        Caspase Recruitment Domain Containing Protein 9 Suppresses Non-Small Cell Lung Cancer Proliferation and Invasion via Inhibiting MAPK/p38 Pathway

        Purpose Caspase recruitment domain containing protein 9 (CARD9) has been demonstrated to be a pro-tumor factor in various cancers. However, our previous study found a significant decrease of CARD9 in malignant pleural effusion compared with benign pleural effusion. So we investigated the role of CARD9 in non-small cell lung cancer (NSCLC) and its working mechanism. Materials and Methods Immunohistochemistry, western blot, and quantitative real-time polymerase chain reaction were used to detect the expression of CARD9 in specimens of NSCLC patients. The Cancer Genome Atlas (TCGA) database was also used to analyze the expression of CARD9 in NSCLC and its predicting value for prognosis. Immunofluorescence was used for CARD9 cellular location. Cell growth assay, clonal formation assay, wound healing assay, matrigel invasion assay, and flow cytometry were used to test cell proliferation, migration, invasion, apoptosis, and cycle progression of NSCLC cells with CARD9 knockdown or CARD9 overexpression. Co-immunoprecipitation was used to identify the interaction between CARD9 and B-cell lymphoma 10 (BCL10). SB203580 was used to inhibit p38 activation. Results CARD9 was decreased in NSCLC tissues compared with normal tissues; low CARD9 expression was associated with poor survival. CARD9 was expressed both in tumor cells and macrophages. Downregulation of CARD9 in NSCLC cells enhanced the abilities of proliferation, invasion, and migration via activated mitogen-activated protein kinases (MAPK)/p38 signaling, while overexpression of CARD9 presented antitumor effects. BCL10 was identified to interact with CARD9. Conclusion We demonstrate that CARD9 is an independent prognostic factor in NSCLC patients and inhibits proliferation, migration, and invasion by suppressing MAPK/p38 pathway in NSCLC cells.

        <P>We present multi-band results for GRB071010B based on Swift, Suzaku, and ground-based optical observations. This burst is an ideal target to evaluate the robustness of the E-peak(src) - E-iso and E-peak(src) - E-gamma relations, whose studies have been in stagnation due to the lack of the combined estimation of E-peak(src) and long-term optical monitoring. The joint prompt spectral fitting using Swift/Burst Alert Telescope and Suzaku/Wide-band All-sky Monitor data yielded the spectral peak energy as E-peak(src) of 86.5(-6.3)(+6.4) keV and E-iso of 2.25(-0.16)(+0.19) x 10(52) erg with z = 0.947. The optical afterglow light curve is well fitted by a simple power law with temporal index alpha = -0.60 +/- 0.02. The lower limit of temporal break in the optical light curve is 9.8 days. Our multi-wavelength analysis reveals that GRB071010B follows E-peak(src) - E-iso but violates the E-peak(src) - E-gamma and E-iso - E-peak(src) - t(jet)(src) at more than the 3 sigma level.</P>

        Clustering and Recommendation for Semantic Web Service in Time Series

        Promoted by cloud technology and new websites, plenty and variety of Web services are emerging in the Internet. Meanwhile some Web services become outdated even obsolete due to new versions, and a normal phenomenon is that some services work well only with other services of older versions. These laggard or improper services are lowering the performance of the composite service they involved in. In addition, using current technology to identify proper semantic services for a composite service is time-consuming and inaccurate. Thus, we proposed a clustering method and a recommendation method to deal with these problems. Clustering technology is used to classify semantic services according to their topics, functionality and other aspects from plenty of services. Recommendation technology is used to predict the possible preference of a composite service, and recommend possible component services to the composite service according to the history information of invocations and similar composite services. The experiments show that our clustering method with the help of Ontology and TF/IDF technology is more accurate than others, and our recommendation method has less average error than others in the series of missing rate.
