        This study was conducted to investigate the physico-chemical characteristics of various bone extracts in Hanwoo cattle. The experimental materials are as follows; T1(Shank bone extract), T2(Tail extract), T3(Beef feet extract), and T4(The other bones extract). In the proximate composition, the T1 and T2 showed higher moisture and fat contents than those of T3 and T4(p<0.05), and the T3 had the highest protein content among the bone extracts(p<0.05). The quality properties including salinity, sugar content, turbidity and yellowness(b*) values were significantly higher in the T3 than the other bone extracts(p<0.05). However, the pH value of T3 was significantly lower than the other bone extracts. In the mineral contents, calcium contents of bone extracts were 36.33-48.57mg/kg, and the content of phosphorus was the lowest in T1(4.62mg/kg) and T2(19.88mg/kg) was the highest(p<0.05). Contents of sodium and potassium were the highest in T3(215.83mg/kg) and T2(54.59mg/kg), while the content of magnesium was the lowest in T3(12.28mg/kg). In conclusion, these results could be suggested to be the basis information for processed meat extract products industry in korea using Hanwoo bone extract.

        Programmed cell death ligand 1 alleviates psoriatic inflammation by suppressing IL-17A production from programmed cell death 1-high T cells

        <P>Background: Psoriasis is one of the most common chronic inflammatory diseases of the skin. Recently, IL-17-producing T cells have been shown to play a critical role in psoriatic inflammation. Programmed cell death 1 (PD-1) is a coinhibitory receptor expressed on T cells in various chronic inflammatory diseases; however, the expression and function of PD-1 during psoriatic inflammation have not previously been characterized. Objective: We examined PD-1 expression on IL-17A-producing T cells from imiquimod-treated mice and patients with psoriasis. Additionally, we investigated the therapeutic effect of recombinant programmed cell death ligand 1 (PD-L1) protein on imiquimod-induced psoriatic inflammation. Methods: PD-1 expression on IL-17A-producing gamma delta T cells from imiquimod-treated mice was examined by means of multicolor flow cytometric analysis. In the psoriatic skin of patients, PD-1 and IL-17A expression was analyzed by using immunofluorescence. The therapeutic effect of PD-L1-Fc fusion protein (PD-L1-Fc) was assessed in imiquimod-treated mice ex vivo and in vivo. Results: During imiquimod-induced psoriatic inflammation, PD-1 is overexpressed on CD27(-)V gamma 1(-) gamma delta T cells. Furthermore, PD-1 expression on IL-17A(+) T cells was confirmed in psoriatic skin tissues from patients and imiquimod-treated mice. In the CD27(-)V gamma 1(-) gamma delta T-cell population, V gamma 4(-) gamma delta T cells with V gamma 6 mRNA expression showed a high level of PD-1 expression. Furthermore, these PD-1(hi)V gamma 4(-)(V gamma 6(+)) gamma delta Tcells were specialized for anti-CD3-induced IL-17A production, which was inhibited by PD-L1-Fc treatment. In imiquimod-treated mice PD-L1-Fc reduced psoriatic inflammation when given alone and enhanced the therapeutic effect of anti-p40 when given in combination. Conclusion: PD-1 is overexpressed in IL-17A-producing T cells in both imiquimod-treated mice and patients with psoriasis. Moreover, recombinant PD-L1-Fc alleviates psoriatic inflammation in imiquimod-treated mice.</P>

        Association of tumor necrosis factor- gene polymorphisms with advanced stage endometriosis

