        Ganglioside are ubiquitous membrane component in mammalian cells and suggested to play important roles in various cell functions such as cell-cell recognition, differentiation and transmembrane signaling. These compounds are localized in a glycosphingolipid-enriched microdomain on the cell surface and regulated by the glycosphingolipid composition. However, the role that gangliosides play in the adhesive interaction response by xenotransplantation is not yet clearly understood. In this study, we investigated expression patterns of gangliosides in tight contact between human umbilical vein endothelial cells (HUVECs) and micro-pig aortic endothelial cells (MPAECs). Analysis of ganglioside expression pattern showed that differential expression patterns of ganglioside in tight contact between HUVECs and MPAECs. Furthermore, we confirmed that cell adhesion molecule expression in tight contact between HUVECs and MPAECs. Taken together, these results suggested that differential expression of gangliosides in tight contact between HUVECs and MPAECs may play a role in adhesive interaction response in successful clinical xenotransplantation.

        Ganglioside GT1b Mediates Neuronal Differentiation of Mouse Embryonic Stem Cells

        It has been reported that ganglioside GT1b is expressed during neuronal cell differentiation from undifferentiated mouse embryonic stem cells (mESCs), which suggests that ganglioside GT1b has a direct effect on neuronal cell differentiation. Therefore, this study was conducted to evaluate the effect of exogenous addition of ganglioside GT1b to an in vitro model of neuronal cell differentiation from undifferentiated mESCs. The results revealed that a significant increase in the expression of ganglioside GT1b occurred during neuronal differentiation of undifferentiated mESCs. Next, we evaluated the effect of retinoic acid (RA) on GT1b-treated undifferentiated mESCs, which was found to lead to increased neuronal differentiation. Taken together, the results of this study suggest that ganglioside GT1b plays a crucial role in neuronal differentiation of mESCs.

      • Comparison of gangliosides expression patterns between human adipose and dental pulp-derived mesenchymal stem cells differentiation into osteoblast

        Human adipose-derived stem cells (hADSCs) and dental pulp-derived stem cells (hDPSCs) have been considered alternative sources of adult stem cells because of their potential to trans-differentiate into multiple cell lineages. This study investigated the possible role of gangliosides in the osteoblast differentiation of hADSCs and hDPSCs. First, we investigated characterization of hADSCs and hDPSCs using FACS analysis. Mesenchymal stem cell specific markers, CD44 and CD105, were expressed but not hematopoetic markers, CD45 and CD117 in both of hADSCs and hDPSCs. High-performance thin-layer chromatography analysis showed that increased gangliosides were associated with differentiation of hADSCs and hDPSCs into osteoblasts. RT-PCR analysis confirmed that osteoblast specific genes, ALP, BMP-2, collagen were expressed in differentiated osteoblasts, however, the another osteoblast specific gene, osteocalcin, was not expressed. When hADSCs and hDPSCs were cultured under osteoblast- differentiation conditions, alkaline phosphatase (ALP) activity was increased in comparison to hADSCs and hDPSCs. Furthermore, specifically both ALP activity and ganglioside expression increased more in hDPSCs-derived osteoblasts than hADSCs-derived osteoblasts. These results suggest that gangliosides play a more important role in regulating the osteoblast-differentiation of hDPSCs compared to hADSCs.

      • Expression Pattern of Gangliosides on the Differentiation of Human Mesenchymal Stem Cells into Dopaminergic Neuron-like Cells

        Gangliosides, sialic acid-conjugated glycosphingolipids, are involved in cell proliferation and differentiation. This study investigated the possible role of gangliosides in the dopaminergic neuron-like cells of human mesenchymal stem cells (hMSCs). We have reported the in vitro conditions for inducing hMSCs into dopaminergic neuron-like cells. hMSCs were induced in vitro to become dopaminergic neuron-like cells in 12 days by using a cocktail that includes sonic hedgehog and fibroblast growth factors. First, we investigated the characterization of hMSCs through FACS analysis. The mesenchymal stem cell specific markers, CD44 and CD105, were expressed in hMSCs while the hematopoetic markers, CD45 and CD117 were not. The cells developed a neuronal morphology expressing the neuronal markers Map2 and βⅢtubulin. RT-PCR analysis revealed the expression of dopamine specific genes such as TH, Pitx3, Nurr1 and DA transporter. Immunocytochemistry showed that hMSCs expressed GM3, GM2, GD1a, GD3 and GT1b based on tyrosine hydroxylase (TH) expression. Specifically, a significant decrease in GM3 and GT1b expression was observed during dopaminergic neuron differentiation. These results suggest that ganglioside GT1b may play a role in the dopaminergic neuron differentiation and development process of hMSCs.

