy. These findings suggest that the <I>D. morbifera</I> branch extract prevents cardiac hypertrophy by downregulating the activation of Sp1/GATA4 and MAPK signaling pathways.</P>

        Hepatotoxic mechanism of diclofenac sodium on broiler chicken revealed by iTRAQ-based proteomics analysis

        Background: At the therapeutic doses, diclofenac sodium (DFS) has few toxic side effects on mammals. On the other hand, DFS exhibits potent toxicity against birds and the mechanisms remain ambiguous. Objectives: This paper was designed to probe the toxicity of DFS exposure on the hepatic proteome of broiler chickens. Methods: Twenty 30-day-old broiler chickens were randomized evenly into two groups (n = 10). DFS was administered orally at 10mg/kg body weight in group A, while the chickens in group B were perfused with saline as a control. Histopathological observations, serum biochemical examinations, and quantitative real-time polymerase chain reaction were performed to assess the liver injury induced by DFS. Proteomics analysis of the liver samples was conducted using isobaric tags for relative and absolute quantification (iTRAQ) technology. Results: Ultimately, 201 differentially expressed proteins (DEPs) were obtained, of which 47 were up regulated, and 154 were down regulated. The Gene Ontology classification and Kyoto Encyclopedia of Genes and Genomes pathway analysis were conducted to screen target DEPs associated with DFS hepatotoxicity. The regulatory relationships between DEPs and signaling pathways were embodied via a protein-protein interaction network. The results showed that the DEPs enriched in multiple pathways, which might be related to the hepatotoxicity of DFS, were "protein processing in endoplasmic reticulum," "retinol metabolism," and "glycine, serine, and threonine metabolism." Conclusions: The hepatotoxicity of DFS on broiler chickens might be achieved by inducing the apoptosis of hepatocytes and affecting the metabolism of retinol and purine. The present study could provide molecular insights into the hepatotoxicity of DFS on broiler chickens.

      • Spatial correlation of aerodynamic forces on 5:1 rectangular cylinder in different VIV stages

        To better understand the vortex-induced vibration (VIV) characteristics of a 5:1 rectangular cylinder, the distribution of aerodynamic force and the non-dimensional power spectral density (PSD) of fluctuating pressure on the side surface were studied in different VIV development stages, and their differences in the stationary state and vibration stages were analyzed. The spanwise and streamwise correlations of surface pressures were studied, and the flow field structure partitions on the side surface were defined based on the streamwise correlation analysis. The results show that the variation tendencies of mean and root mean square (RMS) pressure coefficients are similar in different VIV development stages. The RMS values during amplitude growth are larger than those at peak amplitude, and the smallest RMS values are observed in the stationary state. The spanwise correlation coefficients of aerodynamic lifts increase with increase of the peak amplitude. However, for the lock-in region, the maximum spanwise correlation coefficient for aerodynamic lifts occurs in the VIV rising stage rather than in the peak amplitude stage, probably due to the interaction of vortex shedding force (VSF) and self-excited force (SEF). The streamwise correlation results show that the demarcation point positions between the recirculation region and the main vortex region remain almost constant in different VIV development stages, and the reattachment points gradually move to the tailing edge with increasing amplitude. This study provides a reference to estimate the demarcation point and reattachment point positions through streamwise correlation and phase angle analysis from wind tunnel tests.

      • Smoking, <i>Helicobacter Pylori</i> Serology, and Gastric Cancer Risk in Prospective Studies from China, Japan, and Korea

        <P>Smoking is an established risk factor for gastric cancer development. In this study, we aimed to assess prospectively the association of smoking with gastric cancer risk in 1,446 non-cardia gastric cancer cases and 1,796 controls from China, Japan, and Korea with consideration of <I>Helicobacter pylori</I> infection as a potential effect modifier. Applying logistic regression models stratified by study and adjusted for age and sex we found that current, but not former, smoking was significantly associated with gastric cancer risk [OR = 1.33; 95% confidence interval (CI), 1.07–1.65]. However, the association was significant only in <I>H. pylori</I> sero-positive individuals determined by 3 different sero-markers: overall sero-positivity, sero-positivity to the onco-protein CagA, and sero-positivity to the gastric cancer associated sero-marker HP0305 and HP1564. Specifically, a significant interaction was found when stratifying by HP0305/HP1564 (<I>P</I><SUB>interaction</SUB> = 0.01) with a 46% increased risk of gastric cancer among HP0305/HP1564 sero-positive current smokers (95% CI, 1.10–1.93) as opposed to no increased gastric cancer risk among HP0305/HP1564 sero-negative current smokers (OR = 0.93; 95% CI, 0.65–1.33). We confirmed that current smoking is associated with an increased gastric cancer risk, however, only among individuals that are simultaneously sero-positive for the leading causal factor for gastric cancer, <I>H. pylori</I>.</P>

        Police-Public Partnership among Chinese Communities during COVID-19

        COVID-19으로 인하여 사회 거의 모든 부문에 큰 영향과 많은 변화가 발생했다. COVID-19 대응을 위한 일선 경찰은 평상시와 다르게 방역 관련 업무들을 수행하게 되고, 많은 새로운 문제와 상황을 직면하게 된 것이 사실이다. 지역사회 경찰활동에서 나타난 새로운 도전을 대처하기 위하여 중국 경찰은 풍교경험(楓橋經驗)이라는 지역사회 경찰황동 전략을 새롭게 발전시키고 실천함으로 공중과의 보다 긴밀한 파트너십을 적극적으로 구축하였다. COVID-19 대응을 위한 지역사회 경찰의 노력은 사회질서 유지, 방역활동 지원, 지역사회 전염병 전파 통제에 매우 중요한 역할을 하였다. 본 연구는 중국 COVID-19 대응을 위한 경찰-공중 파트너십을 구축하는 노력과 실천을 주목하여 발전하고 전염병의 위협이 지속되는 상황에서 지역사회 경찰활동의 새로운 전략을 모색하고자 한다. 세계적인 문제인 COVID-19을 극복하고 이기기 위해 중국경찰의 노력과 대처방안을 세계 다른 국가와 나누고자 한다. In view of the policing practice during COVID-19, the efforts against such an epidemic have imposed new requirements on the construction of policing among communities. The Experience of Fengqiao is the basic direction of Chinese police when combat the risk of COVID-19. Chinese police aimed at building a harmonious partnership between the people and community society continuously. The innovative development of community-based policing played a vital role in the epidemic control and social order maintenance. In order to help police in various countries to combat the COVID-19 all over the world, this study focused on the specific practices of Chinese police and public in combating the epidemic, which especially emphasized the core value of Chinese police called people-centered policing is a good attempt to promote the modernization of social governance at the grassroots level.