        <P>BACKGROUND: This study was performed to investigate whether specific haplotypes and several single nucleotide polymorphisms in the promoter region of the tumor necrosis factor (TNF)-alpha gene are associated with the risk of advanced stage endometriosis in a Korean population. METHODS: This study comprised women with (n = 246) or without (n = 248) endometriosis. The TNF:g.[-1031T > C], TNF:g.[-863C > A] and TNF:g.[-857C > T] polymorphism in the TNF-alpha gene were assessed by PCR-restriction fragment length polymorphism analysis, which utilized digestion by BbsI, HypCH4IV and HypCH4IV restriction enzymes, respectively. In silico haplotypes were deduced by using the Haploview version 3.32. RESULTS: The genotype distribution of TNF:g.[-1031T > C] was significantly different between total endometriosis patients and the controls (T/T of 56.9 versus 60.1%, T/C of 35.4 versus 37.5% and C/C of 7.7 versus 2.4%, respectively, P = 0.027). This difference at the TNF:g.[-1031T > C] tends to increase in Stage IV endometriosis (P = 0.01). However, there was no difference in the TNF:g.[-863C > A] and TNF:g.[-857C > T] site between the two groups. Even when the endometriosis cases were subdivided into American Society for Reproductive Medicine Stages III and IV, genotype differences were not found. The CC homozygote at TNF:g.-863 was more frequently found in the controls than Non-CC group (P = 0.04; odds ratio = 0.67; 95% confidence interval = 0.45-0.98). All haplotypes and diplotypes, deduced by in silico analysis, showed no association with subgroups or controls. CONCLUSIONS: Our results suggest that the genotype frequencies at the TNF:g.[-1031T > C] and the TNF:g.[-863C > A] sites may be associated with advanced stage endometriosis in the Korean population.</P>

      • 가열시간이 한우갈비 추출액의 품질특성에 미치는 영향

        본 연구는 갈비추출액을 제조할 때 적절한 추출시간을 알아보기 위하여 진행하였으며, 추출시간은 1시간, 6시간, 10시간, 24시간, 48시간으로 하였다. 갈비추출액은 추출시간이 증가할수록 수분은 감소하였고, 단백질과 회분은 증가하는 경향을 나타내었다. 또한 갈비추출액은 염도, 당도 및 색차값과 같은 품질특성에서도 추출시간이 증가할수록 증가하는 경향을 나타내었다. 관능평가에서 갈비추출액의 맛과 전체기호도가 추출 10시간부터 높은 수준을 나타내는 것으로 보아 한우갈비 추출액은 10시간 이상 가열이 품질에 우수한 것으로 판단된다. This study was carried out to determine proper extraction time of rib bone extract in Hanwoo. Experimental design was devided into 5 treatments (T1: 1h, T2: 6h, T3: 10h, T4: 24h and T5: 48h). As the extraction time increased, the moisture content was reduced whereas the protein, fat, and ash were increased (p<0.05). Also, quality characteristics including salinity, sugar content, and color were improved by the extraction time increase. In the sensory characteristics, T4 and T5 showed higher all sensory scores than T1, T2 and T3. Therefore, the extraction time of Hanwoo rib bone is appropriate for more than 10 hours.

      • 국내 유통중인 기계발골계육의 품질특성 및 저장특성 비교

        This study was undertaken to compare the quality characteristics and storage characteristics between mechanically deboned chicken meat (MDCM) in 5 different companies(T1-T5) in Korean. In the chemical composition, T2 showed higher(p<0.05) moisture, T4 showed higher protein(p<0.05), T3 showed higher fat(p<0.05) and T5 showed higher ash(p<0.05). In the pH value, T5 showed lower(p<0.05) pH value than the other raw meat. In the TBA value, T3 showed lower(p<0.05) value than other products from 0 day to 7 day of storage. In the total microbial count, T3 showed lower(p<0.05) value compared to other raw meat from 0 day. But T1 showed lower(p<0.05) from 7 day. In the VBN value, T5 showed higher(p<0.05) from 0 day. And all raw meats showed higher than standard value from 7 day. As a result, MDCM showed difference the quality and storage characteristics the between companies in Korea. It is impossible to store MDCM more than 7 days when stored at 4℃ refrigerator.