However, the role that gangliosides play in immune rejection response by xenotransplantation is not yet clearly understood. In this study, differential expression patterens of gangliosides of Micro-Pig solid organs, which are heart, kidney, liver, pancreas and spleen. We examined expression level of high-performance thin-layer chromatography (HPTLC) showed that all of solid organs contained GM3, GM2, GM1 and GD1a as major gangliosides. Especially, ganglioside GD1b expressed in liver and pancreas, not in heart, kidney and spleen. Also, GT1b expressed in liver, kidney and spleen, not in heart and pancreas. As a results suggest that differential expression patterens of gangliosides of Micro-Pig solid organs are related immune rejection response in xenotransplantation by biomarker.

      • Regulatory elements and transcriptional control of chicken <i>vasa</i> homologue ( <i>CVH</i> ) promoter in chicken primordial germ cells

        <P><B>Background</B></P><P>Primordial germ cells (PGCs), the precursors of functional gametes, have distinct characteristics and exhibit several unique molecular mechanisms to maintain pluripotency and germness in comparison to somatic cells. They express germ cell-specific RNA binding proteins (RBPs) by modulating tissue-specific <I>cis</I>- and <I>trans</I>-regulatory elements. Studies on gene structures of chicken <I>vasa</I> homologue (<I>CVH</I>), a chicken RNA binding protein, involved in temporal and spatial regulation are thus important not only for understanding the molecular mechanisms that regulate germ cell fate, but also for practical applications of primordial germ cells. However, very limited studies are available on regulatory elements that control germ cell-specific expression in chicken. Therefore, we investigated the intricate regulatory mechanism(s) that governs transcriptional control of <I>CVH</I>.</P><P><B>Results</B></P><P>We constructed green fluorescence protein (GFP) or luciferase reporter vectors containing the various 5′ flanking regions of <I>CVH</I> gene. From the 5′ deletion and fragmented assays in chicken PGCs, we have identified a <I>CVH</I> promoter that locates at −316 to +275 base pair fragment with the highest luciferase activity. Additionally, we confirmed for the first time that the 5′ untranslated region (UTR) containing intron 1 is required for promoter activity of the <I>CVH</I> gene in chicken PGCs. Furthermore, using a transcription factor binding prediction, transcriptome analysis and siRNA-mediated knockdown, we have identified that a set of transcription factors play a role in the PGC-specific <I>CVH</I> gene expression.</P><P><B>Conclusions</B></P><P>These results demonstrate that <I>cis</I>-elements and transcription factors localizing in the 5′ flanking region including the 5′ UTR and an intron are important for transcriptional regulation of the <I>CVH</I> gene in chicken PGCs. Finally, this information will contribute to research studies in areas of reproductive biology, constructing of germ cell-specific synthetic promoter for tracing primordial germ cells as well as understanding the transcriptional regulation for maintaining germness in PGCs.</P>

