<P><B>Abstract</B></P> <P>Leptin, a hormone primarily derived from adipose tissue, is known to induce tumor growth, but its underlying mechanisms of action are not clearly understood. Inflammasomes, acting as signaling platforms for controlling inflammatory responses, modulate tumor growth in a complicated manner. In this study, we investigated the role of inflammasomes in leptin-induced growth of breast cancer cells. Herein, we showed that leptin activated NLRP3 inflammasomes in MCF-7 breast cancer cells, as determined by activation of caspase-1, maturation of interleukin-1β, and increased expression of the inflammasome components, including NLRP3 and ASC. Interestingly, inhibition of the inflammasome by treatment with a pharmacological inhibitor of caspase-1 or gene silencing of NLRP3 prevented leptin-induced increase in cell viability. Moreover, suppression of apoptosis and cell cycle promotion by leptin were also significantly abolished by gene silencing of NLRP3, clearly indicating a crucial role of NLRP3 inflammasomes in leptin-induced breast cancer growth. In addition, inhibition of estrogen receptor signaling or ROS production markedly blocked leptin-induced activation of NLRP3 inflammasomes, suggesting that estrogen receptor signaling and ROS production mediate inflammasomes activation by leptin. The stimulatory effect of leptin on inflammasomes activation was also observed in MCF-7 tumor xenograft model. Furthermore, the critical roles of inflammasomes activation in leptin-induced tumor growth, suppression of apoptotic gene expression, and induction of the genes stimulating cell cycle were confirmed in a tumor xenograft model. Taken together, these results demonstrate that inflammasomes activation plays a pivotal role in leptin-induced growth of breast cancer cells via modulation of both apoptosis and cell cycle.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Nano scale networks are futuristic networks deemed as enablers for the Internet of Nano Things, Body area nano networks, target tracking, anomaly/ abnormality detection at molecular level and neuronal therapy / drug delivery applications. Molecular communication is considered the most compatible communication technology for nano devices. However, connectivity in such networks is very low due to inter-symbol interference (ISI). Few research papers have addressed the issue of ISI mitigation in molecular communication. However, many of these methods are not adaptive to dynamic environmental conditions. This paper presents an enhancement over original Memory-1 ISI cancellation scheme using maximum likelihood estimation of a channel parameter (λ) to make it adaptable to variable channel conditions. Results of the Monte Carlo simulation show that, the connectivity (Pconn) improves by 28% for given simulation parameters and environmental conditions by using enhanced Memory-1 cancellation method. Moreover, this ISI mitigation method allows reduction in symbol time (Ts) up to 50 seconds i.e. an improvement of 75% is achieved.
Case Studies on Sexual Reproductive Health and& Women with Disability

Nepal Census report 2011 states that 2 percent of total population has some kind of disability in Nepal with women consisting 1.71 prevalence rate. While men with disability can still be eligible to get married, get a job, settle with family, women with disability are judged, are considered to be ineligible to get married and have children and even to get a job. Women with disability are deprived of their basic rights, the issue of Sexual and Reproductive Health is excluded. Women with disability face stigma associated with marriage, sexual behavior and use of contraceptives. Due to the lack of proper education and lack of open nature of society, family and even the service providers are not being able to provide sufficient information and services to women with disability. The following case studies in the paper tells us the scenario of women living with disability in the society where there is a huge gap between the social status of men and women. These case studies talk about the experience of women living with disabilities and their challenges facing social stigma, barriers to access sexual reproductive health information and services and struggle to get the equal rights as women and as women with disability.
Pattern of Liver Enzymes in Alcohol Dependence Syndrome Patients

Aims: Alcohol dependence syndrome (ADS) has become a global public health challenge because of its high prevalence and the concomitant increase in risk of liver disease, cardiovascular disease and premature death. Influence of alcohol use on liver metabolism is well recognized. This study was aimed at examining the association of liver enzymes like γ-glutamyltransferase (GGT) and aminotransferase, with alcohol dependence syndrome patients. Methods: This cross-sectional study was conducted in Tribhuvan University Teaching Hospital. ADS patients were screened by the consultant psychiatrist using the Alcohol Use Disorder Identification Test (AUDIT) questionnaire. A total of 89 patients scored positive on the AUDIT as having alcohol-related problems and were included in the study. Blood Pressure and other anthropometric parameters were measured while blood samples were analyzed for liver enzymes and serum protein. Results: Mean age of cases and controls was 35.42 ± 5.6 & 34.53 ± 3.5 years respectively. The mean values of Gamma GT, SGOT and SGPT were largely elevated in cases as compared to the controls with a statically significance (P<0.001). Among the ADS cases serum GGT level was elevated in 97% patients. The SGOT/SGPT ratio was also significantly higher in cases (2.02 ± 0.39) and control (1.45±0.62). It was found that 15.1 % cases had low serum protein level and 32.9% cases were low serum albumin level. Albumin to globulin ratio was also significantly decreased in cases (1.16 ±0.29). Conclusions: These findings support the hypothesis that, alcohol may affect the pattern of liver enzymes and also damage the liver cells. Decrease in serum albumin and elevation of SGOT to SGPT ratio more than two is suggestive of development of liver cirrhosis in alcohol dependence patients.