      • 6-Trifluoromethyl-2-thiouracil possesses anti-Toxoplasma gondii effect in vitro and in vivo with low hepatotoxicity

        Since pyrimethamine, the general therapeutic drug for toxoplasmosis, presents several adverse side effects, the need to develop and evaluate new drugs for the condition is critical. In this study, anti-Toxoplasma gondii activities of 3-[{2-((E)-furan-2-ylmethylene)hydrazinyl}methylene]-1,3-dihydroindol-2-one (ATT-5126) and 6-trifluoromethyl-2-thiouracil (KH-0562) were evaluated in vitro using a 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay and in vivo by measuring amount of the tachyzoites in mice ascites. Biochemical parameters such as lipid peroxidation (LPO), glutathione (GSH), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) were also evaluated in livers of mice at 4days post-infection. As a result, the ATT-5126 and KH-0562 showed anti-T. gondii activity in vitro. Treatment of ATT-5126 or KH-0562 decreased the amount of tachyzoites in T. gondii infected ICR mice. The relative weight of liver and spleen increased by T. gondii infection were decreased by treatment of ATT-5126 or KH-0562. The levels of LPO, ALT and AST, which are biochemical parameters involved in liver injury, were also significantly recovered by treatment of ATT-5126 or KH-0562 (p<0.05). In particular, the recovered levels by KH-0562 were similar to those of pyrimethamine-treated group (p<0.05). However, the level of GSH, which is an antioxidant indicator, showed insignificant statistics. The results suggest that KH-0562 show anti-T. gondii activities in vitro and in vivo with low hepatotoxicity. Therefore, KH-0562 may be a useful candidate for treating T. gondii infection.

        Nutritional and Hormonal Induction of Fatty Liver Syndrome and Effects of Dietary Lipotropic Factors in Egg-type Male Chicks

        This experiment was conducted with male chicks to investigate the influence of hormones and nutrients on the development of fatty liver syndrome (FLS) as well as the effects of dietary lipotropic factors on hepatic fat accumulation and lipogenic enzyme gene expression. A total of two-hundred sixteen 4-wk-old Hy-Line male chicks were divided into six groups and fed an experimental diet (T1, low-energy diet with low levels of lipotropic factors; T2, high-energy diet with low levels of lipotropic factors; T3 and T5, low-energy diet with high levels of lipotropic factors; T4 and T6, high-energy diet with high levels of lipotropic factors) for six weeks. The chicks in T5 and T6 groups were treated with intramuscular injections of estradiol benzoate for three days prior to biopsy and clinical analysis of FLS. Chicks treated with estrogen had significantly greater liver weights than untreated chicks. The abdominal fat contents were increased in chicks consuming high-energy diets as compared to those consuming low-energy diets. Treatment with estrogen significantly increased the concentrations of serum cholesterol, triacylglycerol and phospholipid (p<0.05). The hepatic triacylglycerol levels were tenfold higher in the estrogen treated chicks than in the untreated chicks. There were no significant differences in malondialdehyde levels between the treatment groups. Estrogen treatment dramatically increased the levels of fatty acid synthetase, acetyl-CoA carboxylase and ApoB mRNA. The results indicated that treatment with exogenous estrogen in growing male chicks induced hepatic fat accumulation, which might be partially due to increased lipogenic enzyme gene expression.

      • Mesoporous Silica-Coated Hollow Manganese Oxide Nanoparticles as Positive <i>T</i> <sub>1</sub> Contrast Agents for Labeling and MRI Tracking of Adipose-Derived Mesenchymal Stem Cells

