        Determination of the Amino Acid Requirements and Optimum Dietary Amino Acid Pattern for Growing Chinese Taihe Silky Fowls in Early Stage

        A mathematical model has been constructed to estimate the amino acid requirements for growing Taihe silky fowls in early stage. A requirement was taken as the sum of the needs for maintenance, for gain in carcass weight without feathers, and for the feathers. The maintenance requirement was considered to be the sum of the needs for replacing skin and intestinal losses and for the obligatory creatinine excretion in the urine. A comparative slaughter trial and nitrogen balance trials with growing and adult Taihe silky fowls, respectively, were conducted to estimate the parameters in the model. The amino acid requirements were then calculated with the constructed models. The results showed as following: the replacement needs for skin nitrogen loss was determined at 213.41 mg/d for adult male fowls (body weight 1.60 kg); creatinine excretion in these birds was 4.04 mg/d. when fed an nitrogen-free diets, the adult male fowls with body weight 1.60 kg excreted a total of 246.10 mg/d endogenous nitrogen. The net protein requirement for maintenance was estimated at $11.24mg/w_g{^{0.75}}/d$Per gram of body weight gain contained 27.18 mg carcass nitrogen for growing birds in early stage, but feathers nitrogen in per gram of body weight gain increased with age. The amino acid requirements for growing Taihe silky fowls were slightly higher than for starting and growing pullets, but lower than that of broiler chicks. The amino acid requirements patterns changed with weeks of age.

        Traditional Chinese Medicine Prescriptions Enhance Growth Performance of Heat Stressed Beef Cattle by Relieving Heat Stress Responses and Increasing Apparent Nutrient Digestibility

        The present aim was to investigate the effects of traditional Chinese medicine prescriptions (TCM) on body temperature, blood physiological parameters, nutrient apparent digestibility and growth performance of beef cattle under heat stress conditions. Twenty-seven beef cattle were randomly divided into three groups as following; i) high temperature control (HTC), ii) traditional Chinese medicine prescriptions I+high temperature (TCM I) and iii) traditional Chinese medicine prescriptions II+high temperature (TCM II) (n = 9 per group). The results showed that the mean body temperature declined in TCM II treatment (p<0.05). Serum $T_3$ and $T_4$ levels with TCM I and TCM II treatments elevated (p<0.05), and serum cortisol levels of TCM I treatments decreased (p<0.05), compared with the HTC group. Total protein, albumin, globulin in TCM II treatments elevated and blood urea nitrogen levels of both TCM treatments increased, but glucose levels of both TCM treatments decreased, compared with the HTC group (p<0.05). The apparent digestibility of organic matter and crude protein with TCM I treatment increased, and the apparent digestibility of acid detergent fiber elevated in both TCM treatments (p<0.05). Average daily feed intake was not different among three groups, however average daily gain increased and the feed:gain ratio decreased with both TCM treatments, compared with the HTC group (p<0.05). The present results suggest that dietary supplementation with TCM I or TCM II improves growth performance of heat stressed beef cattle by relieving heat stress responses and increasing nutrient apparent digestibility.

        Characterization of a GH8 β-1,4-Glucanase from Bacillus subtilis B111 and Its Saccharification Potential for Agricultural Straws

        Herein, we cloned and expressed an endo-β-1,4-glucanase gene (celA1805) from Bacillus subtilis B111 in Escherichia coli. The recombinant celA1805 contains a glycosyl hydrolase (GH) family 8 domain and shared 76.8% identity with endo-1,4-β-glucanase from Bacillus sp. KSM-330. Results showed that the optimal pH and temperature of celA1805 were 6.0 and 50°C, respectively, and it was stable at pH 3-9 and temperature ≤50°C. Metal ions slightly affected enzyme activity, but chemical agents generally inhibited enzyme activity. Moreover, celA1805 showed a wide substrate specificity to CMC, barley β-glucan, lichenin, chitosan, PASC and avicel. The K<sub>m</sub> and V<sub>max</sub> values of celA1805 were 1.78 mg/ml and 50.09 μmol/min/mg. When incubated with cellooligosaccharides ranging from cellotriose to cellopentose, celA1805 mainly hydrolyzed cellotetrose (G4) and cellopentose (G5) to cellose (G2) and cellotriose (G3), but hardly hydrolyzed cellotriose. The concentrations of reducing sugars saccharified by celA1805 from wheat straw, rape straw, rice straw, peanut straw, and corn straw were increased by 0.21, 0.51, 0.26, 0.36, and 0.66 mg/ml, respectively. The results obtained in this study suggest potential applications of celA1805 in biomass saccharification.

