Down syndrome with asymptomatic neuroglial cyst : A case report and review of the literature

Down syndrome (DS, trisomy 21) is associated with neuroanatomical abnormalities, including choroid plexus cysts and various types of brain tumors. Trisomy 21 is associated with oncogenic factor, especially in brain tumor. The brain of DS patients had a smaller volume of gray and white matter and an unbalanced cerebellum volume, indicating a smaller volume overall than normal. We report a case of a DS male patient who had an incidentally discovered neuroglial cyst in left cerebellar vermis. He visited our hospital with gait disturbance and fatigue. But, the neurologic exam was normal. To the best of our knowledge, this is the first reported case of a neuroglial cyst in a trisomy 21 patient. As the developmental mechanisms of a cyst and the choroid plexus are related, more research is needed.

Brain ischemia brings about hypoxic insults. Hypoxia is one of the major pathological factors inducing neuronal injury and central nervous system infection. We studied the involvement of mitogen-activated protein (MAP) kinase in hypoxia-induced apoptosis using cobalt chloride in C6 glioma cells. In vitro cytotoxicity of cobalt chloride was tested by MTT assay. Its IC_(50), value was 400μM. The DNA fragment became evident after incubation of the cells with 300μM cobalt chloride for 24h. We also evidenced nuclear cleavage with morphological changes of the cells undergoing apoptosis with electron microscopy. Next, we examined the signal pathway of cobalt chloride-induced apoptosis in C6 cells. The activation of extracellular signal-regulated protein kinase 1/2 (ERK 1/2) started to increase at 1 h and was activated further at 6h after treatment of 400M cobalt chloride. In addition, pretreatment of PD98059 inhibited cobalt chloride-induced apoptotic cell morphology in Electron Microscopy. These results suggest that cobalt chloride is able to induce the apoptotic activity in C6 glioma cells, and its apoptotic mechanism may be associated with signal transduction via MAP kinase (ERK 112).
Inhibitory effects of KHG26377 on glutamate dehydrogenase activity in cultured islets

GDH has been known to be related with hyperinsulinism-hyperammonemia syndrome. We have screened new drugs with a view to developing effective drugs modulating GDH activity. In the present work, we investigated the effects of a new drug, KHG26377 on glutamate formation and GDH activity in cultured rat islets. When KHG26377 was added to the culture medium for 24 h prior to kinetic analysis, the Vmax of GDH was decreased by 59% whereas Km is not significantly changed. The concentration of glutamate decreased by 50% and perfusion of islets with KHG26377 reduced insulin release by up to 55%. Our results show that KHG26377 regulates insulin release by inhibiting GDH activity in primary cultured islets and support the previous studies for the connection between GDH activity and insulin release. Further studies are required to determine in vivo effects and pharmacokinetics of the drug. [BMB reports 2010; 43(4): 245-249]

Parkinson's disease (PD) is a common chronic neurodegenerative disease mainly caused by the death of dopaminergic neurons. However, no complete pharmacotherapeutic approaches are currently available for PD therapies. 1-methyl-4-phenylpyridinium (MPP<sup>+</sup>)-induced SH-SY5Y neurotoxicity has been broadly utilized to create cellular models and study the mechanisms and critical aspects of PD. In the present study, we examined the role of a novel azetidine derivative, 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride (KHG26792), against MPP<sup>+</sup>-induced neurotoxicity in SH-SY5Y cells. Treatment of KHG26792 significantly attenuated MPP+-induced changes in the protein levels of Bcl-2 and Bax together with efficient suppression of MPP+-induced activation of caspase-3 activity. KHG26792 also attenuated mitochondrial potential and levels of ROS, Ca<sup>2+</sup>, and ATP in MPP<sup>+</sup>-treated SH-SY5Y cells. Additionally, KHG26792 inhibited the induced production of nitric oxide and malondialdehyde. Moreover, the protective effect of KHG26792 is mediated through regulation of glutathione peroxidase and GDNF levels. Our results suggest a possibility that KHG26792 treatment significantly protects against MPP<sup>+</sup>-induced neurotoxicity in SH-SY5Y cells and KHG26792 may be a valuable therapeutic agent for the treatment of PD induced by an environmental toxin. <P>A regioselective, reagent-based method for the cyclization reaction of 2-amino-1,3,4-oxadiazole and 2-amino-1,3,4-thiadiazole core skeletons is described. The thiosemicarbazide intermediate <B>3</B> was reacted with EDC·HCl in DMSO or <I>p</I>-TsCl, triethylamine in <I>N</I>-methyl-2-pyrrolidone to give the corresponding 2-amino-1,3,4-oxadiazoles <B>4</B> and 2-amino-1,3,4-thiadiazoles <B>5</B> through regioselcective cyclization processes. The regioselectivity was affected by both R<SUP>1</SUP> and R<SUP>2</SUP> in <I>p</I>-TsCl mediated cyclization. It is shown in select sets of thiosemicarbazide <B>3</B> with R<SUP>1</SUP>(benzyl) and R<SUP>2</SUP>(phenyl). 2-Amino-1,3,4-oxadiazole <B>4</B> was also shown in the reaction of <I>p</I>-TsCl mediated cyclization. Dynamic self-assembly of droplets, regular structure formation of moving deformable objects in a confinement environment is a challenging problem in nonlinear dynamics and engineering patterned structure. In the current work, we investigated how the local kinematic history affects the dynamic self-assembly of picoliter-sized droplets near the expansion regions in microfluidic devices. The local kinematic history was controlled by the shape of the expansion region and characterized using computational fluid dynamics. Sizecontrolled aqueous droplets in light mineral oil were continuously generated at T-junction microchannel and transported toward the expansion region. The fast dynamics of the droplets was tracked using high-speed video microscopy. We found three types of dynamic droplet arrays: 1D, 2D zigzag, and irregular. The orderdisorder transition was associated not only with the droplet size, but also with the controlled local kinematic history, which results in the transient deformation of droplet and droplet-droplet interactions. The present results provide us with insight into the dynamic self-assembly of droplets and could be a useful guide for practical applications of droplet-based microfluidics.

A series of compounds containing both 1,3,4-oxadiazole and thiohydantoin were synthesized as a promising scaffold for application in medicinal chemistry. The key step of the synthesis is a microwave-assisted cyclization of semicarbazides possessing a thiohydantoin moiety at one of the acyl termini using $POCl_3$ as a dehydrating reagent. A wide range of semicarbazides were prepared through the substitution of hydrazides with an N-acylated thiohydantoin derived from the cyclization of the corresponding isothiocyanate with an amino acid and subsequent N-acylation of the resultant thiohydrantion. Consequently, the 58 number of 1,3,4-oxadiazole derivatives having a thiohydantoin substituent were prepared in good overall yields.
Single-File Diffusion of Protein Drugs through Cylindrical Nanochannels

<P>A new drug delivery device using cylindrical block copolymer nanochannels was successfully developed for controlled protein drug delivery applications. Depending on the hydrodynamic diameter of the protein drugs, the pore size in cylindrical nanochannels could be controlled precisely down to 6 nm by Au deposition. Zero-order release of bovine serum albumin (BSA) and human growth hormone (hGH) by single-file diffusion, which has been observed for gas diffusion through zeolite pores, was realized up to 2 months without protein denaturation.