        <P/><P>Mesoporous silica-coated hollow manganese oxide (HMnO@mSiO<SUB>2</SUB>) nanoparticles were developed as a novel <I>T</I><SUB>1</SUB> magnetic resonance imaging (MRI) contrast agent. We hypothesized that the mesoporous structure of the nanoparticle shell enables optimal access of water molecules to the magnetic core, and consequently, an effective longitudinal (<I>R</I><SUB>1</SUB>) relaxation enhancement of water protons, which value was measured to be 0.99 (mM<SUP>−1</SUP>s<SUP>−1</SUP>) at 11.7 T. Adipose-derived mesenchymal stem cells (MSCs) were efficiently labeled using electroporation, with much shorter <I>T</I><SUB>1</SUB> values as compared to direct incubation without electroporation, which was also evidenced by signal enhancement on <I>T</I><SUB>1</SUB>-weighted MR images in vitro. Intracranial grafting of HMnO@mSiO<SUB>2</SUB>-labeled MSCs enabled serial MR monitoring of cell transplants over 14 days. These novel nanoparticles may extend the arsenal of currently available nanoparticle MR contrast agents by providing positive contrast on <I>T</I><SUB>1</SUB>-weighted images at high magnetic field strengths.</P>

      • 포도주박에 숙성시킨 돼지 장족의 품질 및 저장특성

        본 연구는 포도주박 숙성 돼지 장족을 가열한 후 품질특성과 냉장 보관 중 저장특성을 분석하였다. 일반성분 분석 결과 지방함량의 변이가 있었 으며, 수분, 단백질, 회분은 유의적인 차이를 보이지 않았다. 포도주박 숙성은 장족의 pH, 명도, 황색도 값을 감소시켰고, 전단력은 처리구간 변이가 크게 나타났다. 또한 저장기간 중 지방산패도와 총미생물수를 감소시키는 결과를 보여주었다. 이상의 결과를 종합해보면 돼지 장족의 포도주박 숙성은 저장특성을 증가시키는 것으로 판단되며 족발제품의 보존성 향상에 활용할 수 있을 것으로 판단된다. This study was conducted to evaluate the quality and storage characteristics of pork hocks by marination of grape pomace during storage periods. Experimental design included 5 treatments : Control, T1: 1% Grape pomace powder + 3 Hours Marination, T2: 1% Grape pomace powder + 6 Hours Marination, T3: 2% Grape pomace powder + 3 Hours Marination, T4: 2% Grape pomace powder + 6 Hours Marination. In the proximate analysis, there were no significant differences except for fat content. In the quality characteristics of pork hocks, the pH, lightness and yellowness were decreased by the marination of grape pomace. The water holding capacity values were 83.61–88.26 and there were large varitions in shear force values. Also, During cold storage, the marination of grape pomace reduced TBA values and total microbial count in pork hocks. As a result, the marination of grape pomace is determined to increase the storage characteristics and could be able to take advantege of the improved storage stability of the pork hocks products.

      • Characteristics of critical current and index n of Bi-2223/Ag tape up to 30 T at 4.2 K

        We fabricated 55-filament Bi-2223/Ag tape and evaluated the dependence of critical current (I<SUB>c</SUB>) and index n in an external magnetic field up to 30 T at 4.2 K, in order to explore the possible application for a magnet in persistent current mode. I<SUB>c</SUB> and index n were characterized as a function of temperature and the magnitude and orientation of the magnetic field by increasing and decreasing field. It was observed that I<SUB>c</SUB> was dependent on the magnitude and orientation of the magnetic field. The I<SUB>c</SUB> decreased exponentially as the field increased; in a parallel and increasing field, I<SUB>c</SUB> was 128 A at 30 T, which is approximately 40% of the I<SUB>c</SUB> in self-field. On the other hand, index n was not significantly dependent on the field up to 30 T, nor did it vary significantly on increasing or decreasing field; the n value in the range of 0.1∼1 μV/cm was 23.0±5.2 and 27.8±8.0 on increasing and decreasing field, respectively, in the range of 0∼30 T. In addition, we observed that local I<SUB>c</SUB> and index n of a long length of tape (4.2 m) were dispersed over a wide range and that the variation of each of these two values with the position of the tape was similar, suggesting that these properties depend on the microstructural uniformity.