        Characterization of a Thermostable Lichenase from Bacillus subtilis B110 and Its Effects on β-Glucan Hydrolysis

        Lichenase is an enzyme mainly implicated in the degradation of polysaccharides in the cell walls of grains. Emerging evidence shows that a highly efficient expression of a thermostable recombinant lichenase holds considerable promise for application in the beer-brewing and animal feed industries. Herein, we cloned a lichenase gene (CelA203) from Bacillus subtilis B110 and expressed it in E. coli. This gene contains an ORF of 729 bp, encoding a protein with 242 amino acids and a calculated molecular mass of 27.3 kDa. According to the zymogram results, purified CelA203 existed in two forms, a monomer, and a tetramer, but only the tetramer had potent enzymatic activity. CelA203 remained stable over a broad pH and temperature range and retained 40% activity at 70℃ for 1 h. The K<sub>m</sub> and V<sub>max</sub> of CelA203 towards barley β-glucan and lichenan were 3.98 mg/ml, 1017.17 U/mg, and 2.78 mg/ml, 198.24 U/mg, respectively. Furthermore, trisaccharide and tetrasaccharide were the main products obtained from CelA203-mediated hydrolysis of deactivated oat bran. These findings demonstrate a promising role for CelA203 in the production of oligosaccharides in animal feed and brewing industries.

        Nicotinic acid changes rumen fermentation and apparent nutrient digestibility by regulating rumen microbiota in Xiangzhong black cattle

        Objective: The aim of this study was to investigate the impact of dietary nicotinic acid (NA) on apparent nutrient digestibility, rumen fermentation, and rumen microbiota in uncastrated Xiangzhong black cattle. Methods: Twenty-one uncastrated Xiangzhong black cattle (385.08±15.20 kg) aged 1.5 years were randomly assigned to the control group (CL, 0 mg/kg NA in concentrate diet), NA1 group (800 mg/kg NA in concentrate diet) and NA2 group (1,200 mg/kg NA in concentrate diet). All animals were fed a 60% concentrate diet and 40% dried rice straw for a 120-day feeding experiment. Results: Supplemental NA not only enhanced the apparent nutrient digestibility of acid detergent fiber (p<0.01), but also elevated the rumen acetate and total volatile fatty acid concentrations (p<0.05). 16S rRNA gene sequencing analysis of rumen microbiota revealed that dietary NA changed the diversity of rumen microbiota (p<0.05) and the abundance of bacterial taxa in the rumen. The relative abundances of eight Erysipelotrichales taxa, five Ruminococcaceae taxa, and five Sphaerochaetales taxa were decreased by dietary NA (p< 0.05). However, the relative abundances of two taxa belonging to Roseburia faecis were increased by supplemental 800 mg/kg NA, and the abundances of seven Prevotella taxa, three Paraprevotellaceae taxa, three Bifidobacteriaceae taxa, and two operational taxonomic units annotated to Fibrobacter succinogenes were increased by 1,200 mg/kg NA in diets. Furthermore, the correlation analysis found significant correlations between the concentrations of volatile fatty acids in the rumen and the abundances of bacterial taxa, especially Prevotella. Conclusion: The results from this study suggest that dietary NA plays an important role in regulating apparent digestibility of acid detergent fiber, acetate, total volatile fatty acid concentrations, and the composition of rumen microbiota.