        유치원 교사의 일과 삶의 균형 분석

        본 연구는 유치원 교사의 일과 삶의 균형에 대한 전반적인 경향 및 변인에 따른 차이를 알아보는데 목적이 있었다. 연구대상은 유치원에 근무하는 교사 222명이었으며, 연구도구로는 김정운, 박정열(2008)이 개발한 직장인 대상의 ‘일과 삶의 균형(Work-Life Balance) 척도’를 유치원 교사에게 적합하게 수정․보완하여 사용하였다. 연구결과, 첫째, 유치원 교사의 일과 삶의 균형은 보통보다 약간 낮은 정도의 수준으로 나타났다. 하위요인에서는 일-가정 균형, 일-여가 균형, 일-성장 균형은 유사한 반면, 전반적 평가는 상대적으로 낮은 경향을 보였다. 둘째, 변인에 따른 차이에서는 교사가 근무하는 기관유형, 근무시간에 따라 일과 삶의 균형 전체와 모든 하위요인에 유의한 차이가 있었고, 경력과 교육정도에 따라 일과 삶의 균형 전체 혹은 일부 하위요인에 유의한 차이가 있었으나, 결혼여부, 월급여에 따라서는 유의한 차이가 나타나지 않았다. 즉, 근무기관이 공립단설유치원인 경우에 일과 삶의 균형이 낮았으며, 근무시간이 길수록 일과 삶의 균형이 낮았고, 경력이 적거나 학력이 높을수록 일과 삶의 균형이 높아지는 경향이 있었다. 이러한 결과는 유치원 교사의 일과 삶의 균형에 지속적인 관심을 가지고, 일과 삶의 균형을 높이기 위한 실제적 지원 방안을 마련할 필요가 있음을 시사한다. The purpose of this study was to examine overall aspects of kindergarten teachers" work-life balance and investigate differences of the teachers" work-life balance depending on their personal and school-related variables. The subjects of this study were 222 kindergarten teachers, and the research tool was a modified version of the Work-Life Balance Scale developed by Kim and Park (2008). The results of this study are as follows. First, the mean score of kindergarten teachers" work-life balance was below average. In terms of the subordinate factors, the scores of ‘work-life harmony’, ‘work-leisure harmony’, and ‘work-growth harmony’ were similar, whereas the score of ‘overall work-life rating’ was relatively low. Second, the total score of work-life balance and the scores of all subordinate factors were significantly different depending on types of kindergartens and hours of duty. In addition, the total score of work-life balance and/or the scores of some subordinate factors were significantly different depending on their teaching careers and education levels. On the other hand, there was no significant difference on their work-life balance depending on their marital statuses and salaries. In other words, teachers working at an independent public kindergarten showed significantly lower work-life balance than teachers working at other types of kindergartens; teachers who had longer working hours showed lower work-life balance; teachers who had short teaching career or high education level showed a tendency of higher work-life balance than others. These results imply that continuous attention to kindergarten teachers’ work-life balance is essential, and practical support to increase their work-life balance should be planned and implemented.

        탄소-탄소 복합재료의 하프늄 탄화물 코팅재의 열적/기계적 특성

        본 논문에서는 C/C-SiC 복합재료의 하프늄 탄화물 코팅재에 대한 열적, 기계적 특성을 평가하였으며 특히 코팅에 의한 내산화성과 내마모성의 향상여부를 평가하였다. 하프늄 탄화물(HfC)을 용사시켜 코팅한 샘플들을 가공한 후, 공기 중에서 열적 특성평가 및 마모, 압입시험 평가에 대한 연구를 수행하였다. 공기 중에서 1200oC의 온도까지 승온시킨 후 1시간 유지하는 싸이클을 10싸이클 진행하여 각 싸이클마다의 무게변화를 통해 탄소의 산화저항성을 평가하였고, 초경 구(tungsten carbide)를 사용하여 마모시험과 압입시험을 수행하여 그 결과를 비교하였다. 열피로 시험 수행 결과 하프늄 탄화물 코팅재가 상대적으로 무게감소가 적어 상대적으로 내산화성이 높은 것으로 평가되었다. 코팅된 하프늄 탄화물에 의해 탄성계수가 상대적으로 증가하였으며, 또한 C/C-SiC 복합재료는 하프늄 탄화물의 코팅에 의하여 내마모성이 향상되어 동일조건에서 마모량이 상대적으로 적었고 낮고 안정된 마찰계수가 유지되었다. This study investigates thermal and mechanical characterization of Hafnium carbide coating on the Cf-C composites. The hafnium carbide coatings by vacuum plasma spray on the C/C-SiC composites are prepared to evaluate oxidation and wear resistance. We perform the thermal durability tests by thermal cycling at 1200℃ for 10cycles in air and investigates the weight change of each cycle. We also evaluate the wear and indentation behavior using tungsten carbide ball indenter as a mechanical evaluation. As a result, the HfC coating is beneficial to reduce of weight loss during thermal cycling test and improve the elastic property of C/C-SiC composite. Especially, the HfC coating improves the wear resistance of C/C-SiC composite.
